2. A GRX-SOD module maintains superoxide homeostasis and meristem development in maize.
PMID:日期:2026-09-04The continuous initiation of plant aerial organs relies on the maintenance of dynamic stem cell homeostasis within the shoot apical meristem (SAM). Although reactive oxygen species (ROS) have emerged as crucial signals determining cell fate, how plants precisely maintain SAM-specific ROS homeostasis remains to be elucidated. Here, we report that a redox regulatory module comprising the maize CC-type glutaredoxin MSCA1 and its homologs regulates meristem architecture by maintaining superoxide homeostasis in the SAM. We found that the superoxide anion (O) is specifically enriched in the central zone of the maize SAM, whereas the loss of the GRX module leads to strikingly decreased O accumulation, resulting in SAM size reduction and impaired leaf formation. MSCA1 physically interacts with the superoxide dismutase ZmCSD5, and loss of GRX function is associated with elevated total SOD activity, supporting a working model in which MSCA1 may restrain ZmCSD5-associated SOD activity and thereby limit excessive O scavenging in the SAM. Furthermore, loss-of-function zmcsd5 mutants exhibit a significantly enlarged SAM, genetically supporting the proposed MSCA1-ZmCSD5 regulatory module. Our findings reveal how superoxide homeostasis is precisely maintained in the SAM to control development, providing potential genetic targets for the molecular improvement of crop architecture and yield traits.

4. MYDGF as a telomerase activator and therapeutic target for osteoarthritis.
PMID:日期:2026-08-31Osteoarthritis (OA) involves progressive cartilage breakdown. This is driven by impaired chondrocyte function and an imbalance in the extracellular matrix homeostasis. However, the key upstream regulators that maintain healthy chondrocytes are still not fully known. Here, we report that myeloid-derived growth factor (MYDGF) is a novel regulator of telomerase activity and is critical for cartilage health. Using a genome-wide CRISPR-Cas9 screen with a TERT reporter system, MYDGF was identified as a strong positive regulator of telomerase. Functional studies confirmed that MYDGF positively regulates TERT expression and telomerase activity in both HeLa and ATDC5 cells. In a mouse surgical destabilization of the medial meniscus (DMM) OA model, MYDGF knockout (KO) mice exhibited more severe cartilage damage. Conversely, delivering MYDGF into the knee joint using AAV virus partially alleviated cartilage injury. Transcriptome profiling revealed that MYDGF-deficient chondrocytes exhibit downregulated key pathways involved in matrix building and upregulated inflammatory signals, indicating a shift in cell state. In summary, our work establishes MYDGF as a key upstream factor for both telomerase and cartilage matrix homeostasis remodeling. It connects telomerase function to chondrocyte health. These findings also highlight MYDGF as a promising new target for treating osteoarthritis.

5. BEST4⁺ cells: a potential hub of intestinal ion transport and diarrhea manipulation.
5. 最佳4⁺ 细胞:肠道离子转运和腹泻控制的潜在枢纽PMID:日期:2026-08-12BEST4⁺ cells are a recently identified, functionally specialized intestinal epithelial cell population characterized through single-cell and spatial transcriptomic analyses in humans, pigs, rats, and other vertebrates. These cells characteristically express BEST4, OTOP2, CA7, and GUCY2C, and exhibit particularly high CFTR expression in the small intestine, supporting their roles in luminal pH sensing, fluid-electrolyte homeostasis, and mucus hydration. Emerging evidence from organoid and cross-species studies supports a predominant role for the NOTCH-SPIB signaling axis in driving BEST4⁺ cell differentiation, but their lineage identity and regulatory mechanisms remain incompletely resolved. This review focuses on intestinal epithelial BEST4⁺ cells as a critical cellular hub that connects ion transport physiology with the pathophysiology of secretory diarrhea. We summarize the molecular identity, regional distribution, and cross-species conservation of BEST4⁺ cells. We then evaluate their roles in luminal pH regulation, electrogenic fluid secretion, and mucus barrier integrity. We detail how bacterial enterotoxins activate the GC-C/cGMP/CFTR and cAMP/PKA/CFTR pathways in BEST4⁺ cells to drive pathological fluid hypersecretion, and discuss how viral infections may indirectly engage or amplify BEST4⁺ cell-associated ion-transport pathways. We also outline their relevance to viral diarrhea, inflammatory bowel disease, and cystic fibrosis-associated intestinal dysfunction. Finally, we assess established antidiarrheal approaches and investigational strategies that modulate BEST4⁺ cell-associated pathways, and emphasize the need for cell-type-specific validation in physiologically relevant in vivo models.

6. A safety-aware intake modeling framework for translating edible algal biomass into nutritional bioproducts.
PMID:日期:2026-08-10Edible algal biomass has emerged as a promising platform for nutritional bioproduct development, but its practical translation depends on compositional variability, intake feasibility, and safety constraints. Here, we integrated biochemical profiling, trace element screening, age-specific intake modeling, and community consumption context to evaluate commercially available and supplier-provided microalgal and seaweed products within realistic application boundaries. Selected products differed substantially in macronutrient composition, fatty acid profiles, pigments, polyphenols, and mineral composition, indicating product- and species-specific roles as nutritional biomass rather than interchangeable ingredients. Across species and age groups, approximately 0.7 to 3.1 g dry biomass provided 20% of age-specific alpha-linolenic acid intake, with approximately 1 g sufficient for several products. Multi-species allocations improved omega-3 representation while remaining within practical intake limits and trace element-based screening thresholds. Community consumption data from Pacific Island children showed low and episodic seaweed intake, highlighting a gap between biomass potential and routine dietary use. These findings support safety-aware modeling as a preliminary screening and decision-support strategy for evaluating edible algal biomass before further product-specific validation.

