2. The relationship between plasma long-chain fatty acids and hypertension and its underlying molecular mechanisms.
PMID:日期:2026-09-01The associations between plasma long-chain fatty acids and hypertension remain incompletely understood, particularly in middle-aged and older adults, and the underlying mechanisms are still unclear. This study aimed to investigate the associations between plasma long-chain fatty acids and hypertension and to explore the underlying molecular mechanisms using an integrated network pharmacology and molecular docking approach. We included 2092 middle-aged and older adults with available measurements of plasma long-chain fatty acid concentrations. Stepwise adjusted logistic regression and Bayesian kernel machine regression were used to assess the associations of individual fatty acids and mixed fatty acid exposure with hypertension. Potential targets of fatty acids and hypertension were identified from public databases, followed by protein-protein interaction network construction, hub target identification, enrichment analyses, and molecular docking. After adjustment for sex, age, sociodemographic characteristics, lifestyle factors, and body mass index, myristoleic acid, arachidonic acid, erucic acid, and docosahexaenoic acid were inversely associated with hypertension, whereas eicosapentaenoic acid and docosatetraenoic acid were positively associated. Mixed-exposure analysis suggested a modest overall effect, with the main contributions arising from these key fatty acids. MAPK3, PTGS2, and ESR1 were identified as hub targets and were mainly involved in inflammation, immune responses, lipid metabolism, and vascular function. In conclusion, plasma long-chain fatty acid profiles were associated with hypertension in middle-aged and older adults and may provide new epidemiological evidence and mechanistic insights into hypertension risk.

3. Metabolic syndrome risk occurs at relatively low adiposity levels in Korean adults.
PMID:日期:2026-08-30Body mass index (BMI) is widely used to assess obesity-related cardiometabolic risk, but the adiposity measures corresponding to a given BMI may differ across populations. We hypothesized that in Korean adults, a BMI of 25 kg/m² would correspond to substantial metabolic syndrome (MetS) risk at lower adiposity levels than previously reported in Western populations and that BMI and adiposity measures would demonstrate differential discriminatory performance for MetS. We analyzed adults aged 18-80 years from the Korea National Health and Nutrition Examination Survey were analyzed, including 3023 participants with dual-energy X-ray absorptiometry (DXA)-derived whole-body fat percentage (WBF%) and trunk fat percentage (2008-2011) and 9185 participants with bioelectrical impedance analysis (BIA)-derived WBF% (2022-2023). Age- and sex-adjusted complex-sample logistic regression was used to estimate the probability of MetS at BMI 25 kg/m² and the corresponding adiposity measures. The estimated prevalence of MetS at BMI 25 kg/m² was 29.6% in DXA-based dataset and 29.7% in BIA-based dataset. Corresponding WBF% values were 22.3% in men and 33.2% in women in DXA-based dataset, and 24.7% and 33.2%, respectively, in BIA-based dataset. Corresponding trunk fat percentages were 24.4% in men and 33.2% in women. BMI demonstrated greater discriminatory performance for MetS than WBF% in both datasets, and trunk fat percentage outperformed WBF% in DXA dataset. These findings support the hypothesis that Korean adults experience substantial metabolic risk at lower adiposity levels than reported in U.S. populations and highlight the importance of considering both total and regional adiposity in nutritional and metabolic risk assessment.

4. Vitamin D concentrations and neutrophil-to-lymphocyte ratio are associated with disability and pain sensitivity in individuals with migraine: a cross-sectional study.
PMID:日期:2026-08-29This secondary analysis aimed to examine the associations between biochemical biomarkers, lipid profiles and clinical outcomes in individuals with migraine. A cross-sectional study including 72 adults with a confirmed migraine diagnosis according to the ICHD-III criteria was conducted. Clinical outcomes comprised pressure pain thresholds (PPTs) at trigeminal and extratrigeminal sites, migraine-related disability (MIDAS), perceived stress (PSS-14), and handgrip strength. Biochemical parameters included serum vitamin D concentrations, the (Neutrophil-to-Lymphocyte Ratio (NLR), lipid profile and fasting glucose. Independent associations were assessed using adjusted multiple linear regression models. Higher NLR was associated with greater migraine-related disability (MIDAS; r = 0.38, p < .05) and higher perceived stress (PSS-14; r = 0.43, p < .05). Vitamin D concentrations were significantly and positively correlated with higher PPTs, particularly at the anterior temporalis (r = 0.36, p < .05), middle temporalis (r = 0.24, p < .05), and sternocleidomastoid muscles (r = 0.28, p < .05). Regression models further showed that elevated NLR was linked to increased disability (B = -0.35, 95% CI -0.47 to -0.17, p = .001) and lower PPTs at masseter (B = -0.40, 95% CI -0.75 to -0.14, p = .01) and middle temporalis (B = -0.34, 95% CI -0.46 to -0.13, p = .009). Also, lower vitamin D concentrations were associated with decreased PPTs at the middle temporalis (B = -0.38, 95% CI -0.52 to -0.22, p = .001). Higher NLR, as a marker of systemic inflammation, was associated with greater migraine-related disability and increased pain sensitivity, whereas lower vitamin D concentrations were associated with increased pain sensitivity.

