1. Effects of Different Treatment on the Chemical Composition, Ruminal Degradation Kinetics and In Vitro Digestibility of Cottonseed.
PMID:This study evaluated the effects of gamma ray (GR), electron-beam (EB) irradiation, microwave irradiation (MI) and roasting (R) on the chemical composition, ruminal degradability and in vitro digestibility of whole cottonseed (CS). Cottonseed samples were treated with gamma irradiation (20 and 40 kGy), electron beam (20 and 40 kGy), microwave irradiation (3 and 5 min), and roasting (at 140°C for 15 and 30 min). Chemical composition was analyzed by standard methods, whereas ruminal degradability of dry matter (DM) and crude protein (CP) was measured using the nylon bag technique with three fistulated cows. In vitro digestibility was determined with the Tilley and Terry method, and amino acid degradability was assessed across treatments. All processing methods decreased neutral detergent fiber (NDF) and acid detergent fiber (ADF) of CS compared to untreated CS (p < 0.05). Gamma and microwave irradiation decreased DM and CP degradability of CS at ruminal outflow rates of 0.02, 0.05 and 0.08 h (p < 0.05). All treatments decreased effective rumen degradability (ERD) of CS at ruminal out flow rates of 0.02, 0.05 and 0.08 h (p < 0.05). Roasting and microwave irradiation for 5 min decreased water soluble fraction and increased the slowly degradable fraction of CS compared to untreated CS (p < 0.05). In vitro digestibility of 40 kGy irradiated CS and roasted CS for 30 min was significantly higher than other treatments and untreated CS (p < 0.05). Ruminal disappearances of essential and non-essential amino acids after 16 h incubation in rumen were decreased by electron beam irradiation (p < 0.05). These results indicate that physical processing methods, especially electron beam and roasting, enhance nutritional value of CS for ruminants.

2. Effects of Integrated Feeding of Tithonia diversifolia, Katuk Leaves and Palm Kernel Meal-Based Concentrate on Feed Intake, Digestibility, and Milk Production of Sapera Goats Lactation.
PMID:This study aimed to determine the effects of a mixture of tithonia (Tithonia diversifolia) forage, palm kernel meal-based concentrate, and katuk leaves (Sauropus androgynus) on nutrient digestibility, feed intake, milk production, and the composition of milk in first-lactation Sapera dairy goats. The experiment was conducted using a completely randomized design with four treatments: A (control; commercial forage and commercial concentrate), B (60% tithonia + 20% commercial concentrate + 20% palm kernel meal-based concentrate), C (B + 1.5% katuk leaves), and D (B + 3% katuk leaves). The addition of katuk leaves to the diet resulted in a significant (p < 0.05) increase in feed intake, protein digestibility, and milk yield by 18% compared with the control group. The fat, protein, and total milk solids content also increased by 9%, 12%, and 7%, respectively. The combination of tithonia and katuk leaves in this tropical feed system improved nutrient utilization efficiency and the quality of Sapera goat milk production in a sustainable manner.

3. Effects of the Switch From a Short-Day Photoperiod With Natural Light to a Long-Day Photoperiod With White Light-Emitting Diode or Induction Lighting on Milk Production and Endocrine Profiles in Lactating Dairy Cows.
PMID:This study evaluated the response of milk production and endocrine profile to a switch from short-day photoperiod (SDPP) with natural light to long-day photoperiod (LDPP) with white light-emitting diode (WLED) or induction lighting (IL) in lactating dairy cows. Nine Holstein cows (198 ± 10.0 days in milk) were initially exposed to SDPP of 10:14 h light-dark cycle (10L:14D) with natural light (SDPP period), followed by a long-day photoperiod (LDPP; 16L:8D) using either WLED (443 nm peak, LDPP-W, n = 5) or IL (IL; 529 nm peak, LDPP-I, n = 4). Each period comprised a 1-week adaptation and a 2-week sampling period. Milk yield decreased after switching to LDPP-W (p < 0.05) but remained unchanged with LDPP-I. The slope of milk yield over the entire trial period (from the beginning of SDPP to the end of LDPP period) differed depending on lighting type (p = 0.05), being negative in the LDPP-W and positive in the LDPP-I. Plasma prolactin concentration in the light period increased after switching to LDPP-I but not to LDPP-W. These results indicate that LDPP using IL is likely more effective in promoting milk yield than WLED, potentially due to enhanced prolactin secretion.

