2. Rapid Detection of Component Contents in Polyphenol-Rich Plant Leaves Based on Near-Infrared Spectroscopy.
PMID:日期:2026-09-30Leaves of Rhus punjabensis var. sinica, Rhus potaninii, Caesalpinia spinosa (Tara), and Phyllanthus emblica (Indian gooseberry), and other species are rich in tannic and gallic acids, which are polyphenols widely utilized in the pharmaceutical, food, and chemical industries. Conventional quantification involves tedious solvent extraction and chemical assays, which are time-consuming, costly, and incapable of simultaneous multi-component analysis. To enable rapid, non-destructive detection, Fourier transform near-infrared spectroscopy was used to establish quantitative models for synchronous determination of tannic acid, gallic acid, and moisture in polyphenol-rich plant leaves. Eighty-five leaf samples from six species were collected; reference values were obtained by chemical methods. Near-infrared spectra of plant leaves were fitted to traditional chemical values. Synergy interval partial least squares (SIPLS) selected characteristic intervals to build a partial least squares (PLS) model; performance was assessed by correlation and error metrics. Optimal pretreatments were FD+MSC (moisture), MSC (tannic acid), and FD (gallic acid) with number of principal factors 4, 9, and 6. Characteristic bands: moisture 9403.7-6098.1 and 5450.1-4597.7 cm⁻¹; tannic acid 9403.7-7498.2 and 6102-4246.7 cm⁻¹; gallic acid 9403.7-8451 and 6102-5446.3 cm⁻¹. For moisture, tannic acid, and gallic acid, Rc² = 0.9574, 0.9595, 0.9115; RMSECV = 0.433%, 1.47%, 0.362%; RPD = 4.81, 4.96, 3.36; Rp² = 0.9669, 0.9792, 0.9443; RMSEP = 0.274%, 1.25%, 0.241%. Bias was not significant (P > 0.05), and average RSD < 2%. The results demonstrate that near-infrared spectroscopy combined with chemometric methods demonstrates high predictive capability and practical application value for the quantitative determination of tannic acid, gallic acid, and moisture in polyphenol-rich plant leaves. A rapid, non-destructive NIR spectroscopy method was developed for simultaneous quantification of tannic acid, gallic acid, and moisture in polyphenol-rich plant leaves. Optimized chemometric modeling (SIPLS-PLSR) achieved high prediction accuracy (Rp² > 0.94, RPD > 3.0), demonstrating strong robustness and suitability for practical applications.

3. Determination of cadmium, lead and mercury in cannabis flowers by microwave plasma atomic emission spectrometry.
PMID:日期:2026-09-24The rapid expansion of legal medicinal and recreational cannabis markets has increased the need for quality control strategies and regulatory monitoring of contaminants in cannabis products. Toxic elements are of particular concern because cannabis plants can accumulate trace metals from environmental and cultivation sources. To evaluate microwave plasma atomic emission spectrometry (MP AES) as a cost-effective alternative to conventional argon plasma-based techniques for the determination of cadmium (Cd), lead (Pb), and mercury (Hg) in cannabis flowers. Samples were prepared by microwave-assisted acid digestion using nitric acid. Cd and Pb were determined sequentially by MP AES, while Hg was quantified using a dedicated cold vapor generation coupled to MP AES (CV-MP AES). The analytical methodologies were optimized and validated according to Eurachem guidelines. Limits of quantification were 0.40 µg/g for Cd, 0.18 µg/g for Pb, and 0.015 µg/g for Hg. Accuracy was evaluated using reference materials and spiked samples, with recoveries of 95-101% and 81-90%, respectively, and relative standard deviations below 15%. Good linearity was achieved (R² > 0.999). The methodologies were applied to 30 commercial cannabis flower samples, where Cd and Hg were not detected, and Pb was quantified at 2.12 ± 0.13 µg/g. MP AES and CV-MP AES provide adequate accuracy and precision for the determination of Cd, Pb, and Hg in cannabis flowers. The proposed methodologies represent a practical, cost-effective, and sustainable alternative to argon-based plasma techniques for routine quality control and compliance testing of cannabis products.

