1. CRISPR-Programmable Nanoparticle Transport in Salt-Gated Porous Media for Visual DNA Detection.
2026-07-18Paper-based diagnostics typically treat porous cellulose as a passive capillary medium, with nanoparticle transport governed by channel geometry and capture chemistry. Here we show that CRISPR-programmable nanoparticle surface chemistry can actively regulate transport in paper through a salt-activated adhesion mechanism. Gold nanoparticles functionalized with hydrophobic Cy5-labeled DNA exhibit strong adhesion to cellulose fibers at elevated ionic strength (~1.5 M local NaCl generated upon rehydration of preloaded salt), whereas particles lacking hydrophobic domains migrate freely. This phenomenon enables a distance-based visual readout, in which the length of the colored nanoparticle front provides a semi-quantitative measure of DNA target concentration. Optical and electron microscopy reveal that nanoparticle immobilization arises from extensive fiber adhesion rather than pore occlusion or particle aggregation. As a proof of concept, coupling Cas12a-mediated cleavage of Cy5-labeled DNA probes to this salt-gated transport system enabled naked-eye, distance-based detection of Herpes simplex virus 1 (HSV-1) DNA with a limit of detection of 100 aM in ~45-60 min. Evaluation of 40 clinical samples showed excellent agreement with qPCR, achieving 96.2% sensitivity and 100% specificity. More broadly, this work establishes a general strategy for programming nanoparticle transport in porous media by encoding functionality directly into nanoparticle surface chemistry.

2. Bioprinting of miRNA-Induced Spheroids for Vascularized, Heterocellular Bone Regeneration.
[英文摘要] Successful bone regeneration requires coupled osteogenic and vascular development; however, achieving simultaneous multicellular differentiation within engineered tissues remains challenging. Here, we developed a microRNA (miR)-guided spheroid platform to induce dual osteogenic and endothelial differentiation of human adipose-derived stem cells (hASCs) for vascularized bone regeneration. hASCs were transfected with miR-148b or miR-210 to promote osteogenic and vascular-associated phenotypes, respectively, and assembled into spheroids that were bioprinted within an nHA-containing GelMA microgel environment using aspiration-assisted bioprinting (AAB). The integrated platform combined miR-guided osteogenic and endothelial differentiation, spatially organized AAB-based spheroid assembly, and an nHA-containing GelMA microgel environment to support vascularized bone tissue regeneration. The engineered constructs maintained high cell viability (> 90%) and supported active cell spreading and migration within the microgel matrix, together with increased osteogenic and endothelial gene expression. To further verify their regenerative potential, the constructs were implanted into mouse critical-size calvarial defects, where those containing miR-transfected hASCs improved bone regeneration, achieving ~91% of defect closure, and promoted the formation of vessel-like CD31-positive structures compared to controls. Together, these findings demonstrate that combining miR-mediated dual-lineage differentiation with spatially organized spheroid assembly and a supportive microgel environment provides a promising strategy for vascularized bone tissue engineering.

3. Remediation of Pb contaminated soil with recovered P-laden engineered biochar: Effects on Pb speciation and soil microbial community.
2026-03-15This study investigates the concept of using engineered biochar to reclaim phosphorus (P) in water and subsequent reuse of recovered P-laden engineered biochar for remediation of lead (Pb) contaminated soil. Hickory wood biochar (BC) was loaded with Mg oxides/hydroxides (EBC) via wet-impregnation pretreatment and Mg/Fe layered double hydroxides (LDH) via co-precipitation, respectively. Both engineered biochar types demonstrated effective P adsorption, with maximum capacities of 196.0 mg P/g for EBC and 88.3 mg P/g for LDH. The recovered P-laden biochar (EBC-P and LDH-P) were incubated with a Pb contaminated soil for four months, and changes in labile Pb content, Pb speciation, and soil microbial community were monitored. EBC-P and LDH-P significantly reduced extractable Pb (CaCl and TCLP methods) by over 95% stabilization efficiency. Soil XRD spectra indicated gradual crystallization of hydroxypyromorphite within the first month, yet this Pb mineral phase was not detected in later months by XANES. Pb was consistently bonded on biochar surfaces across all treatments. Biochar applications also altered the soil microbial community structure, increasing the relative abundance of taxa (e.g., strains) in association with increased soil pH and reduced Pb toxicity. This change suggests enhanced microbial functionalities to cope with environmental stresses such as pH and heavy metals as well as assimilation and generation of precursor metabolites. Ultimately, this study provides a technical foundation for evaluating P-enriched engineered biochar as a sustainable strategy for remediating Pb-contaminated soils.

