1. [Immunological methods for the diagnosis of bovine tuberculosis--state of the art].
PMID:日期:2000-10-01An overview is given about the most common immunological methods for the diagnosis of bovine tuberculosis. These methods are based on cell mediated immune reactions on the one hand (intradermal skin test, interferon-gamma-test, lymphocyte transformation test) or on antibody production against Mycobacterium bovis on the other hand (ELISA). The advantages and disadvantages of the methods as well as their specificity and sensitivity are discussed.

2. [Reliability of the interpretation of lymph node cytology by investigators with little cytologic experience--a study with students of veterinary medicine].
PMID:日期:2000-10-01The aim of this study was to calculate the reliability of the examination of lymph node cytologies by individuals with little experience in the field of cytology. After an introduction comparable to the content of a cytology seminar, 11 students of veterinary medicine were asked to assess 20 slides with different lymphadenopathies (hyperplasia: n = 4, lymphadenitis: n = 4, malignant lymphoma: n = 4, different other neoplasias: n = 8). A correct classification according to these categories was achieved on the average (median value) in 18 of the 20 cases (minimum: 16, maximum: 20). The majority of the investigators also found a correct further differentiation for some slides, particularly with respect to the type of non-lymphatic neoplasia. The results of this study emphasize that the experience gained in a single cytology seminar enables only few individuals to reliably differentiate lymphadenopathies. The error rate, which was 10 to 20% for the majority of the investigators, however, has to be assessed critically due to the clinical consequences of misdiagnosis. This emphasizes the necessity of experience even for interpretation of lymph node cytology.

3. [Chytridiomycosis in amphibians--first report in Europe].
PMID:日期:2000-10-01Declining of amphibian populations is a worldwide phenomenon. A cutaneous mycosis as a cause of death in free-living amphibians as well as in captive ones due to an chytrid fungus (Batrachochytrium dendrobatidis) was reported at first in 1998. This infections were reported hitherto from Australia, North, Central and South America. This is the first report on chytrid infections in captive anurans from Europe. Dendrobates auratus and D. pumilo imported from Costa Rica and P. vittatus imported from French Guayana died with chytridiomycosis within a week after arrival in Europe. Batrachocytrium was also found on captive bred frogs in Germany and Belgium. Clinical signs, diagnosis and conclusions for protecting free-living amphibian populations and captive frogs are discussed.

4. [Occurrence of zearalenone, alpha- and beta-zearalenol in bile of breeding sows in relation to reproductive performance].
PMID:日期:2000-10-01Although zearalenone-induced reproductive disorders and the clinical appearance of hyperestrogenism were reproduced and documented quite often the role of zearalenone-contaminated fodder as a cause for fertility problems in sow breeding is still discussed controversial. Therefore the correlation of zearalenone and zearalenone-derivatives in bile (n = 794) and feed (n = 158) with fertility problems of unknown origin was investigated in this study. For the analysis of zearalenone and its derivatives in bile a HPLC/EIA combination was used. On the one hand, this procedure guaranteed the quantitatively reproducible detection, on the other hand, the investigation expenditure could be kept small with regard to a later effort in the routine diagnostics. The detection limits for zearalenone, alpha- and beta-zearalenol in bile were at 1.0 ng/ml, 1.0 ng/ml, and 3.0 ng/ml, respectively. Results were confirmed by GC-MS. Zearalenone and zearalenone-derivatives were detected in almost every bile analysed. The contamination rate was 96.2%. In opposition to recent investigations beta-zearalenol was perceived as a relevant metabolite in swine. The contamination rate of feeding stuffs was 25.9%. Incubation of samples with beta-glucosidase did not elevate the detected amounts of zearalenone. As the measurable concentrations in bile and fodder were only slightly correlated the analysis of bile represents a reasonable alternative for fodder investigation. However, a correlation between the occurrence of zearalenone, alpha- and beta-zearalenol in bile of sows and non-infectious reproductive disorders could not be established at the loading level found. These results are in line with those statements obtained in feeding experiments regarding the risk evaluation of zearalenone in sow reproduction.

5. [Anti-inflammatory, antibacterial and antimycotic effects of dark sulfonated shale oil (ichthammol)].
PMID:日期:2000-10-01Dark sulfonated shale oil (Ichthammol, ammoniumbituminosulfonate) is an active ingredient of natural origin that was included in Annex II of European Council Regulation (EEC) 2377/90. Therefore, this well-tolerated substance is available further on as a dermatological agent for the application to animals from which food is produced for human consumption. The manifold pharmacological actions could be substantiated in numerous in-vitro studies using the Ichthammol from Seefeld, Austria. The antiinflammatory action which is well-known from clinical experience could be traced back to an influence of the substance on the formation, secretion and effect of inflammation mediators. Antibacterial and antimycotic actions which are well-known from clinical use could be confirmed in in-vitro studies as well.

