1. Engineering sustainable and bioactive 3D-printed cellulose nanofiber scaffolds from kraft pulps with varying lignin levels.
2026-10-02Growing interest in cellulose nanofibers (CNFs) for wound-care applications has increased the need for sustainable production routes to deliver high-performance biomaterials. Here, CNFs were produced from bleached and unbleached kraft pulps with different lignin contents and formulated as inks for extrusion-based 3D printing. The influence of pulp composition on TEMPO-mediated oxidation, nanofibrillation, rheological behavior, scaffold architecture, Zn functionalization, and biological performance was systematically evaluated. Residual lignin reduced carboxylation and fibrillation efficiency, resulting in larger fibrillar structures and lower rheological recovery. Nevertheless, both CNF suspensions enabled the fabrication of highly porous (>90%) scaffolds with tunable architectures and comparable mechanical properties. Fibroblast studies confirmed cytocompatibility and revealed sustained cell proliferation, while macrophage cultures showed an early pro-inflammatory response characterized by TNF-α and IL-6 secretion. Zn functionalization generated a burst-release profile during the first hour without affecting scaffold mechanics. Although Zn incorporation markedly reduced fibroblast adhesion, cell viability remained above the cytocompatibility threshold. In addition, Zn-loaded scaffolds produced a moderate reduction in bacterial viability and biofilm formation by Staphylococcus aureus and Pseudomonas aeruginosa. Despite their different lignin contents, both pulps yielded scaffolds with comparable Zn loading, release behavior, and overall biological performance. These results show that less-refined lignin-containing pulps can be used to produce printable CNFs and functional wound-dressing scaffolds, supporting the use of more sustainable cellulose sources without compromising scaffold performance.

2. Can tissue engineering applications replace cardiovascular medical devices? Searching for the key to clinical translation.
PMID:日期:2026-10-01Cardiovascular diseases are the number one killer worldwide. Malfunction of the conduction system, heart valves, the myocardial muscle or the vessels irrigating the tissue have been traditionally addressed with medical devices such as pacemakers, left ventricular assist devices, heart valves and vascular grafts. These positively impact critical outcomes such as functionality and mortality. However, by their non-biological nature, they cannot operate with the same efficiency as their original natural counterpart. Conversely, tissue engineering is an interdisciplinary field aimed at fabricating human tissues and organs in the lab. The same tissues that are replaced with device-based solutions are currently the focus of intense research in this area. In the present review, we provide a comparative overview of current medical device-based solutions and their tissue engineered counterparts in four highly impacting cardiovascular areas involving the cardiac conduction system, heart valves, myocardium (muscle) and the vessels. For each, we describe the characteristics of the diseases and their impact on cardiovascular homeostasis, as well as the existing device-based clinical solutions, before summarizing the main work in the tissue engineering field. Finally, we reflect on strategic initiatives that are required to advance experimental bioengineered systems to become a translational reality, analogous to the developmental path of implantable medical devices.

4. An injectable ROS-responsive PRP-loaded hydrogel attenuates osteoarthritis by reducing oxidative stress, inflammation and chondrocyte apoptosis.
2026-09-26Osteoarthritis (OA) is a degenerative joint disease mainly characterized by oxidative stress, inflammatory responses, and loss of cartilage matrix. Although platelet-rich plasma (PRP) has been applied in OA treatment, its intra-articular use is still limited by insufficient local retention and short duration of action. To address this issue, we constructed a reactive oxygen species (ROS)-responsive injectable hydrogel based on quaternized chitosan and tannic acid and loaded it with PRP (PRP@QCTH) as a local delivery strategy for OA treatment. The hydrogel showed favorable injectability, gel-forming ability, and ROS-responsive release behavior. In vitro experiments demonstrated that, compared with the model group, PRP@QCTH improved chondrocyte viability, reduced apoptosis and intracellular ROS accumulation, decreased the levels of IL-1β, TNF-α, and IL-6, and partially restored the expression of Aggrecan and COL2A1. In vivo evaluation further demonstrated that PRP@QCTH alleviated cartilage degeneration, preserved cartilage matrix integrity and improved subchondral bone-related parameters in OA joints, as confirmed by histological and cartilage-specific marker analyses. Transcriptomic analysis suggested that these protective effects might be associated with pathways involved in inflammatory regulation, extracellular matrix remodeling and tissue repair. In conclusion, PRP@QCTH showed protective effects in experimental OA and may serve as a potential local intervention strategy.

