1. A kinesin motor complex localizes microtubule-severing proteins to direct gravitropism in Physcomitrium patens.
1. 一种驱动蛋白马达复合物定位微管切割蛋白以引导立碗藓的向重力性PMID:日期:2026-10-05Gravitropism enables plants to adjust growth direction and architecture in response to gravity. The classical Cholodny-Went theory posits that asymmetric auxin redistribution drives differential growth during gravitropism in seed plants, but the nature of the equivalent asymmetric process in moss protonemata is still unclear. Here, through forward genetic screening, we identified GTRD, which encodes a coiled-coil protein whose loss reverses the direction of protonemal gravitropism. We further demonstrated that GTRD forms a complex with the kinesin-14 motor GTRC, and that GTRC drives the minus-end-directed movement of GTRD along microtubules. This GTRC-GTRD complex transports a microtubule anchoring module MSD1-WDR8 that positions katanin. Under gravistimulation, katanin-dependent microtubule severing facilitates the asymmetric formation of a microtubule focus on the upper flank of the tip cell, where it spatially constrains the apical actin cluster. This cytoskeletal reorganization guides the upward growth of the protonemata. Together, the GTRC-GTRD motor complex spatially organizes katanin-mediated microtubule remodeling to establish cytoskeletal polarity, which serves as a key asymmetric cue that directs gravitropic growth in protonemal tip cells.

2. Nuclear localization controls BED-NLR Xa1-mediated immunity in rice.
2. 核定位控制水稻中BED-NLR Xa1介导的免疫PMID:日期:2026-10-03Transcription activator-like effectors (TALEs) are major virulence factors of Xanthomonas oryzae, the causal agent of bacterial blight in rice. The BED-domain nucleotide-binding leucine-rich repeat receptor Xa1 confers resistance by recognizing TALEs, but the mechanisms underlying Xa1 activation remain unclear. Here we show that structurally conserved canonical TALEs differ in their ability to activate Xa1, revealing unexpected specificity in effector recognition. Using transient expression assays, endogenous epitope tagging, and prime editing of the native Xa1 locus, we demonstrate that nuclear localization is essential for Xa1-mediated immunity. We identified three functional nuclear localization signals (NLSs) within the Xa1 N-terminal region. Prime-edited rice lines carrying mutations in these motifs exhibited compromised resistance, with mutation of the first NLS causing the strongest susceptibility phenotype. Disruption of the NLS motifs in either Xa1 or TALEs did not prevent their physical interaction, indicating that effector binding can occur independently of nuclear localization. However, only nuclear-localized Xa1 triggered cytosolic Ca2+ influx, a hallmark of immune activation. Together, our results establish nuclear localization as a prerequisite for Xa1 immune function and reveal a nuclear surveillance mechanism for BED-NLR-mediated immunity in plants.

4. Single-cell epigenomics in plants.
4. 植物单细胞表观基因组学PMID:日期:2026-09-23Single-cell technologies enable epigenomics characterization at single-cell resolution, offering novel insights into gene expression regulation and a wide range of biological processes, including cell fate determination, developmental differentiation, and environmental adaptation. In recent years, plant single-cell epigenomics has witnessed rapid progress, empowering the construction of cell type-specific chromatin accessibility atlases, the identification of candidate cis-regulatory elements (CREs), the inference of gene regulatory networks, and the elucidation of regulatory mechanisms underlying plant development, stress responses, species evolution, and complex agronomic traits. Nevertheless, single-cell methodologies for profiling histone modifications, DNA methylation, three-dimensional genome organization, and spatial epigenomic states in plants are still in their infancy. In this review, we systematically elaborate the technological framework, bioinformatic workflows, key research advances, and existing challenges in the field of plant single-cell epigenomics, and highlight future perspectives for its in-depth development.

