1. A NAC-MYB-cysteine regulatory module enhances drought and salt tolerance in soybean.
1. 一个NAC-MYB-半胱氨酸调控模块增强大豆的耐旱性和耐盐性PMID:日期:2026-10-04Drought and salinity are two major factors limiting soybean productivity worldwide, and both stresses can negatively affect plant growth in similar manners by disrupting cellular redox homeostasis and metabolism. However, the regulatory networks coordinating transcriptional control and metabolic adaptation under both types of stress remain poorly understood. Here, we demonstrate that the R2R3-MYB transcription factors GmMYB60a and GmMYB60b serve as core negative regulators of drought and salt tolerance in soybean. Loss-of-function mutations in GmMYB60a/b clearly increased stress tolerance, which was associated with reduced reactive oxygen species accumulation and increased antioxidant capacity. The GmMYB60 proteins directly bind to and transcriptionally repress GmCYS20, a key gene involved in cysteine biosynthesis, thereby reducing cellular cysteine and glutathione pools. Notably, the GmCYS20 protein physically interacts with GmMYB60 in the nucleus and weakens its ability to bind DNA, resulting in the formation of a transcription-metabolic feedback loop that stabilizes redox homeostasis under stress. Furthermore, the stress-responsive NAC transcription factor GmNAC3 directly suppresses GmMYB60 expression, thereby linking environmental signals to metabolic regulation. Together, these findings reveal a hierarchical GmNAC3-GmMYB60-GmCYS20 regulatory module that coordinates cysteine homeostasis and oxidative stress responses by integrating transcriptional repression with metabolic feedback. This work provides mechanistic insights into soybean stress adaptation and identifies promising genetic targets for improving soybean resistance to drought and salinity.

3. SlBPC1 regulates fruit elongation by directly repressing SlOFP2/SlOFP21 and SUN expression in tomato.
3. SlBPC1通过直接抑制番茄中SlOFP2/SlOFP21和SUN的表达来调控果实伸长PMID:日期:2026-10-04Fruit shape is an important agronomical trait that influences consumer preference. Despite the dramatic variations in fruit shape among tomato germplasms, the underlying genes and regulatory mechanisms remain poorly understood. Here, we found that knockout of the transcription factor SlBPC1 leads to elongated fruits, resulting from altered cell division patterns during ovary development. Further studies revealed that SlBPC1 regulates fruit shape by repressing its target genes SlOFP21 and SUN. Overexpression of SlOFP21 resulted in the formation of elongated fruits with an increased fruit shape index. Moreover, SlBPC1 also represses the expression of SlOFP2, a homolog of SlOFP21. Consistently, overexpression of SlOFP2 also led to elongated fruits in tomato. SlOFP2 and SlOFP21 were found to interact with fruit elongation activator SlTRM5 and repressors SlTRM17/20a and SlTRM19. Together, our results revealed an SlBPC1-SlOFP2/SlOFP21/SUN regulatory module that provides new insights and molecular tools for the genetic improvement of fruit shape traits in tomato breeding programs.

5. S-nitrosylation of SlMYB86 within a SlGRF4-SlMYB86-SlGSNOR module enhances nitrate tolerance in tomato.
PMID:日期:2026-09-30Excessive application of nitrogen (N) fertilizers, particularly nitrate (NO ), in protected agricultural systems has led to secondary soil salinization, posing a global threat to crop yield and quality. The regulatory network underlying tolerance to excess nitrate in tomato (Solanum lycopersicum) remains largely unexplored. Here, we report that S-nitrosoglutathione reductase (GSNOR) positively regulates nitrate tolerance by reducing nitric oxide (NO) and S‑nitrosothiol (SNO) accumulation. We then identified the transcription factor SlMYB86 as a direct upstream activator of SlGSNOR, and further found that nitrate‑induced NO triggers S‑nitrosylation of SlMYB86 at Cys68 and Cys263. This post‑translational modification enhances SlMYB86 protein stability by blocking 26S proteasome‑mediated degradation, while also increasing its DNA‑binding affinity and the transcriptional activation of SlGSNOR, thereby enhancing nitrate tolerance. Moreover, Growth-regulating factor 4 (SlGRF4) functions as an upstream regulator of this pathway, activating SlMYB86 expression under nitrate stress and thereby enhancing tomato tolerance to excessive nitrate stress. Collectively, these findings establish a SlGRF4-SlMYB86-SlGSNOR module in which SlMYB86 S‑nitrosylation establishes a negative feedback loop that couples NO signaling to NO clearance, maintaining redox homeostasis and promoting nitrate tolerance in tomato.

