1. Modern Nanocarriers as a Factor in Increasing the Bioavailability and Pharmacological Activity of Flavonoids.
PMID:日期:2022-01-01This review is devoted to modern systems of nanocarriers that ensure the targeted delivery of flavonoids to various organs and systems. Flavonoids have wide range of effects on the human body due to their antioxidant, anti-inflammatory, antitumor, antimicrobial, antiplatelet and other types of activity. However, the low bioavailability of flavonoids significantly limits their practical application. To overcome this disadvantage, serious efforts have been made in recent years to develop nanoscale carriers for flavonoids. This is particularly important in view of the known antitumor effect of these compounds, which allows them to target tumor cells without affecting surrounding healthy tissues. Nanocarriers provide increased penetration of biologicals into specific organs in combination with controlled and prolonged release, which markedly improves their effectiveness. This review summarizes data on the use of phytosomes, lipid-based nanoparticles, as well as polymeric and inorganic nanoparticles; their advantages and drawbacks are analyzed; the prospect of their use is discussed that opens new possibilities for the clinical application of flavonoids.

2. The Effect of Disinfectants on the SARS-CoV-2 RNA Detection in Swabs from Various Surfaces.
PMID:日期:2022-01-01The COVID-19 pandemic has spread rapidly around the world; some countries have introduced controls on imported products, including testing for viral nucleic acids. In this work, the influence of disinfectants for treating various SARS-CoV-2-contaminated surfaces on the detection of viral RNA fragments in swabs from these surfaces was analyzed using quantitative RT-PCR. Quaternary ammonium salt, hydrogen peroxide, 1-propanol, and sodium salt of dichloroisocyanuric acid, as well as ultraviolet irradiation, were tested as such disinfecting agents. Our results show that without exposure to disinfectants, viral RNA can be detected on the surface of all examined materials for at least 3 days. UV irradiation or irrigation with a disinfectant containing 0.2% active chlorine had the greatest effect on the decontamination of nonporous surfaces as measured by RT-PCR of swabs from these surfaces. Irrigation with disinfectants of porous surfaces (cardboard) had practically no effect on the detection of SARS-CoV-2 RNA by RT-PCR.

3. Rapid Assessment of Neutralizing Antibodies Using Influenza Viruses with a Luciferase Reporter.
PMID:日期:2022-01-01Influenza viruses cause acute respiratory infections, especially in the autumn-winter period. They are characterized by a high mutation frequency and cause annual seasonal epidemics. The detection of antibodies that neutralize the virus is an important criterion in the assessment of population immunity and the influenza vaccine effectiveness. In this study, a method for determining the titer of virus-neutralizing antibodies in blood serum has been developed. A new test called the luciferase neutralization assay uses a bioluminescent signal for detection. The assay is based on engineered influenza reporter viruses with various surface antigens and a nanoluciferase reporter protein in the NS1 reading frame. Using the developed method, we studied paired sera of volunteers obtained before and after vaccination. The proposed assay was compared with the conventional antibody assessment methods (microneutralization and hemagglutination inhibition assay); a high degree of correlation was observed. At the same time, the use of the luciferase neutralization assay made it possible to reduce the time required for the analysis and to simplify the detection procedure. 4. Microalgae in Terms of Biomedical Technology: Probiotics, Prebiotics, and Metabiotics.
PMID:日期:2022-01-01Green, red, brown, and diatomic algae, as well as cyanobacteria, have been in the focus of attention of scientists and technologists for over 5 decades. This is due to their importance as efficient and economical producers of food additives, cosmetics, pharmaceuticals, biofertilizers, biofuels, and wastewater bioremediation agents. Recently, the role of microalgae has increasingly been considered in terms of their probiotic function, i.e., of their ability to normalize the functioning of the microbiota of humans and agricultural animals and to produce biologically active substances, including hormones, neurotransmitters, and immunostimulators. A separate brief subsection of the review deals with the potential functions of microalgae with respect to the brain and psyche, i.e., as psychobiotics. Moreover, algal polysaccharides and some other compounds can be broken down to short fragments that will stimulate the development of useful intestinal microorganisms, i.e., function as efficient prebiotics. Finally, many components of microalgal cells and chemical agents produced by them can exert important health-promoting effects per se, which indicates that they are as potentially valuable metabiotics (the term preferred by late Prof. B.A. Shenderov), which are alternatively denoted as postbiotics in the literature.