4. Lipidomic and Metabolomic Profiling of Breast Muscle Between Squabs and Adult White King Pigeons.
PMID:This study systematically investigated age-related variations in meat quality traits, lipidomic profiles, and metabolomic profiles of breast muscle from White King pigeons, comparing 28-day-old squabs (D28) and 12-month-old adult pigeons (M12). Six biological replicates were used per group (n = 6). Physicochemical analyses and ultra-high performance liquid chromatography-tandem mass spectrometry were employed to characterize these differences. Results showed significant disparities in meat quality between the two age groups: D28 pigeons exhibited significantly higher lightness (L*), redness (a*), and yellowness (b*) values at 24-h postslaughter, along with higher water-loss rate alongside lower shear force (p < 0.05), reflecting more tender muscle texture despite weaker water retention capacity. In contrast, M12 pigeons had higher springiness but lower cohesiveness and gumminess (p < 0.05). Lipidomic analysis identified 553 lipid molecules, with 174 differential lipids categorized into six classes. D28 pigeons had significantly higher levels of glycerolipids (GLs) and fatty acyls (FAs), while M12 pigeons showed elevated saccharolipids (SLs) and prenol lipids (PRs). Metabolomic profiling detected 19,019 metabolites, among which 984 were differentially abundant (screening criteria: variable importance in projection (VIP) ≥ 1, fold change (FC) > 1.2 or FC < 0.83, FDR-adjusted p < 0.05), predominantly enriched in amino acid metabolism. Correlation analysis revealed that acylcarnitines (AcCa) and phosphatidylinositol (PI) were positively correlated with water-loss rate and L24 values (r > 0.8, p < 0.05), whereas lysophosphatidylethanolamines (LPEs), L-threonic acid, and β-glycerophosphoric acid showed positive associations with shear force and cooking loss (r > 0.8, p < 0.05). These findings reveal statistical associations between lipid/metabolite profiles and age-driven differences in pigeon breast meat quality, and identify potential candidate lipid and metabolite biomarkers correlated with pigeon meat quality phenotypes. These results identify candidate biomarkers associated with breast meat quality in male White King pigeons at two ages and offer preliminary exploratory omics data for subsequent pigeon meat research, rather than universal guidance for breeding and production.

6. Meta-Analysis of the Mitigation Effects of Supplementation With Calcium Salts of Unsaturated Fatty Acids on Enteric Methane Emissions in Beef Cattle and Its Impact on Meat Quality.
PMID:This study evaluated the effects of calcium salts of unsaturated fatty acids (CSUFAs) on enteric methane (CH) emissions and meat quality in beef cattle by integrating data from trials on beef cattle of different production stages, sexes, and breeds. Specifically, a meta-analysis of four studies including 44 animals (22 with CSUFAs and 22 as a control) was performed to assess CH-related outcomes. Daily CH emissions in all experiments were estimated by multiplying the breath CH:CO concentration ratio by estimated CO emissions. CSUFA supplementation tended to reduce CH emissions per unit of total digestible nutrient intake (CH/TDNI; mean difference = -2.2 L/kg, p = 0.077), corresponding to a reduction of 5.6%. Meanwhile, no significant differences were observed for the CH:CO ratio, daily CH emissions, or CH per unit of dry matter intake. Physicochemical analysis of meat from 16 animals revealed no adverse effects of CSUFA supplementation on meat quality. Descriptive sensory evaluation indicated improved juiciness, fat flavor richness, and other sensory attributes upon CSUFA supplementation (p < 0.05). These findings suggest that CSUFAs can reduce enteric CH emissions relative to dietary energy density while maintaining meat quality, supporting their use as a practical strategy for mitigating CH emissions from beef cattle.

7. A Technique for Nonmicrobial Freshness Evaluation of Broiler Breast Meat Using a Handheld Electrical Impedance Meter.
PMID:This study investigated the applicability of a handheld electrical impedance meter (EIM) for nondestructive evaluation of the freshness of broiler breast meat during refrigerated storage. Impedance ratios (2/100 kHz) were measured on Days 0, 1, 2, 3, 5, 7, and 10 of refrigerated storage and were compared with the K-values obtained through high-performance liquid chromatography (HPLC) analysis of adenosine triphosphate (ATP) degradation products. The impedance ratio decreased consistently over time, with SD decreasing from 0.36 on Day 0 to 0.07 on Day 10. A quadratic regression model showed that the impedance ratio decreased with the number of storage days (R = 0.7561). The K-value increased over time and was also well described by a quadratic regression model (R = 0.8395). The impedance ratio was strongly negatively correlated with the K-value (r = -0.730; n = 140; p < 0.001), supporting its utility for tracking freshness-related changes. Although the K-value demonstrated a slightly higher explanatory power, the impedance ratio showed sufficient accuracy for practical applications. These findings suggest that the EIM method offers a rapid, simple, and nondestructive alternative to conventional biochemical assays, with potential for implementation in poultry processing and retail environments for on-site freshness assessment.