4. Targeted Photodynamic Therapy for Multispecies Dental Biofilms Using Liposome Encapsulated p-Toluquinone.
2026-02-10Oral infections remain highly prevalent among individuals with limited access to routine dental care. Blue light (BL) offers a promising alternative for managing these infections, but its efficiency is limited without BL-activated photosensitizers. Here, we engineered an ionizable liposome to encapsulate a newly identified BL-activated photosensitizer, p-toluquinone (p-TQ). The resultant p-TQ@Lipo responded to the acidic pH produced by bacteria, facilitating the targeted delivery of the cargo to the pathogens. p-TQ@Lipo had a pH-dependent surface potential change from +29.5 ± 2.1 mV at pH=5, to +8.2 ± 0.1 mV at pH=7, and -6.4 ± 0.5 mV at pH=8. When combined with 405 nm BL at a fluence as low as 27 J/cm, p-TQ@Lipo reduced bacterial load by >99%, with minimal cytotoxicity to human gingival fibroblasts, achieving a remarkable 359-fold improvement in selectivity over free p-TQ. Confocal microscopy validated more efficient delivery of the cargo to Gram-negative than to Gram-positive bacteria, reflecting differences in bacterial surface charges. We then established polymicrobial dental biofilms in vivo and in vitro, comprising BL-sensitive and -refractory and Gram-positive and -negative species. The polymicrobial biofilms were efficiently disrupted on ferret tooth surfaces in an ex vivo infection model and eradicated in an in vivo mouse oral infection model by repeated therapies of p-TQ@Lipo combined with BL, whereas either treatment alone was ineffective. Importantly, the treatment significantly decreased periodontal inflammation and hyperplasia. These findings establish p-TQ@Lipo plus BL as a safe and effective antimicrobial platform with translational potentials for oral infection management.

5. NeuProIO: A dual-Action Solid Drug Nanoparticle Topical Formulation for Simultaneous Intraocular Pressure Reduction and Neuroprotection for Glaucoma.
2026-01-26Glaucoma is a leading cause of irreversible blindness, primarily due to elevated intraocular pressure (IOP) that damages the retina and optic nerve. Most topical glaucoma treatments focus solely on IOP-lowering without direct neuroprotection, as drug delivery to the posterior eye is challenging. Here, we developed NeuProIO, a dual-action solid drug nanoparticle (NP) eye drop for simultaneous IOP reduction and neuroprotection. Using flash nanoprecipitation in a multi-inlet vortex mixer, we fabricated the first NeuProIO solid drug NPs incorporating the repurposed neuroprotective agent maprotiline (MAP) and the IOP-lowering agent betaxolol (BX), forming MAP/BX NPs for synergistic glaucoma treatment. The NeuProIO improved stability and ocular biocompatibility versus conventional eye drops and markedly enhanced corneal transport. Single topical dosing sustained an effective IOP control (15% IOP reduction or higher) for 72 h. Beyond its IOP-lowering effect, BX significantly enhanced the posterior segment delivery of MAP in MAP/BX NPs, resulting in retinal and optic nerve MAP levels that were approximately 5-fold and 2-fold higher, respectively, than those observed with MAP alone. In silicone oil-induced ocular hypertension glaucoma mouse model, the MAP/BX combination exhibited significantly improved neuroprotection effects than MAP monotherapy, including >21% greater ganglion cell complex thickness, >7-fold axon preservation, and >4-fold retinal ganglion cell survival. A comprehensive biosafety evaluation confirmed the safety of NeuProIO, with assessments of corneal thickness, RGC viability, and major organ histology revealing no adverse local or systemic effects. Taken together, NeuProIO has the potential to redefine glaucoma treatment, offering a more effective and long-lasting solution to preserving vision and improving patient outcomes.