4. A Risk-Based Framework to Optimize Microbiological Testing of Primary Packaging Materials.
PMID:日期:2026-09-24Risk-based approaches are increasingly encouraged to optimize microbiological control strategies for non-sterile pharmaceutical materials. Primary packaging materials represent a critical interface, yet routine microbiological testing is often applied uniformly without considering material characteristics or historical performance. This study aimed to develop and evaluate a risk-based framework to support reducing routine microbiological testing for non-sterile primary packaging materials by integrating water activity (Aw), historical microbiological data, and intrinsic material and process attributes. Common primary packaging materials, including PVC, PVC-PVDC, aluminum foil, PET, HDPE, LDPE, and polymeric tubes, were evaluated. Water activity was measured and compared against an upper specification limit (Aw ≤ 0.60). Historical microbiological performance was assessed using at least 20 consecutive compliant batches without out-of-specification (OOS) or out-of-trend (OOT) results. A one-sided process capability analysis (Cpk) was performed to characterize Aw variability relative to the specification limit. Historical data supported sustained control across materials, with at least 20 consecutive compliant batches and Aw values consistently ≤ 0.60. Most materials showed low variability and operated with a substantial margin relative to the Aw limit, whereas HDPE and LDPE exhibited comparatively higher variability. These findings were not associated with evidence of microbiological proliferation within the evaluated dataset. Cpk values quantitatively described observed performance and were interpreted as supportive indicators given the limited dataset. These findings support a risk-based strategy to optimize the frequency of microbiological testing. Historical microbiological performance represents the primary basis for reduced testing, while Aw and material attributes provide complementary evidence. This approach enables optimization of testing under controlled conditions without replacing microbiological control, in alignment with ICH Q9(R1), ICH Q6A, and relevant compendial guidance. A risk-based framework that integrates water activity, historical microbiological data, and material attributes is proposed to optimize microbiological testing of non-sterile primary packaging materials. All evaluated materials showed Aw ≤ 0.60, consistent with conditions unfavorable to microbial proliferation, while historical data (≥20 compliant batches) provided the primary evidence of sustained control. Differences in Aw variability were observed, particularly for HDPE and LDPE, highlighting the importance of material-specific characteristics in risk assessment. Overall, the approach supports the reduction-rather than replacement-of microbiological testing under controlled conditions, in alignment with current regulatory expectations.

5. Edible Fish as a Source of Klebsiella pneumoniae: Incidence, Virulence, Biofilm Formation, Antimicrobial Resistance and Control Strategies.
PMID:日期:2026-09-21Fish sold in retail markets are prone to contamination with pathogenic bacteria. Investigations on occurrence of Klebsiella pneumoniae, its characterization in seafood are essential for the protection of consumer health. The present study investigated the incidence of K. pneumoniae in fish and characterized their virulence profile, biofilm-forming ability, antimicrobial resistance and their control using water-soluble chitosan and postbiotics. In total, 144 fish and processing water samples from fish retail markets of Visakhapatnam, India were screened for presence of K. pneumoniae using MacConkey agar and amplifying khe gene. Virulence characterization (entB, mrkD, kfu, allS, rmpA, ybtS, iutA) antibiotic susceptibility test (disc diffusion assay against 28 antibiotics) biofilm-forming ability and anti-K. pneumoniae activity of water-soluble chitosan (microtiter plate assay) were studied. Eleven fish samples showed presence of K. pneumoniae. All the isolates possessed entB gene, however, 91% of the isolates carried mrkD and kfu genes. Antibiogram studies showed that all isolates were multidrug resistant (MDR), and their Multiple Antibiotic Resistance (MAR) index ranged from 0.19 to 0.39. Resistance genes detected in K. pneumoniae were bla SHV (91%), bla TEM (27%), sul1(18%), tetA (9%), tetD (9%), and catA2 (9%). Majority of K. pneumoniae isolates were moderate biofilm formers (63.6%) and the biofilm-forming ability was potentiated by supplementation with lactose (100%) followed by fructose (90.9%) and ribose (90.9%) sugars. Water-soluble chitosan (1%) showed effective control of all K. pneumoniae. The present study showed majority of the K. pneumoniae isolates from seafood were found to be virulent, multidrug-resistant, ESBL producers, and moderate biofilm formers indicating potential health risk to fish consumers. Water-soluble chitosan showed a significant inhibitory effect on growth of all K. pneumoniae isolates. The detection of virulent and MDR K. pneumoniae in edible fish meat, albeit at a low level, poses a significant threat to consumers.

