1. Vinylogous Mannich reaction in total synthesis of alkaloids.
PMID:日期:2026-10-02Covering: up to July 2026The vinylogous Mannich reaction (VMR) enables γ-selective carbon-carbon bond formation between vinylogous nucleophiles and imines or iminium ions, yielding δ-aminocarbonyl frameworks. Compared with the classical Mannich reaction, VMR shows distinct advantages in remote functionalization and convergent bond formation, facilitating the rapid assembly of nitrogen-containing heterocycles, multiple stereogenic centers, and densely functionalized intermediates. As a result, this reaction has become an increasingly valuable strategy for generating complexity in the total synthesis of alkaloids and has been successfully applied to a wide range of alkaloid families, including piperidine, pyrrolidine, indole, , , phthalideisoquinoline, steroidal, and . This review presents a structured and concept-oriented overview of VMR-based alkaloid synthesis. Representative applications are organized according to the nature of the vinylogous nucleophiles, including both acyclic and cyclic dienol equivalents. Particular emphasis is placed on reaction design, stereochemical control, complexity generation, and the integration of VMR into asymmetric, collective, and biomimetic synthetic strategies. Overall, this review highlights the evolution of VMR from a methodological variant of the classical Mannich reaction into a broadly applicable approach for the efficient synthesis of structurally diverse alkaloids. It also provides perspectives on its future development in alkaloid synthesis.

2. {"_":"Expanding bile acid diversity host and microbial conjugation pathways.","i":["via"]}
PMID:日期:2026-09-24Covering: 2019 to April 2026Bile acids, originally identified as digestive emulsifiers, are now recognized as signaling molecules at the nexus of metabolism and immunity. Recent advances in metabolomics have revealed unexpected chemical diversity of BAs, derived from previously unrecognized conjugation pathways involving both the gut microbiota and the host. This includes microbial conjugation of the bile acid carboxy terminus with diverse amino acids and other biogenic amines as well as host-derived conjugation with cysteamine derivatives. In addition, microbial acylation of the steroid core has been shown to introduce succinyl, acetyl, and other chemical modifications, including expansion of the canonical bile acid carbon skeleton. This review synthesizes recent discoveries (2019-2026) in bile acid conjugation chemistry and highlights how the expanded bile acid diversity regulates host physiology through activation of different nuclear receptors and G-protein coupled receptors, including therapeutic implications for metabolic and immune disorders such as non-alcoholic steatohepatitis, inflammatory bowel disease, and cardiometabolic diseases. The emerging picture reveals that bile acid pools result from an intricate network of host-microbe metabolic pathways, and that considering both host and microbial contributions is essential for understanding bile acid biology in health and disease.

3. The roles of glycosyltransferases in shaping flavonoid diversity in plants.
PMID:日期:2026-09-21Covering: up to 2026Flavonoids belong to one of the largest families of specialized metabolites in plants and play pivotal roles in plant adaptation and human health. In their biosynthesis, the formation of core aromatic structures is followed by various chemical modifications that further decorate these scaffolds, shaping their chemical diversity, biological functions, and physicochemical properties. Among these modifications, glycosylation is the most prevalent, transferring monosaccharides from activated nucleotide sugars to acceptor molecules to form glycosidic bonds. This reaction is catalyzed by UDP-glycosyltransferases (UGTs), a class of enzymes that generate structurally diverse glycosides from a relatively limited set of flavonoid core structures. Through these catalytic processes, UGTs have driven the structural and functional diversification of flavonoids across plant lineages. This review highlights the structure, biochemistry, and phylogeny of UGTs participating in flavonoid biosynthesis. We discuss how substrate promiscuity, together with broad specificity toward both sugar donors and acceptor substrates, allows UGTs to expand flavonoid chemical diversity. Additional focus is placed on how gene expression is integrated into tissue-specific and stress-responsive regulatory networks that shape flavonoid diversity, and how UGT-dependent glycosylation plays a critical role in the stabilization and subcellular sequestration of diverse flavonoids. Lastly, we highlight emerging strategies for engineering plant flavonoid UGTs and glycosylation pathways, including the opportunities and limitations of computational and artificial intelligence-assisted approaches.

5. Advancing biosynthetic pathway discovery through short-read-directed long-read sequencing.
PMID:日期:2026-09-21Time span of literature: 2020-todayMetagenomic methods have rapidly advanced, enabling the identification of biosynthetic pathways directly from complex microbiome data. Short-read sequencing, while accurate and cost-effective, often generates fragmented assemblies that can lead to incomplete biosynthetic gene cluster (BGC) recovery. Although long-read sequencing offers a solution to the fragmentation problems, technical requirements and higher costs have limited its scalability. Here, we examine BGC fragmentation in short-read sequencing data across large databases of metagenome-assembled genomes (MAGs) and estimate the targeted genome contiguity required to recover 'complete' biosynthetic gene clusters. We argue that the increasing availability of MAGs recovered from short-read metagenomes with recent advancements in ultra-low input DNA amplification for high-fidelity PacBio sequencing-now requiring as little as nanograms of DNA-can be used sequentially to boost biosynthetic pathway discovery. We demonstrate how natural products researchers can benefit from using short-read MAG comparisons to guide targeted long-read re-sequencing efforts with low amounts of input DNA and/or limited financial resources. Our analysis provides strategic recommendations for the broader scientific community on how to best leverage the strengths of short- and long-read sequencing data to efficiently allocate resources and accelerate natural product discovery.

6. Radical SAM-dependent enzymes: Jacks-of-all-trades in RiPP pathways.
PMID:日期:2026-09-15Covering: up to 2026The cofactor -adenosyl-L-methionine (SAM) plays a key role in the biosynthesis of ribosomally synthesised and post-translationally modified peptide (RiPP) natural products. SAM-dependent enzymes have been shown to catalyse important steps in RiPP biosynthesis, including methylation, desaturation, epimerisation, splicing reactions, and oxidative decarboxylation. Furthermore, a range of SAM-dependent cross-linking reactions result in the formation of complex macrocyclic peptide products, which often exhibit enhanced stability and intriguing biological activities. These post-translational modifications occur in all parts of the peptide molecule, including side chains, peptide backbones, and N- and C-termini. Newly identified RiPP pathways continue to reveal interesting SAM-dependent enzymes that install remarkable chemical moieties. In view of the recent advances in this field, the objective of this review is to provide an overview of the significant chemical diversity of RiPP natural products generated by SAM-dependent enzymes, with a particular focus on post-translational modifications introduced by radical SAM enzymes.