5. Food-derived bioactive peptides with neuroprotective properties: a narrative review.
PMID:日期:2026-08-24Neuroprotective peptides, a class of bioactive peptides naturally derived from food sources, exhibit strong potential in neuroprotection and the promotion of neural development. These peptides are characterized by high bioavailability, favorable safety profiles, and multifunctional physiological benefits. Notably, specific peptides such as PW5 (Pro-Trp-Trp-Trp-Trp-Trp) and WHP (Trp-His-Pro) have demonstrated significant efficacy in reducing oxidative stress, inhibiting neuroinflammation, and attenuating neuronal apoptosis in both in vitro and in vivo models of cognitive impairment. These actions are comparable to neurotrophic factors such as brain-derived neurotrophic factor (BDNF) and glial cell line-derived neurotrophic factor (GDNF), contributing to improved synaptic plasticity and enhanced cognitive function. Clinically, these peptides offer promising adjunct or alternative strategies for managing neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD), especially in early-stage interventions or dietary-based therapies. This review summarizes recent advances in food-derived neuroprotective peptides, with particular emphasis on their neuroprotective mechanisms, bioavailability challenges, BBB transport, and delivery approaches. It further highlights current research gaps and provides perspectives for future investigations aimed at facilitating the rational development and potential application of these peptides. Current research faces key limitations, including the incomplete understanding of structure-activity relationships, insufficient evidence regarding blood-brain barrier (BBB) permeability, limited knowledge of peptide stability in complex biological environments, and the lack of comprehensive long-term safety evaluations. Future studies should prioritize systematic peptide screening, mechanism-oriented structure optimization, and advanced delivery strategies to improve the bioavailability and functional efficacy of food-derived neuroprotective peptides.

6. Paternal DHA supplementation before conception shapes offspring lipid metabolism in a sex- and postnatal diet-dependent manner.
PMID:日期:2026-08-15Paternal obesity results in altered metabolic health and cognitive dysfunction in the offspring. Docosahexaenoic acid (DHA) supplementation has been reported to improve metabolic inflammation, insulin sensitivity, and sperm quality. This study investigated whether paternal DHA supplementation in healthy male rats could prevent metabolic dysfunction induced by a high-calorie diet in their offspring, with a special focus on liver lipid metabolism and sex-specific outcomes. Male Wistar rats received daily saline or DHA-rich fish oil by oral gavage from weaning to adulthood before mating with untreated females. Offspring were then fed either a standard diet or exposed to a free-choice high-fat, high-sugar (FcHFHS) diet. Supplementation with DHA did not alter body weight, circulating lipid profiles, or glucose concentration in the founder males. In offspring, FcHFHS diet feeding induced increased body weight and adiposity in both sexes regardless of paternal DHA supplementation. However, female offspring exhibited greater susceptibility than males to FcHFHS diet-induced dyslipidemia and liver triglyceride accumulation, together with altered hepatic expression of genes involved in lipogenesis and lipid export. Three-way ANOVA analyses revealed that sex and postnatal diet were the primary determinants of these gene expression changes, as well as strong interaction effects between sex and paternal DHA supplementation for key lipogenic genes, including GPAM, SCD1, and FAS. These results show that paternal DHA supplementation did not protect offspring from obesity or metabolic dysfunction. Instead, it induced sex- and diet-dependent programming effects of lipid metabolism driven by strong interactive influences of paternal diet, offspring sex, and postnatal nutritional environment.

7. Immunomodulatory effects of palm-derived tocotrienol-rich fraction in healthy volunteers following influenza vaccination: A randomised clinical trial.
PMID:日期:2026-08-11Most studies on vitamin E and immunity have focused on tocopherols, but the tocotrienol-rich fraction (TRF) from palm oil may exert stronger immunomodulatory effects. This study investigated the effect of various TRF doses on the immune response to influenza immunisation in healthy adults. We hypothesized daily supplementation with lower doses of TRF can boost immune response to the influenza vaccine in healthy Individuals. In a double-blind placebo-controlled trial, 150 participants (≥18 years) were randomised to receive placebo or TRF (50, 100, 200, or 400 mg) daily for 2 months, with vaccination on day 28. Blood samples collected on days 0, 28, and 56 were analyzed for humoral and cellular immunity, antioxidant status, and plasma concentrations of vitamin E. TRF significantly augmented anti-influenza antibody production across all doses, with the 400 mg group exhibiting the highest titers. A selective Th1-type immune response was observed, characterized by significant IFN-γ production upon restimulation in all TRF-supplemented groups compared with placebo. Plasma IL-12 was elevated in the highest dose, whereas the amount of EBI3 produced increased across a broader range of doses compared to placebo. TRF supplementation was associated with increased antioxidant enzyme activities and stable malondialdehyde activity. Elevated α-tocopherol concentrations observed, primarily at higher doses. Supplementation with TRF can enhance immune response to influenza immunisation by modulating humoral, Th1 responses and improving antioxidant status. Critically, antibody production increased similarly across all TRF doses, suggesting that lower doses may offer a cost-effective strategy for supporting humoral immunity, while higher doses provide broader immunomodulatory benefits.