8. Evaluation of Fermentation Quality, Digestive Characteristics, and Nutritive Value of Whole Crop Silage Prepared by Fine-Cutting From Rice Cultivars With Short Panicles and Fed to Dairy Cows.
PMID:Silage fermentation and digestibility characteristics, ruminal fermentation, and chewing activity in dairy cattle fed whole-crop rice silages having short panicle cultivars "Tachisuzuka," "Tachiayaka," and "Tsukisuzuka" were evaluated. All cultivars used in this study were grown under identical conditions, harvested using a fine-cutting harvesting machine, and ensiled in a round baler for the chopped material along with Lactobacillus inoculation. Four nonlactating, nonpregnant, rumen-cannulated Holstein cows were housed in a climate-controlled room and fed one of the three silage diets in a 3 × 3 Latin square design. Every 2 weeks, the cows were housed in respiration chambers for 5 days to measure gaseous exchange, energy and nitrogen balance, apparent total-tract digestibility of nutrients, and chewing activity. All cultivars showed fermentation quality with low pH and high acetic acid content. The digestibilities of fiber and nonfibrous carbohydrates were higher in Tachisuzuka and Tachiayaka than in the other cultivars, respectively. The total digestible nutrient contents in Tachisuzuka, Tachiayaka, and Tsukisuzuka were 61.4%, 60.8%, and 59.4%, respectively. The roughage value index was 79-86. The high digestibility of these silage cultivars suggests that they could be used as feed for lactating cows; however, their high chewing stimulus also needs to be considered.

9. Modified Zymography Protocol: A Rapid and Sensitive Method for Monitoring Calpain Activity.
PMID:Calpain is a key proteolytic enzyme involved in muscle development. Overactivation in diseases can lead to muscle atrophy, and its role in postmortem meat tenderization affects consumer acceptability. To develop a rapid and sensitive zymography protocol for the efficient detection of calpain activity in skeletal muscle cells, we addressed the limitations of traditional methods that require lengthy assay times and high protein inputs. A previously published zymography method was modified by increasing the Ca concentration from 4 to 8 mM and reducing the incubation time from overnight to 6 h. Calpain activity was assessed in differentiated C2C12 myotubes using varying protein concentrations (1-8 μg), and band intensity was quantified using ImageJ software. The optimized conditions enhanced the detection sensitivity up to tenfold and enabled reliable visualization of calpain activity using only 1 μg of protein within 8 h. The 8 mM Ca incubation buffer yielded stronger, clearer lytic bands compared with 4 mM, indicating improved calpain activation. This modified zymography protocol significantly reduces assay time and protein requirement while maintaining high sensitivity. The method serves as an efficient tool for studying calpain-mediated proteolysis in muscle atrophy research and postmortem meat quality evaluation.

10. Analysis of miRNA Expression in Bovine Endometrium During the Estrous Cycle.
PMID:The present study aimed to identify and characterize bovine endometrial miRNAs at different stages of the estrous cycle. Total RNA was extracted from endometrial tissue and subjected to small RNA sequencing. A total of 334 miRNAs were expressed in bovine endometrial tissue during the estrous cycle. Among them, 11 were differentially expressed miRNAs (DEMs) at the follicular stage (F-DEMs) and four at the luteal stage (L-DEMs) in comparison between F and L stages. Bioinformatic analysis of the target genes of these DEMs revealed that miR-2411-3p and miR-320a were the top-ranked hub miRNAs in F- and L-DEMs, respectively. These two miRNAs demonstrated the highest functional coverage in GO annotation. The analysis using cultured bovine endometrial cells showed that the expression level of miR-2411-3p was significantly high in the E2-treated group compared to the control (p < 0.05), whereas the expression level of miR-320a was significantly high in the P4-treated group (p < 0.05). The present study identified the endometrial DEMs during the estrous cycle and predicted their functions based on their target genes. The results demonstrated that miR-2411-3p and miR-320a were identified as characteristic endometrial miRNAs regulated by ovarian steroid hormones in bovine.