7. A total infectome framework for resolving complex disease etiology in aquaculture.
PMID:日期:2026-08-07Traditional pathogen isolation can establish disease causality but often fails in complex infections involving unknown or multiple microbial backgrounds, whereas metatranscriptomics comprehensively captures the total infectome without directly resolving the primary etiological agent. Here, using overwintering syndrome (OWS) in grass carp, a widespread but unresolved aquaculture disease in China, as a disease model, we developed an integrated strategy that links comparative total infectome analysis with targeted isolation, experimental infection, and post-challenge infectome validation. Multi-organ metatranscriptomic profiling of healthy and diseased grass carp identified 107 microorganisms spanning viruses, bacteria, and eukaryotic microbes. Comparative infectomics prioritized Flavobacterium psychrophilum through its consistent disease enrichment, lesion-site abundance, multi-organ dissemination, and recurrence across affected hosts. Infection with F. psychrophilum alone reproduced the clinical signs, tissue pathology, mortality patterns, and infectome signatures of natural OWS, establishing it as the primary etiological agent. Temperature-shift experiments further showed that disease progression is promoted by warming from low temperature conditions, explaining the seasonal emergence of OWS. This work resolves a major grass carp disease and provides a generalizable framework for converting unbiased pathogen discovery into causal inference in complex infectious disease systems.

8. Engineering monosex and sterile fish for food production: from conventional methods to CRISPR-based precision.
PMID:日期:2026-08-04Monosex and sterile fish populations are increasingly recognized as essential tools for enhancing aquaculture productivity while safeguarding ecosystems from genetic introgression and uncontrolled reproduction. Conventional approaches such as hormonal manipulation and triploidy have demonstrated utility but face persistent limitations in efficiency, welfare, and regulatory acceptance. Recent breakthroughs in CRISPR-based genome editing and germ cell transplantation now enable heritable, scalable, and biocontained control of sex and fertility, offering transformative potential for sustainable aquaculture. This paper synthesizes advances across finfish and crustaceans, critically evaluates technical and regulatory bottlenecks, and highlights convergent solutions integrating automation, artificial intelligence, and governance frameworks. By positioning reproductive programming as a cornerstone of climate-resilient and sustainable aquatic food systems, we propose a roadmap toward commercial adoption that balances biological precision, ecological safety, and societal acceptance.

9. Phages released from Acidithiobacillus ferrooxidans enhance chalcopyrite bioleaching by alleviating passivation and promoting sulfur turnover.
PMID:日期:2026-07-27Chalcopyrite is the most abundant copper-bearing sulfide mineral, characterized by a stable crystal structure. Bioleaching of chalcopyrite is inherently slow and further constrained by surface passivation layers during prolonged bioleaching. In this study, bacteriophages released from Acidithiobacillus ferrooxidans were introduced to promote chalcopyrite bioleaching. Mineralogical characterization, microbial profiling, and functional gene analysis were employed to reveal the underlying mechanisms. One day after phage introduction, phage abundance was 9.42 × 10 VLPs mL in the phage-treated group, 150-fold higher than in the control (6.26 × 10 VLPs mL). Over a 60-day leaching period, the copper leaching efficiency of the phage-treated group was 31.72%, significantly higher than that of the control (20.43%). Phage introduction alleviated surface passivation, suppressed elemental sulfur (S) and jarosite-type deposition, and increased the apparent product-layer diffusion rate constant 3.60-fold. Bacterial diversity and the relative abundance of heterotrophic bacteria were elevated. The increase in heterotrophic bacteria may alleviate the inhibitory effects of organic matter on autotrophic bioleaching microorganisms. Genes associated with the Sox system, dissimilatory sulfur metabolism, and organic sulfur transformation were significantly enriched in the phage-treated group, consistent with enhanced S turnover and reduced surface accumulation. Together, these results indicate that phage introduction enhances chalcopyrite bioleaching by alleviating surface passivation and promoting sulfur turnover through coupled interfacial and microbial regulation. This study provides a novel approach for sustainable intensification of bioleaching of refractory sulfide minerals.

10. Metabolic engineering of cereal lipids: from omega-3 fatty acids to wax esters and pheromones.
PMID:日期:2026-07-24Cereals are emerging as attractive platforms for the sustainable production of high-value lipids through metabolic engineering. Although plant lipids play essential biological roles and have considerable economic value, their conventional production from natural sources is often limited by sustainability, scalability and cost. Recent advances in synthetic biology enable the reprogramming of seed lipid metabolism for the tailored synthesis of valuable lipid compounds. In this review, we first summarize the core pathways of fatty acid biosynthesis and triacylglycerol assembly in seeds, together with the genetic transformation and genome editing toolkits available for major cereals. We then highlight recent progress in the heterologous production of specialized lipids, including eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), wax esters, and insect sex pheromones, in engineered plant systems. Finally, we discuss the potential of cereals as scalable and sustainable platforms for the production of high-value lipids. Together, these advances position engineered cereals as promising plant-based factories for applications in agriculture, nutrition, and the emerging bio-based economy.