6. [Contribution to the differentiation of cross-reacting antibodies in brucellosis serology--1. Reactions with various Yersinia serotypes and antibody avidity].
PMID:日期:2000-10-01Brucella and Yersinia enterocolitca O:9 (Y.e.O:9) have antigenic determinants in common causing cross-reactions which interfere strongly in Brucella serology. Hoffmann (1988) observed cross-reactivity between Y.e.O:9, Y.e.O:5, and Y.e.O:6 and used an ELISA for the differentiation between Brucella- and Yersinia antibodies since Y.e.O:5 and Y.e.O:6 produced only low-level cross-reactions with Brucella antigen. The observations of Hoffmann were confirmed and led to the construction of an ELISA with 4 antigens. Sera from natural outbreaks of brucellosis and from artificial infections with Brucella and Y.e.O:9 were properly identifiable by this system. Furthermore, the test material has been used to measure the antibody avidity by a specially designed ELISA. This test produced a further divergence between specific and non-specific reactions.

7. [Influence of diagnostic tests for estimating prevalence during surveillance programs].
PMID:日期:2000-09-01The influence of diagnostic tests on the estimation of the prevalence and the calculation of sample sizes with respect to different sampling schemes are presented in this paper. These sampling schemes are used for the implementation of surveillance programs. Assuming "perfect tests" (i.e. sensitivity = specificity = 100%) the calculated sample sizes, e.g. for an IBR/IPV-surveillance or a paratuberculosis survey, are half of the sample sizes considering sensitivity and specificity of the diagnostic tests. However the probability not to identify infected livestocks may be 4 times higher neglecting the test characteristics and assuming perfect tests.

8. [Determination of breakpoints for veterinary medically relevant antibiotics for resistance assessment of veterinary pathogens].
PMID:日期:2000-09-01This article describes the meaning of the term clinical breakpoint. This is followed by a discussion of the parameters that need to be considered when setting valid breakpoints for active substances in veterinary medicine; in doing so we closely follow equivalent regulations and guidelines on the establishment of breakpoints in human medicine. Along with pharmacokinetic data and the results of clinical efficacy tests, susceptibility data of relevant organisms play a key role in the establishment of breakpoints. Published breakpoints are currently only available for a few modern drugs in veterinary medicine.

9. [Technique of abdominal ultrasonography in newborn foals and normal findings].
PMID:日期:2000-09-01Under field conditions, the diagnosis of foal's diseases relies almost exclusively on the physical examination. As the signs of illness in the equine neonate are frequently vage and non-localizing, the diagnosis of diseases may be problematic. This often causes misinterpretations and leads to ineffective prophylaxis and treatment. The purpose of this study was to evaluate the usefulness of diagnostic ultrasonography of the foal's abdomen under field conditions to provide an optimized technique and to describe the normal findings. Diagnostic ultrasonography of the abdomen was performed after obtaining clinical history and passing the physical examination of 25 foals without signs of abdominal problems. The foals were scanned in a stable box, being restrained by three persons in semi-lateral recumbency. Usually, sedation was not necessary. The ventral abdominal wall was clipped, a generous amount of ultrasound coupling gel was applied and massaged on the skin surface. The ultrasonographic examination was carried out using a portable sector scanner ("Microimager 2000", Ausonics) with 5.0 and 7.5-MHz transducers or a combined 5.0 and 7.5-MHz transrectal linear-array scanner ("450 Enhanced", Pie Medical). Employing the 5.0-MHz sector scanner first, the abdomen was explored from caudal to cranial in left and right semi-lateral recumbency. The 7.5-MHz scanner was used to attain higher resolution of certain structures. The sector scanner turned out to be suitable under field conditions and adequate to examine the abdominal organs. The transrectal linear-array scanner also provided the most important informations, although it was difficult to maintain a good contact area of the scan head. By ultrasonography it was possible to identify the urinary bladder, kidneys, spleen, liver and part of the gastrointestinal tract. Thus, application of ultrasound could successfully be performed on newborn foals under field conditions.

10. [Possibilities for standardization of ELISA for detection of Salmonella antibodies in sera and meat juices of pigs].
PMID:日期:2000-09-01Programmes for controlling salmonella infections in German piggeries are based on the meat-juice-ELISA conducted in various investigation centres by using different test-kits. A usual procedure for harmonization (standardisation) of results is the calculation of the percentage of antibody-concentration from field samples in relation to the extinctions of a set of control-sera with known antibody concentrations. Whether this system is still acceptable in case of using different test-kits seems to be questionable. In principle, difficulties arise by calculating field results from the regression curve of control-sera because the calculated percentages of antibodies do not represent the antibody concentration but, instead, the percentages of the extinctions measured, and secondly, because control-sera presently in use are directed against different salmonella serovars. In regard to the number of laboratories involved and because of a variety of test-kits used it seems to be more adequate to include only one anti-Salmonella Typhimurium standard-serum at a given antibody concentration which is to be tested repeatedly on every test-plate. Simultaneously, further controls should include another anti-Salmonella Typhimurium and one anti-Salmonella Choleraesuis serum which should provide results similar to the Danish system which is regarded as a standard. As well, a negative serum must be included in the test and a minimum difference in extinctions between this negative serum and the standard positive control-serum should be reached to prove the validity of results from the test plate.
