[英文摘要] Management of wounds unveils various crucial clinical constraints, emphasizing the demand for transformed wound dressings that maximize the healing process and facilitate tissue regeneration. Here, a simple, green yet scalable approach was established to fabricate a multifunctional nanocomposite hydrogel employing plant- and animal-derived polymers, specifically almond gum and gelatin, enhanced with bioactive L-menthol and Ae-ZnONPs (MeZ@ALGEL), to improve wound healing by reinforcing the wound environment with its bactericidal, reactive oxygen species (ROS) regulation, and tissue regeneration abilities. MeZ@ALGEL exhibits necessary physicochemical and mechanical properties and significant antibacterial and antioxidant activities, making it ideal for topical administration and beneficial in accelerating wound repair. Under in vitro conditions, MeZ@ALGEL demonstrates a high degree of biocompatibility with fibroblast cells, actively promotes their migration in wound-like cases, and indicates improved hydroxyproline content with upregulation of , , genes, and downregulation of the gene revealed in gene expression analysis. In vivo studies using as a preliminary wound model reveal that the MeZ@ALGEL hydrogel is nontoxic up to the concentration of 125 μg/mL, promotes healing of glass wool-mediated wounds, exhibits strong antimicrobial activity, and accelerates wound repair through a multifaceted approach involving bacterial eradication, regulation of ROS, and stimulation of collagen synthesis, ultimately enhancing wound healing in the model organism. By these findings, the facilely synthesized nanocomposite hydrogel opens up the possibilities for developing multifunctional hydrogels of natural origin by utilizing the synergistic interaction of components and offers great potential in the wound care sector.

3. Ascorbic acid-loaded gellan-g-poly(ethylene glycol) methacrylate matrix as a wound-healing material.
[英文摘要] Ascorbic acid (AA) is one of the important biomolecules involved in all phases of wound healing. The aim of this study was to develop a new hydrogel system that offers topical delivery of ascorbic acid to wounds during wound care management. In this work, we grafted poly (ethylene glycol) methacrylate onto a renewable biopolymer gellan, and the graft copolymer (GPMA) formed was crosslinked covalently and ionically, and used as a matrix for delivering AA to the wounds. By the processes of grafting and crosslinking, the mechanical properties of the gellan increased several fold compared to mechanically weak native gellan. In vitro cytotoxicity evaluation showed that GPMA was non-cytotoxic to fibroblast cells. GPMA hydrogel matrix allowed the sustained release of AA. When AA was incorporated in GPMA, a significant improvement in wound closure was observed in scratch wound assay performed with keratinocytes. Since AA acts as a cofactor in collagen synthesis, the controlled delivery of AA to the wound microenvironment favors the up-regulation of colα1 gene expression. This study revealed that ascorbic acid, at a concentration of 150 μM, has a favorable impact on wound healing when tested in vitro. Overall results indicate that the GPMA matrix could be a promising material for wound healing applications.

[英文摘要] Hydrogel-based scaffolds have been widely investigated for their use in tissue engineering to accelerate tissue regeneration. However, replicating the physiological microenvironments of tissues with appropriate biological cues remains challenging. Recent advances in gradient hydrogels have transformed tissue-engineering research by providing precise structures that mimic the extracellular matrix of natural tissues. Unlike conventional homogeneously structured hydrogels, gradient hydrogels provide a better bio-mimicking microenvironment for combined cell therapies in chronic wound treatment by regulating various cell behaviors, such as proliferation, migration, and differentiation. Here, we present the integration of L929 mouse fibroblast spheroids into gradient hydrogels to mimic the dermal stiffness microenvironment and we investigated their impact on full-thickness skin regeneration. A stiffness gradient was achieved by modulating the concentration of methacrylated hyaluronic acid (HA-MA) with varying degrees of methacrylation, using a dual-syringe pump system. The encapsulation of L929 spheroids with gradient hydrogel facilitated skin cell organization in a hierarchically ordered configuration, leading to full-thickness wound healing that was 1.53 times faster than the untreated group in a rat model. This study provides a method for investigating the potential role of gradient hydrogels in various tissue engineering and regeneration applications.