5. Fermentation-integrated In Situ functionalization of bacterial cellulose with Coptis chinensis and pomegranate peel extracts for antibacterial wound healing.
2026-09-25Infected wound treatment requires multifunctional biomaterials to combat infection, oxidative stress, and impaired tissue repair. Bacterial cellulose (BC) provides a hydrated nanofibrous scaffold for wound dressings; however, its functionalization often suffers from inefficient incorporation of poorly water-dispersible bioactives and harsh purification procedures. Here, we developed a fermentation-integrated strategy that combines the submerged BC-forming capability of Kosakonia oryzendophytica FY-07 with xanthan-gum-assisted dispersion of plant-derived bioactives and temperature-triggered bacterial autolysis for in situ fabrication of functional BC composites. Alkaloid-rich Coptis chinensis extract and polyphenol-rich pomegranate peel extract were incorporated during BC biosynthesis, followed by mild enzymatic purification. The resulting membranes achieved high bioactive incorporation efficiencies of 89.73% and 94.61%, respectively, and exhibited sustained release behavior. The incorporated bioactives endowed BC membranes with enhanced antioxidant and antibacterial activities, while maintaining favorable cytocompatibility and hemocompatibility. In particular, the pomegranate peel extract-incorporated membrane showed strong radical-scavenging capacity, achieving DPPH• and ABTS• scavenging rates of 97.64% and 95.22%, respectively. Moreover, the engineered FY-07 strain enabled temperature-triggered autolysis, resulting in an approximately 5.1-log reduction in viable cells and facilitating mild downstream purification. In a Staphylococcus aureus-infected full-thickness wound model, the composite membranes effectively promoted wound healing. This work establishes a fermentation-integrated manufacturing platform that couples microbial BC production with in situ incorporation and controlled delivery of natural bioactives, providing a sustainable approach for the fabrication of multifunctional BC-based wound biomaterials.

6. The application of engineered exosome-mimetic lipid nanoparticles in the hepatic rejuvenation of aged donor livers.
2026-09-24The severe global shortage of donor organs necessitates the utilization of aged extended-criteria donor (ECD) livers, which are susceptible to ischemic injury and exhibit a sluggish regeneration due to hepatic senescence. While mesenchymal stem cell-derived exosomes show regenerative promise, clinical translation is hindered by manufacturing bottlenecks and batch heterogeneity. In this study, we have developed a highly controllable, microfluidic-engineered exosome-mimetic lipid nanotherapeutic (LMP) to rejuvenate aged livers. The proposed procedure employs a "decoding and synthetic reconstruction" paradigm, where the regenerative cargo of natural exosomes is converted to a standardized 10-component core, comprising three master-regulatory miRNAs and seven functional proteins. In vitro and in vivo evaluations have demonstrated that the LMP exhibits the requisite biosafety, rapid hepatic internalization, and immunological inertness. A systemic administration of LMP effectively revitalizes baseline liver function in aged mice. In terms of mechanism, the multidimensional rejuvenation is driven by a modulating of the collapsed mitochondrial energy networks, attenuating the senescence-associated secretory phenotype (SASP), and reducing age-related fibrotic architectures. The LMP actively remodels the hepatic immune microenvironment by promoting a shift in the relative distribution of macrophage populations toward a pro-resolving M2-like phenotype, significantly attenuating chronic inflammaging. This study presents a highly scalable, multi-pathway nanomedicine that can successfully reset the hepatic senescent network, providing a promising preclinical pharmacological strategy that warrants future testing in rigorous donor-graft and transplantation settings to fully evaluate its clinical translational potential.

7. Metal-phenolic conducting polymer hydrogels for multifunctional cardiac patches and post-infarction myocardial repair.
[英文摘要] Myocardial infarction (MI) causes irreversible cardiomyocyte loss and adverse ventricular remodeling, yet current cardiac patches remain limited by inadequate bioactivity and poor electrical integration with host myocardium. We report a multifunctional metal-phenolic conducting polymer hydrogel (CPH) for post-infarction myocardial repair. A one-pot process integrates a conductive polypyrrole network with dynamic Cu-tannic acid (TA) coordination, thereby combining electrical conductivity, mechanical support, and redox-related functionality within a single hydrogel. Cu-TA CPH exhibited myocardium-matched mechanical properties (∼19 kPa), suitable conductivity, good biocompatibility, and intrinsic antioxidant and antibacterial activities. These combined features enabled the hydrogel patch to provide mechanical support, facilitate electrical communication, and improve the local microenvironment after MI. On postoperative day 7, left ventricular ejection fraction in the Cu-TA CPH-treated MI group was higher than that in the MI group receiving no material treatment. This therapeutic effect was associated with an increased Bcl-2/Bax ratio, enhanced VEGFR-1 expression, reduced infarct size, and improved myocardial tissue preservation. Collectively, Cu-TA CPH represents a promising hydrogel patch for post-infarction myocardial repair.