6. Evaluating Substitution of Hazardous Solvents in USP Monograph HPLC Methods.
PMID:日期:2026-09-10Hazardous solvents, such as dichloromethane (DCM), n-hexane, and acetonitrile (ACN), are widely used in HPLC methods, posing significant health and environmental risks. To evaluate the feasibility and impact of substituting hazardous solvents with greener alternatives in USP monograph methods. Six high-impact solvents were identified from USP-NF monographs. Two representative monographs per solvent were selected. Substitution strategies were assessed, and performance was compared using system suitability and sample acceptance criteria. Greenness improvement was evaluated using the Analytical GREEnness (AGREE) metric. Performance remained equivalent across all twelve substitution cases. In almost every instance, only the mobile phase required modification, either by direct substitution or by adjusting the solvent-to-buffer ratio, except for one case that required a minor adjustment in column temperature. The AGREE Greenness metric increased by 18-65% in ten out of twelve cases; for n-hexane, improvements were modest at just 6% when replaced with n-heptane but exceeded 40% when substituted with supercritical CO₂. Greener solvents are highly likely to replace hazardous solvents used in compendial chromatography methods without loss of performance. Demonstrated performance equivalency for greener solvent substitutions; Quantified greenness improvements using AGREE; Discussed strategies to implement greener solvents in USP monograph methods.

7. Validation of a QuEChERS-UHPLC-MS/MS method for the determination of aflatoxins and ochratoxin A in cannabis flowers: Application to regulated and non-regulated products.
PMID:日期:2026-09-03Cannabis sativa flowers are a complex plant matrix that poses significant analytical challenges due to their chemical composition and the low regulatory limits established for consumer safety. To optimize and validate a QuEChERS-UHPLC-MS/MS method for the simultaneous determination of aflatoxins B1, B2, G1, G2, and ochratoxin A in Cannabis sativa flowers. Critical parameters influencing extraction efficiency were systematically evaluated through a 3³ factorial design, including solvent acidification, extraction time, and evaporation temperature. Optimal conditions were achieved using acetonitrile acidified with 0.1% v/v formic acid, 15 min vortex extraction, and evaporation below 50 °C. Under these conditions, recoveries ranged between 90-109%, with precision (RSD 2.5-8.3%) and trueness demonstrating satisfactory method performance. Linearity was confirmed with R² > 0.99, and LOQs of 1.0 µg/kg for aflatoxins and 5.0 µg/kg for ochratoxin A, consistent with European Pharmacopoeia requirements. Application to 30 cannabis flower samples revealed that all commercial products complied with international maximum residue limits (MRLs), whereas some non-regulated samples exceeded the total aflatoxin limit (4 µg/kg), mainly due to elevated aflatoxin B2. These findings provide the first data on mycotoxin occurrence in cannabis flowers from Uruguay and highlight the importance of monitoring non-regulated products, emphasizing the need for appropriate quality-control strategies. The proposed method provides a simple, robust, and high-throughput approach for routine quality control of cannabis flowers.

8. A Generic Approach for Simultaneous Screening and Quantification of Aflatoxins and Pesticide Residues in Rice by LC-MS/MS: Single Laboratory Validation.
PMID:日期:2026-09-01Chemical residues in rice require highly sensitive analytical methods to verify compliance with stringent residue limits. In addition to pesticide residues, aflatoxins are critical contaminants due to their toxicity and carcinogenicity. To develop and validate a simple, generic LC-MS/MS workflow for simultaneous screening and quantification of pesticide residues and aflatoxins in rice. Samples were extracted using a modified QuEChERS EN 15662:2018 procedure with acidified acetonitrile, followed by dispersive solid-phase extraction cleanup. Analysis was performed using a UHPLC coupled to a triple quadrupole mass spectrometer (QQQ) with heated electrospray ionization in time-scheduled selected reaction monitoring mode. Method performance was assessed in accordance with SANTE guidance using matrix-matched calibration. The optimized LC-MS/MS method demonstrated many transitions with polarity switching while maintaining acceptable chromatographic peak quality and selectivity. Acidified extraction improved the response by 3- 5-fold for the selected analytes. The method achieved LOQs of 0.0005 mg/kg for pesticides and aflatoxins. Calibration was linear over 0.0005-0.050 mg/kg for pesticides and 0.0002-0.010 mg/kg (0.2 -10 µg/kg) for aflatoxins, with R² ≥ 0.99 and residuals within ±20%. Recoveries for all target analytes were within 70- 120%, with precision ≤20% RSD at the evaluated spike levels. The proposed single-extraction LC-MS/MS workflow is selective, sensitive, accurate, and suitable for the simultaneous high-throughput determination of pesticide residues and aflatoxins in rice. The method meets SANTE criteria for pesticides, complies with the Commission Implementing Regulation (EU) 2023/2782 for aflatoxins, and complies with FSSAI and EU MRLs/MLs for rice.