6. Investigating Nanoparticle Distribution and Tissue Architecture in Male and Female Human Cardiac Organoids.
2026-01-22Nanoparticles (NPs) are increasingly recognized as vital tools in targeted cardiac therapies, diagnostics, and tissue regeneration. Despite notable advancements, a comprehensive understanding of NP interactions within cardiac tissue as a whole remains limited. In this study, we employ human cardiac organoids derived from induced pluripotent stem cells to investigate NP distribution and colocalization by cardiomyocytes (CMs) and epicardial cells. We also examine the influence of organoid sex on structural organization, biomechanics, and whether such variations impact NP biodistribution and cellular internalization. Using fluorescent polystyrene NPs combined with cryo-electron, confocal, and super-resolution microscopy, RNA sequencing, and atomic force microscopy (AFM), we compare male and female organoids across multiple parameters. Our findings reveal that NPs preferentially accumulate in regions rich in CMs and extracellular matrix, with CMs showing significantly higher colocalization with NPs than epicardial cells. While super-resolution microscopy and AFM analyses demonstrate substantial architectural and mechanical differences between male and female organoids, no significant sex-based differences in NP distribution or colocalization/proximity were observed. Overall, our findings suggest two key points: (i) CMs play a pivotal role in NP-mediated cardiac therapies, and (ii) the development of cardiac organoids should incorporate additional features, such as immune system components, to better mimic native heart tissue. Such advancements would enhance the utility of organoids as robust models for studying drug and NP interactions.

7. {"_":"Fully Automated [F]AlF Radiolabeling Using a Microdroplet Reactor Platform.","sup":["18"]}
2026-01-09The preparation of F-labeled biomolecules for PET imaging is often challenging due to complex synthesis protocols and the short half-life of F-18 (109.8 min). The aluminum-[F]fluoride ([F]AlF) method offers a straightforward, efficient route for radiofluorinating biomolecules, with demonstrated utility in both preclinical and clinical imaging. However, one technique's flaw is the mismatch between the typical microvolume conditions required for high-yield [F]AlF chelation and the milliliter-scale operation of commercial automated radiosynthesizers. To address this shortcoming, we leverage microscale radiochemistry techniques and report the first fully-automated, droplet-based production of [F]AlF-labeled compounds using a compact and cost-effective platform. The optimized process was first achieved using a NODA analogue (NODA-Tz) as a model substrate at a 15 μL reaction scale, and was subsequently applied to the fully automated synthesis of the clinically-relevant PET probe [F]AlF-FAPI-74 which targets fibroblast activation protein expressed in most cancers. [F]AlF-FAPI-74 (n = 3) was obtained with high radiochemical yield (RCY, 75 ± 5%), radiochemical purity (RCP, >99%), activity yield (67 ± 4%), and apparent molar activity ( , 358-482 GBq/μmol). Starting from 2.6-3.5 GBq of [F]fluoride, the final product quantity (1.8-2.4 GBq) was sufficient for multiple patient doses. Compared to the conventional vial-based methods, the microdroplet approach showed significant improvements including reduced synthesis time (19 ± 1 min vs. 31 min), higher yield, and reduced precursor consumption (5 nmol vs. 80 nmol). This work demonstrates the feasibility and advantages of translating [F]AlF chemistry to a microdroplet platform, enabling more efficient radiotracer production and broader clinical accessibility.