6. Grazing alters the response of ANPP to precipitation variability in a desert steppe.
PMID:日期:2026-09-25Precipitation is a pivotal driver of aboveground net primary productivity (ANPP) in grasslands, while grazing regulates ANPP by reshaping plant community structure. However, how grazing regulates grassland responses to precipitation variability remains incompletely understood. Here, we conducted an 18-year field grazing experiment in a desert steppe, combining long-term ANPP monitoring with daily precipitation data. Our results revealed that grazing significantly reduced community ANPP by 31% and forb ANPP by 61%. This reflected a shift from palatable forbs to stress-tolerant grasses, primarily driven by increased abundance of Stipa breviflora. June-July precipitation amount emerged as the primary driver of inter-annual ANPP variability. Critically, significant interactions between grazing and precipitation demonstrated that grazing fundamentally altered these precipitation-ANPP relationships, strengthening the response slope of grasses while weakening that of forbs and the whole community. Although grazing did not significantly alter legacy effects at the community or grass level, it significantly modulated the legacy effect on forb ANPP; previous-year precipitation remained a significant predictor of ANPP across all organizational levels. Grazing-induced phenotypic adjustments in S. breviflora (increased root length and population density) strengthened the coupling between grasses and early- and mid-summer precipitation, even though their precipitation-use efficiency did not differ between treatments. Our findings demonstrate that grazing reorganizes ecosystem sensitivity to climate by shifting plant functional group dominance and key species traits. These findings provide a mechanistic basis for forecasting grassland responses to climate variability and for guiding grazing management under seasonal precipitation forecasts.

7. RNA regulatory landscapes in fleshy fruits: From regulation mechanisms to quality design.
PMID:日期:2026-09-25Fruit quality is a complex trait shaped by developmental programs, metabolic networks, and environmental responses; yet, the RNA-level regulatory processes that determine functional outputs remain insufficiently integrated and underexplored. In this review, we provide an integrated overview of RNA-mediated regulatory mechanisms underlying fleshy fruit quality formation, focusing on five major layers, including RNA modification, organellar RNA editing, alternative splicing, long non-coding RNAs, microRNAs, and their crosstalk. We summarize their molecular modes of action and discuss how these regulatory mechanisms shape diverse quality traits, including fruit morphology, pigmentation, flavor, texture remodeling, ripening progression, and postharvest performance. Furthermore, we highlight emerging strategies for harnessing RNA-mediated regulation to improve and maintain fruit quality, including RNA-centered multi-omics integration, AI-driven design of RNA regulatory elements, and precision of RNA-level manipulation. We propose that deciphering and manipulating post-transcriptional regulatory codes will provide a conceptual and technological framework for shifting fruit improvement from empirical selection toward rational RNA-guided quality design, with broad implications for crop improvement, nutritional enhancement, and sustainable postharvest management.

9. Pan-genomic analysis of alternative polyadenylation reveals a 3'UTR remodeling regulatory layer controlling salt tolerance in rice.
9. 替代多聚腺苷化的泛基因组分析揭示了控制水稻耐盐性的3'UTR重塑调控层PMID:日期:2026-09-22Alternative polyadenylation (APA) generates mRNA isoforms with variable 3'UTR lengths, yet its genetic basis and functional significance at the population level remain unexplored in plants. Here, we present the population-level APA landscape and the APA quantitative trait loci (3'aQTL) map in rice, through profiling of over 200 rice accessions under normal and salt-stress conditions. We identified 19,938 and 22,251 APA events under normal and salt-stress conditions, respectively, and demonstrate that salt stress globally induces 3'UTR shortening via proximal polyA site selection, which can release miRNA-mediated repression on stress-response genes. We further mapped 1,634 and 1,809 cis-3'aQTLs under normal and salt-stress conditions, respectively, which were markedly enriched in 3'UTRs and transcription termination sites, in contrast to expression QTLs that were enriched in promoter regions. This APA genetic framework reveals a post-transcriptional regulatory layer distinct from transcriptional control. Functional validation of two 3'aQTL-associated genes, OsMyb21 and OsRNS3, through CRISPR knockout and 3'UTR deletion confirmed their roles in salt tolerance via APA-mediated regulation. Our findings provide a community resource for understanding post-transcriptional regulation of stress responses and offer genetic targets for breeding salt-tolerant rice varieties.

10. Beyond hypoxia: Context-dependent transcriptional reprogramming by ERFVIIs.
10. 超越缺氧:ERFVIIs依赖环境的转录重编程PMID:日期:2026-09-22This Commentary highlights the finding that an increased post-submergence oxidative burst reprograms plant cysteine oxidase (PCO)-group VII ethylene response factor (ERFVII) oxygen sensing, inhibiting PCO activity and transcriptionally rewiring ERFVIIs toward oxidative stress recovery. This redefines ERFVIIs as redox-responsive stress integrators and reveals targets to enhance climate-adaptive stress responses.