9. Triplex PCR Assays for Simultaneous Detection of Maize Ingredients, Maize Allergen-Encoding Genes, and the Cry1Ab Transgene in Processed Foods.
PMID:日期:2026-08-26Maize is widely used in processed foods but may cause allergic reactions. Reliable detection of maize ingredients, related allergens, and genetically modified organisms (GMOs) in foods is essential for food authenticity, food safety, and consumer protection. However, DNA degradation during food processing and multi-target detection remain significant analytical challenges. To develop and evaluate triplex PCR assays for the simultaneous detection of maize ingredients, allergen-encoding genes, and the Cry1Ab transgene, thereby enabling integrated food authentication, allergen and GMO screening in processed foods. Genomic DNA was extracted from certified reference materials (0-5% MON810 maize) and processed foods using the CTAB method and a commercial plant kit. Two triplex PCR systems were optimized for the detection of species-specific marker (zein), allergen-encoding genes (Zea m 8, Zea m 14), and the Cry1Ab transgene. The PCR products were analyzed via agarose gel electrophoresis. The CTAB method yielded higher amounts of amplifiable DNA than the commercial kit. The developed assays enabled simultaneous detection of (i) Zea m 8, Zea m 14, and the zein gene, and (ii) Zea m 8, Zea m 14, and the Cry1Ab transgene. The assays successfully detected 0.1% MON810 reference material in the uniplex and 0.5% in the triplex systems. Analysis of processed foods confirmed reliable detection of all targets despite DNA degradation. The proposed triplex PCR assays offer a reliable, rapid, and cost-effective approach for the simultaneous detection of maize ingredients, allergen-encoding genes, and the Cry1Ab transgene, supporting food authentication, allergen and GMO screening in processed foods. Triplex PCR assays enable rapid, simultaneous detection of maize ingredients, allergen-encoding genes (Zea m 8, Zea m 14, zein) and the Cry1Ab transgene in processed foods, detecting 0.1% and 0.5% MON810 reference materials by uniplex and triplex PCR, respectively.

10. Quality evaluation of Sanfu Paste by combining multi-dimensional fingerprint with chemometrics.
PMID:日期:2026-08-21The investigation of Sanfu paste (SFP) has mostly concentrated on its clinical applications, mechanistic study, and the enhancement of dosage forms; conversely, quality evaluation has been comparatively overlooked and hardly documented. This study seeks to develop a thorough technique for SFP quality evaluation by the integration of multidimensional fingerprinting, chemometrics, and multicomponent content analysis. The high-performance liquid chromatography (HPLC) and gas chromatography (GC) fingerprinting techniques were established to evaluate compositional similarity among 16 batches of SFP from various geographical sources. Ten primary bioactive constituents-sinapine thiocyanate, corydaline, coptisine, berberine hydrochloride, dehydrocorydaline, euphol, methyl eugenol, safrole, elemicin, and sinapic acid-were quantitatively assessed. Thorough quality evaluation utilised orthogonal partial least squares-discriminant analysis (OPLS-DA), principal component analysis (PCA), and hierarchical cluster analysis (HCA). Ten distinct chemical compounds were discovered, demonstrating batch-to-batch diversity in concentration; notably, sinapine thiocyanate and methyl eugenol exhibited comparatively larger abundances. The similarity indices between HPLC and GC fingerprints for 16 SFP batches surpassed 0.9. HCA and PCA produced consistent clustering patterns. The OPLS-DA model exhibited stability and robustness, identifying euphol, dehydrocorydaline, sinapine thiocyanate, and methyl eugenol as the primary discriminatory markers. This study developed a chemical fingerprinting and quantitative content-determination approach that offers an accurate and dependable instrument for the quality evaluation and thorough assessment of SFP. GC and HPLC fingerprinting techniques, along with quantitative content-determination assays for SFP, were established for the inaugural time. These approaches are essential for clarifying the pharmacological foundation of its bioactive components and for thorough quality evaluation.