5. Lactobacilli: Legal Regulation and Prospects for New Generation Drugs.
PMID:日期:2022-01-01The global probiotics industry has been undergoing major changes in recent years. Approaches to finding and creating new probiotics, as well as a paradigm of their use in food, medicine, and pharmacology are changing. The catalyst proved to be the increasing popularity and availability of omics technologies, in particular, metagenomic studies of human and animal microbiomes. However, the efficiency and safety of drugs based on probiotic strains, as well as their marketing rates, largely depend on the levels of legal and technical regulation in the field. The present review discusses the aspects of legal regulation in Russia, the European Union and the United States, along with the advantages and disadvantages of probiotics and postbiotics. A consensus is emerging that postbiotics have a number of advantages over classical live probiotic cultures. The review also focuses on the lactobacilli family, which includes the largest number of probiotic strains studied so far and still holds a leading position among probiotics. On the legislative front, Russia is often ahead of its time with adopting such laws as the Federal Law No. 492-FZ on biosecurity, which defined the concept of human and animal microbiota and set forth legislative guidelines for its preservation. The new field of research referred to as microbiome nutrigenomics aims to achieve this goal.

6. Synthesis of a Bioactive Composition of Chitosan-Selenium Nanoparticles.
PMID:日期:2022-01-01A biologically active composition of chitosan-selenium nanoparticles has been developed. Selenium nanoparticles are characterized by a clear bimodal size distribution: 2-3 and ~37 nm. The main active centers of complexation with nanoparticles are the amino and hydroxyl groups of chitosan. In experiments on culturing fibroblasts of the hTERT BJ-5ta cell line on sample films, high biocompatibility of the composition was shown. It was shown that the composition of chitosan-selenium nanoparticles has a corrective effect on the oxidative processes of the body, reducing the activity of free-radical oxidation in the blood of animals. This opens up prospects for the use of this complex in the composition of antioxidant and adaptogenic drugs.

VKM Ac-2618D strain has been identified, and its morphological and physiological features have been studied in relation to the production of the immunosuppressant tacrolimus. The phenotypic variability of the strain was analyzed, and a dissociant with a high level of tacrolimus production was selected. Based on a comprehensive study of morphological, physiological, and chemotaxonomic properties and on phylogenetic analysis, the strain was named VKM Ac-2618D. The strain genome contains the full version of the tacrolimus biosynthetic gene cluster. The advantages of fed-batch cultivation mode for tacrolimus biosynthesis are shown. The results broaden the understanding of the characteristics of polyketide biosynthesis and can be used in the development of technology for tacrolimus production.

8. Loop-Mediated Isothermal Amplification as a Promising Method for Mass COVID-19 Diagnostics.
PMID:日期:2021-01-01Real-time reverse-transcription polymerase chain reaction (RT-PCR) is currently the most popular method for early COVID-19 diagnostics. However, loop-mediated isothermal amplification (LAMP) is superior to real-time RT-PCR in rapidity and simplicity, since it does not require expensive laboratory equipment and trained personnel. LAMP-based diagnostic kits for COVID-19 testing already exist, but corresponding tests are not yet widely available. The method has great potential for mass application. Here, we discuss the technical and methodological aspects of its widespread adoption.

PMID:日期:2021-01-01This work describes a modification of the pRH2521 vector of the pRH2502/pRH2521 system for CRISPR-dCas9-mediated RNA interference. The modification enabled an increase in the cloning efficiency of guide RNA spacers. The ability of the modified pRH2502/pRH2521 system to suppress the transcription of certain genes was evaluated with the use of genes of adenylate cyclases. The results revealed the limitations of the pRH2502/pRH2521 system for CRISPR interference associated with the probability of the detection of a protospacer adjacent motif (PAM) in the gene promoter region.

10. Use of Mathematical Methods for the Biosafety Assessment of Agricultural Crops.
PMID:日期:2021-01-01In Russia and around the world, there are important questions regarding the potential threats to national and biological safety created by genetic technologies and the need to improve or introduce new, justified, and adequate measures for their control, regulation, and prevention. The article shows that a significant volume of the global market is occupied by five major transgenic crops, and producers are ready to switch to crops with an edited genome that has been approved in the United States, Argentina, and other countries. We propose a qualitatively new approach to the risk assessment of edited plants, "Safe Design," and we have also developed an extremely important, fundamentally new approach to the development of methods that combine next-generation sequencing (NGS) and Bioinformatics for the assessment of the crop import biosafety. The proposed mathematical approach provides a detailed analysis of the possible insertions of DNA fragments into the genome of edited crops and a clarification of their biological significance. The developed method can be used in the rapid screening of plants for the presence of potentially dangerous genes, viral sequences, and nonspecific promoter sequences.