8. The circadian clock protein CmLHY integrates strigolactone signaling to regulate short-day-induced flowering in chrysanthemum.
8. 昼夜节律钟蛋白CmLHY整合独脚金内酯信号以调控菊花短日照诱导的开花PMID:日期:2026-09-22Flowering is a key ornamental trait in chrysanthemum and a major agronomic trait affecting rice yield. Strigolactones (SLs) are a class of phytohormones that regulate diverse aspects of plant growth and development, including flowering time. However, the molecular mechanisms underlying SL-mediated regulation of flowering remain poorly understood. Here, we show that SL promotes flowering in chrysanthemum under inductive short-day conditions. The SL-specific signaling complex CmMAX2-CmD14.1-CmD53 was first identified in chrysanthemum. Silencing CmD14.1 or CmMAX2 causes late-flowering and SL insensitivity, while their overexpression accelerates flowering upon SL treatment. In contrast, overexpression of CmD53 or the degradation-resistant variant CmD53m delayed flowering. SL-induced CmD53 degradation depends on both CmD14.1 and CmMAX2. RNA-seq analysis reveals that overexpression of CmD53 or CmD53m significantly downregulates the florigen gene CmFTL3. We confirmed that CmD53 directly binds to the CmFTL3 promoter to repress its transcription, thereby delaying flowering. Moreover, the central circadian clock protein CmLHY represses CmMAX2 and CmD14.1 transcription and subsequently modulates SL-mediated CmD53 degradation. This regulatory cascade fine-tunes CmFTL3 expression and controls flowering in chrysanthemum under inductive SD conditions. Similarly, in rice, OsD3 (a MAX2 homolog) and OsD14 act downstream of OsCCA1 (an LHY/CCA1 homolog) to regulate flowering. Our findings demonstrate that LHY/CCA1-mediated integration of SL signaling in the flowering regulatory pathway is conserved between chrysanthemum and rice, providing new insights into the novel crosstalk between the circadian clock and SL signaling in flowering-time control.

9. Sorbitol dehydrogenase aids grain-fill and integrates carbohydrate metabolism with redox in the hypoxic maize endosperm.
9. 山梨醇脱氢酶有助于灌浆,并在缺氧玉米胚乳中将碳水化合物代谢与氧化还原整合PMID:日期:2026-09-21Extreme low oxygen is typical of microenvironments inside developing grains, thus requiring metabolic adaptations to hypoxia. Potential contributions from sorbitol dehydrogenase (SDH) are tested here for maize (Zea mays) since the reversible reaction (fructose + NADH ↔ sorbitol + NAD+) can balance both redox state and metabolism during assimilate import into kernels. To do so, we developed and analyzed maize sdh1 mutants and over-expression lines. Dysfunction of Sdh1 decreased seed weight by 17%, impaired starch accumulation, and enhanced levels of glucose, fructose, and sucrose. Redox balance in the inner endosperm was also disrupted (NADH and NADPH levels rose) and adenylate energy charge (AEC) decreased. Moreover, genes responsive to oxidative stress were upregulated in mutant kernels. In contrast, Sdh1-OE lines rescued the small-grain phenotype without exceeding wild-type kernel size. Metabolite profiles of mutant and Sdh1-OE kernels revealed reciprocal levels for sucrose, hexoses, and key points of redox sensitivity and C/N balance. Results are distinct from the classical involvement of sorbitol in long distance transport (as in apples) and its contribution to desiccation tolerance in diverse species. Rather, our analyses support a two-fold contribution by an SDH-modulated sorbitol reservoir that 1) aids redox balance in the hypoxic endosperm by storing excess reductant and 2) minimizes fructose levels that can otherwise inhibit sucrose import, metabolism, and signaling pathways. Collective evidence reveals a previously unrecognized role for sorbitol and SDH in kernel-fill that integrates carbohydrate metabolism and redox balance in the low-oxygen endosperm of this developing grain.

10. BR-ENHANCED EXPRESSION 1/2/3 fine-tune plant photomorphogenesis by regulating the phyA/B-SPA1 cascade.
10. BR增强表达1/2/3通过调控phyA/B-SPA1级联精细调节植物光形态建成PMID:日期:2026-09-21BR-ENHANCED EXPRESSION1 (BEE1), BEE2, and BEE3, key transcriptional components of brassinosteroid signaling, have been shown to play critical roles in regulating light signaling. However, the precise molecular mechanism by which BEE1/2/3 repress photomorphogenesis remains elusive. In this study, we found that light triggers the degradation of BEE1/2/3 proteins through the 26S proteasome system. BEE1/2/3 interact with phyA and phyB, and this interaction promotes the degradation of BEE3. Mutations in BEE1/2/3 exhibit short hypocotyls and partially suppress the long hypocotyl phenotype of the phyA phyB double mutant. BEE1/2/3 attenuate the interaction between phyB and PIF3, leading to PIF3 stabilization and subsequent repression of photomorphogenesis. In addition, BEE1/2/3 interact with SPAs to disrupt the phyA/B-SPA1 association, thereby further attenuating the exaggerated light signaling. The COP1-SPAs complexes stabilize BEE1/3 under both dark and light conditions. Mutations in BEE1/2/3 enhance the constitutively photomorphogenic phenptype of spa1/2/4, whereas overexpression of BEE1/3 partially rescue the short hypocotyls of cop1 and spa1/2/4. Collectively, these findings demonatrate that BEE1/2/3 fine-tune the phyA/B-COP1/SPA1 signaling hub to negatively regulate photomorphogenesis in response to excessive light signaling. Our study provides insights into the pivotal roles of BEE1/2/3 as photomorphogenic repressors, facilitating adaptation to complex light conditions.