8. Oxidative stress and iron dysmetabolism: key targets and novel evidence for dietary intervention in MAFLD.
PMID:日期:2026-08-08Metabolic dysfunction-associated fatty liver disease (MAFLD) is a critical global public health challenge with limited treatment options. Recent studies have identified the "oxidative stress-iron dysmetabolism axis" as a key pathological hub driving its onset and progression. This review systematically elaborates on the core mechanisms by which this interactive axis propels MAFLD, primarily involving the mutual exacerbation of reactive oxygen species (ROS) burst and iron overload, the activation of ferroptosis pathways, and the consequent inflammatory signaling, which collectively disrupt hepatocyte homeostasis and exacerbate injury. Based on this framework, the article summarizes novel dietary intervention strategies targeting this axis, such as specific dietary patterns, various dietary bioactive compounds, nutrients, and probiotics, which aim to break the vicious cycle and improve liver pathology through multiple pathways. Future research needs to focus on in-depth mechanistic dissection, developing personalized precision interventions, and strengthening clinical translation to advance these dietary strategies as an integral component of the comprehensive MAFLD prevention and management system.

9. Serum 25(OH)D concentrations are differentially associated with innate and adaptive immune cell markers, with threshold effects near 40 ng/mL: A cross-sectional study.
PMID:日期:2026-08-05The nonlinear relationship and threshold effects of vitamin D on immune function in healthy adults remain unclear. This cross-sectional study of 316 healthy adults (January 2022 to June 2023) investigated associations between serum 25-hydroxyvitamin D (25(OH)D) concentrations and peripheral blood immune parameters (CD3+ T lymphocytes, CD4+ T cells, CD19+ B cells, and natural killer [NK] cells) quantified by flow cytometry. Restricted cubic spline and piecewise regression models evaluated nonlinear relationships and inflection points. Compared to the low 25(OH)D group (<28.45 ng/mL), the high group (>28.45 ng/mL) had higher NK cell counts (306.00 vs 272.50, P < .01) and percentages (15.52% vs 14.21%, P < .01), but lower CD3+ T cell percentage (69.99% vs 73.20%, P < .01). Multivariate regression confirmed that 25(OH)D was negatively associated with CD3+ T cell percentage (β = -0.16, P < .001), and positively with NK cell count (β = 3.62, P = .002) and NK cell percentage (β = 0.16, P < .001). Restricted cubic spline analyses revealed significant threshold effects, with inflection points at 40.94 ng/mL for CD3+ T cell percentage (P = .003), 37.86 ng/mL for NK cell count (P = .005), and 40.94 ng/mL for NK cell percentage (P = .022). Subgroup analyses showed stronger correlations in males and individuals 40 to 59 years of age (P < .05). Our findings suggest that vitamin D may exert threshold-dependent dual immunomodulatory effects-enhancing innate immunity (NK cells) while suppressing adaptive immune activation (CD3+ T cells). Higher 25(OH)D concentrations (around 40 ng/mL) were associated with distinct immune profiles in this healthy cohort. These associations may provide preliminary insights for future studies, although causal relationships cannot be inferred from this cross-sectional design.

10. Source-specific dietary nitrate and nitrite intakes are differentially associated with the incidence of hypertension: Tehran Lipid and Glucose Study.
PMID:日期:2026-07-30In the current study, we have investigated the association between dietary nitrate (NO)/nitrite (NO) intakes and the incidence of hypertension (HTN). This study included a total of 1576 men and women (mean age of 36.4 ± 11.7 and 39% men) in the third (2006-2008) phase of the Tehran Lipid and Glucose Study and were followed up until 2018-2022. At the beginning of the study, a semiquantitative food frequency questionnaire was applied to measure dietary intakes of NO and NO. Multivariable-adjusted Cox proportional hazard models for multiple variables were used to calculate hazard ratios (HRs) and 95% confidence intervals (CIs) of incidence of HTN across different quartiles of NO and NO dietary intakes. Median (interquartile range) daily intake of NO and NO were 465 (253) mg/d and 9.6 (4.8) mg/d, which were mostly derived from plant-based sources (95.4% and 55.8%, respectively). Participants in the highest quartile of total and plant-based NO₃ intake were associated with a significantly lower risk of HTN (HR= 0.46; 95% CI, 0.23-0.91; HR=0.44; 95% CI, 0.22-0.87, in total and plant-based sources, respectively). No significant association was found between animal-based NO₃ intake and HTN incidence. The highest quartile of plant-based NO₂ intake, compared to the lowest, was associated with a lower risk of HTN (HR = 0.57; 95% CI, 0.31-0.99) and there was no significant association between animal-based NO₂ intake and the risk of HTN. Higher intakes of total and plant-based NO and NO, compared with lower intakes, were significantly associated with a lower risk of HTN.