8. 4D Printing of a Bioadhesive Shape Memory Elastomer.
2025-12-17Tissue adhesive materials for biomedical applications demand exceptional stretchability, flexibility, and robust adhesion. Herein, we introduce a bioadhesive shape memory elastomer (SME) that is 4D printed by digital light processing (DLP). The ink for printing this SME consists of N-vinylpyrrolidone (NVP), dodecyl acrylate (DA), and a new Poly(ethylene glycol-co-dodecanedioic acid) diacrylate (AcP) prepolymer. By employing AcP, a linear oligomer with tailored hydrophobic and hydrophilic segments, as a crosslinker, the resulting poly(AcP-DA-NVP) SME exhibits impressive mechanical and adhesion performance. Depending on the weight ratios of the ink precursors, this bioadhesive SME shows tunable transition temperatures in a range of 9-68 °C, Young's modulus of up to 24 MPa, and tensile strength of ~ 18.5 MPa, stretchability of ~ 700%, and adhesion strengths of ~ 600 kPa to dry/wet aluminum foils and 90 kPa to porcine skin. Empowered by these combinational properties, the SME offers an unprecedent capability of being a shape-programmable, adhesive substrate for on-skin soft electronics and shows potential for tissue engineering, wound healing, and biomedical implantation.

9. Enhancing Arsenate Adsorption Kinetics in Iron(III)-Crosslinked Chitosan Beads.
2025-10-16Iron(III)-crosslinked chitosan (Fe-Ch) offers great potential for the sustainable removal of arsenic from contaminated water sources due to its high adsorption capacity and selectivity for As(V) over background competitors such as phosphate (P(V)). However, this material can have low surface area and high activation energy for chemisorption, which can contribute to a low adsorption rate limiting its scalability. This study investigated parameters impacting the adsorption kinetics of Fe-Ch beads and assessed methods to improve their performance. Batch adsorption experiments and molecular dynamics simulations demonstrated that the identity of anions present during synthesis plays a significant role in the final structure and degree of crosslinking of the beads. Acetate in the synthesis was found to lead to a lower degree of crosslinking and higher surface area, enhancing adsorption kinetics. Moreover, glycine in the synthesis was found to lead to the highest adsorption rates, as its interaction with iron increases the hydrophilicity of the beads and may provide a coordination environment that has higher affinity for arsenate than crosslinked chitosan evidenced by density function theory calculations. We also compared the impact of drying techniques on adsorption performance. Freeze-dried beads were found to be rate-limited by chemisorption while air-dried beads were found to be rate-limited by intraparticle diffusion until the beads reached a swollen state. With the approaches evaluated in this study, the pseudo-second-order rate constant of Fe-Ch beads was increased by 24-fold compared to literature studies without altering selectivity, enhancing their suitability for As(V) remediation.

10. Tunable lysis of functionalized nanocultures for efficient microbial and nucleic acid recovery.
PMID:日期:2025-09-22The cultivation of low-abundance and fastidious microorganisms remains a critical challenge in microbiology. To overcome this, we developed -a microencapsulation platform that enables high-throughput cultivation of microbes under near-native conditions. These nanoliter-sized polymeric microcapsules are generated via droplet microfluidics and provide a mechanically robust yet tunable environment for microbial growth, isolation, and interrogation. By engineering the capsule shell chemistry, we control the mechanical properties to allow on-demand lysis and efficient retrieval of intracellular contents. We assessed three physical disruption methods-osmotic swelling, sonication, and bead beating-to evaluate their performance in releasing viable cells and nucleic acids from nanocultures. Osmotic swelling yielded the highest recovery of both viable microbes and high-integrity nucleic acids, outperforming the other methods. This approach preserves the biological relevance of the cultivated communities while enabling downstream processing for species identification and functional analysis. Nanocultures offer a scalable, high-throughput strategy to culture diverse microbial species in environments that mimic natural habitats. Their mechanical tunability provides precise control over microcapsule lysis and content recovery, positioning them as a powerful tool for microbial discovery, diagnostics, and synthetic ecology.