8. Platelet membrane-camouflaged Fe-PDAP nanozymes for synergistic photodynamic therapy and immune microenvironment remodeling in breast cancer.
2026-09-23Photodynamic therapy (PDT) is a photosensitizer-based targeted therapeutic approach that has been widely explored in tumor therapy. Nevertheless, the hypoxic features and immunosuppressive status of the tumor microenvironment (TME) can severely limit the therapeutic efficacy of PDT. To address this critical bottleneck, this study constructed a multifunctional biomimetic nanoprobe to enhance PDT efficacy by synergistically alleviating hypoxia and regulating the TME. Using Fe-doped polydiaminopyridine (Fe-PDAP) nanozymes as carriers, the photosensitizer indocyanine green (ICG) and immunomodulator rapamycin (RAPA) were encapsulated, and the nanoparticles were further coated with platelet membrane (PM) to obtain the Fe-PDAP-ICG-RAPA@PM biomimetic nanosystem. Platelet-membrane modification endows nanoparticles with favorable tumor-targeting capacity and effectively improves their accumulation efficiency in tumor tissues. Within the TME, Fe-PDAP works synergistically with ICG to mitigate hypoxia-related limitations of PDT. Meanwhile, RAPA can drive the polarization of immunosuppressive M2 tumor-associated macrophages (TAMs) toward the antitumor M1 phenotype, thereby efficiently remodeling the immunosuppressive microenvironment. In addition, this biomimetic nanosystem possesses dual-modal magnetic resonance/fluorescence imaging capability. In summary, this study establishes a multifunctional biomimetic nanoplatform integrating dual-modal imaging and synergistic therapy. It can serve as a potential combination therapeutic strategy for breast cancer and provide a reference for the optimized design of subsequent multifunctional nanotheranostic systems.

9. A triple-immunomodulatory quercetin nanoplatform to reinforce T-cell function and remodel tumor microenvironment.
[英文摘要] Immune checkpoint inhibitors (ICIs) often exhibit limited clinical efficacy due to insufficient infiltration of T cells in the tumor microenvironment (TME). To address this challenge, we utilized quercetin, an active ingredient of traditional Chinese medicine, as an alternative ICI, and developed a hyaluronic acid-coated manganese-embedded mesoporous silica nanoparticle (HA-MSN(Mn) for TME-triggered co-delivery quercetin (Que) and Mn to potentiate cancer immunotherapy. This nanoplatform exploits the dual pH/redox-responsive MnO bond cleavage within the TME, triggering structural disintegration of the nanoparticles and subsequent controlled release. Through in vitro and in vivo models, we demonstrated that this nanoplatform employs a triple immunomodulatory mechanism to enhance T-cell function and reverse the immunosuppressive TME. Specifically: (1) both Que and Mn could independently promote dendritic cell (DC) maturation, contributing to T-cell activation; (2) Mn and Que could independently facilitate M1 macrophage polarization, thereby recruiting T cells to tumors by secreting immune cytokines; (3) Que loaded HA-MSN(Mn) downregulates PD-L1 expression on tumor cells, overcoming immune escape and enhancing T cell cytotoxicity. Furthermore, the nanoplatform exhibits antitumor effects associated with immune modulation, alongside enhanced inhibition of tumor growth, with minimal off-target toxicity. Importantly, this study provides deeper insights into the multi-immunomodulatory role of quercetin, offering a promising nanotechnology-driven approach for immunotherapy.

10. Polyphenols-functionalized hydroxyapatite nanocomposites/carbohydrazide grafted gelatin/oxidized dextran adhesive hydrogel with osteogenic and angiogenic activity for bone defect repair.
PMID:日期:2026-09-22Current bone adhesives face critical limitations in osseointegration and vascularization, severely restricting their efficacy in bone defect repair. To address these challenges, we report an injectable adhesive hydrogel synergizing tannic acid-functionalized hydroxyapatite nanocomposites and deferoxamine to remodel microenvironment for bone defect repair. Structural characterization via scanning electron microscope confirmed a porous network with homogeneous nanocomposites dispersion. Meanwhile, rheological studies revealed shear-thinning injectability and self-healing capability. Besides, the hydrogel exhibited interfacial adhesion and cytocompatibility, supporting >90% cell viability with enhanced migration and proliferation. Notably, in vitro studies demonstrated multifunctionality: the hydrogel enhanced angiogenic activity through increased endothelial tubule formation. Moreover, the hydrogel upregulated osteogenic differentiation via elevated expression of runt-related transcription factor 2, osteocalcin, and alkaline phosphatase. In a femoral defect rat model, micro-CT analysis revealed greater new bone volume and superior trabecular thickness and trabecular number in hydrogel-treated groups at 8 weeks. Furthermore, histological examination including H&E and Masson staining demonstrated that the hydrogel contributed to newly forming bone and mature collagen deposition. Immunohistochemical evaluation confirmed enhanced neovascularization in vivo. Hence, this work developed an injectable adhesive hydrogel as a promising bone adhesive for bone defect repair through material-mediated microenvironment with synergistic multifunctionality in osteogenesis and angiogenesis.