[英文摘要] Chronic diabetic wounds are a severe complication of diabetes, often leading to high treatment costs and high amputation rates. Numerous studies have revealed that nitric oxide (NO) therapy is a promising option because it favours wound revascularization. Here, base-paired injectable adhesive hydrogels (CAT) were prepared using adenine- and thymine-modified chitosan (CSA and CST). By further introducing S-nitrosoglutathione (GSNO) and binary l-arginine (bArg), we obtained a NO sustained-release hydrogel (CAT/bArg/GSON) that was more suitable for the treatment of chronic wounds. The results showed that the expression of HIF-1α and VEGF was upregulated in the CAT/bArg/GSON group, and improved blood vessel regeneration was observed, indicating an important role of NO. In addition, the research findings revealed that following treatment with the CAT/bArg/GSON hydrogel, the viability of Staphylococcus aureus and Escherichia coli decreased to 14 ± 2 % and 6 ± 1 %, respectively. Moreover, the wound microenvironment was improved, as evidenced by a 60 ± 1 % clearance of DPPH. In particular, histological examination and immunohistochemical staining results showed that wounds treated with CAT/bArg/GSNO exhibited denser neovascularization, faster epithelial tissue regeneration, and thicker collagen deposition. Overall, this study proposes an effective strategy to prepare injectable hydrogel dressings with dual NO donors. The functionality of CAT/bArg/GSON has been thoroughly demonstrated in research on chronic wound vascular regeneration, indicating that CAT/bArg/GSON could be a potential option for promoting chronic wound healing. STATEMENT OF SIGNIFICANCE: This article prepares a chitosan hydrogel utilizing the principle of complementary base pairing, which offers several advantages, including good adhesion, biocompatibility, and flow properties, making it a good material for wound dressings. Loaded GSNO and bArg can steadily release NO and l-arginine through the degradation of the gel. Then, the released l-arginine not only possesses antioxidant properties but can also continue to generate a small amount of NO under the action of NOS. This design achieves a sustained and stable supply of NO at the wound site, maximizing the angiogenesis-promoting and antibacterial effects of NO. More neovascularization and abundant collagen were observed in the regenerated tissues. This study provides an effective repair hydrogel material for diabetic wound.

[英文摘要] Despite the advancements of pharmacological treatments and gauze dressings in the field of skin wound healing, these methods present numerous limitations. Therefore, developing a multifunctional material capable of efficiently promoting skin wound healing is particularly crucial. Citric acid (CA)-modified chitosan (CS) loaded with Shikonin (SK) (CA-CS-SK) hydrogel was prepared via the freeze-thaw method. The physical properties of the hydrogel were profiled through FTIR, SEM, rotational rheometry, swelling experiment, degradation rate analysis, and drug release experiments. Furthermore, the biocompatibility of the hydrogel was comprehensively evaluated through hemolysis assay, CCK-8 cytotoxicity detection, and live/dead cell staining. Antimicrobial activity against E. coli and S. aureus of the hydrogel was gauged in vitro, and its therapeutic performance was ultimately validated in a mouse full-thickness wound model through H&E staining and ELISA. The CA-CS-SK hydrogel exhibited appropriate rheological properties, swelling ratio, degradation rate, and drug release rate. It effectively suppressed the proliferation of E. coli and S. aureus, with superior inhibitory effects compared to CA-CS hydrogel and SK alone. Additionally, the hydrogel showed no significant toxicity to human dermal fibroblasts and did not cause erythrocyte rupture. Animal model experiments demonstrated that, compared to cotton gauze, CA-CS hydrogel, and SK, the CA-CS-SK hydrogel reduced levels of TNF-α and IL-6 at the wound site, alleviated the inflammatory response, and promoted wound healing. The CA-CS-SK hydrogel possesses high antibacterial activity, excellent biocompatibility, and efficient wound healing promotion capabilities, making it a highly promising material for skin wound treatment.

[英文摘要] Skin wound healing will become a pressing and difficult problem following injury to the skin structure. Persistent wounds, in particular, become more vulnerable to bacterial infections, which can contribute to persistent skin inflammation. Therefore, it is critical to create a wound dressing that promotes wound healing while also being antimicrobial. In the present work, a multifunctional biological activity hydrogel formed by enzymatic cross-linking was developed by introducing graphene oxide (GO) and lactoferrin to gelatin hydrogel. Furthermore, by incorporating lactoferrin, the composite hydrogels exhibit excellent in vitro antibacterial and biocompatibility. According to cell experiments, the LTF-GO/Gel hydrogel can improve wound healing by enhancing L929 cell migration. Interestingly, under near-infrared light, LTF-GO/Gel hydrogel increases the generation of singlet oxygen (O) and hydroxyl radical (-OH), making the hydrogel system excellent antioxidant and antibacterial capabilities, these results demonstrate that the LTF-GO/Gel hydrogel has clinical promise as a wound dressing for wound healing. In vivo experiments unequivocally establish the capacity of the LTF-GO/Gel hydrogel to expedite wound healing and mitigate inflammation. This hydrogel, therefore, harbors immense potential for applications in wound healing.
