1. Techniques for analytical estimation of COVID-19 clinical candidate, niclosamide in pharmaceutical and biomedical samples.
PMID:日期:2022-09-08Niclosamide is a well-known broad-spectrum antiparasitic drug used for human as well as veterinary tapeworm infections. Recently, it attracted attention as an antiviral agent for treating coronavirus disease 2019. It is administered orally in humans to treat tapeworm infections. Furthermore, it is a registered pesticide and molluscicide to control infections in the aquaculture industry. Its chronic environmental exposure has potential toxicities when such contaminated seafood is consumed. Therefore, monitoring its residual concentration in food products (seafood, water, water waste, etc.) and pharmaceuticals (active pharmaceutical ingredients, bulk drugs, and formulations) is imperative. The present review critically investigates the sophisticated techniques employed for analyzing niclosamide, its degradation products, and metabolites in various samples and matrices. The future scope for green analytical methods, green sample extraction and preparation is also deliberated.

2. Online Monitoring of Small Volume Reactions Using Compact Liquid Chromatography Instrumentation.
PMID:日期:2022-03-31A wide variety of analytical techniques have been employed for monitoring chemical reactions, with online instrumentation providing additional benefits compared to offline analysis. A challenge in the past for online monitoring has been placement of the monitoring instrumentation as close as possible to the reaction vessel to maximize sampling temporal resolution and preserve sample composition integrity. Furthermore, the ability to sample very small volumes from bench-scale reactions allows the use of small reaction vessels and conservation of expensive reagents. In this study, a compact capillary LC instrument was used for online monitoring of as small as 1 mL total volume of a chemical reaction mixture, with automated sampling of nL-scale volumes directly from the reaction vessel used for analysis. Analyses to demonstrate short term (~2 h) and long term (~ 50 h) reactions were conducted using tandem on-capillary ultraviolet absorbance followed by in-line MS detection or ultraviolet absorbance detection alone, respectively. For both short term and long term reactions (10 and 250 injections, respectively), sampling approaches using syringe pumps minimized the overall sample loss to ~0.2% of the total reaction volume.

3. UHPLC assay and impurity methods for diphenhydramine and phenylephrine hydrochloride oral solution.
PMID:日期:2020-01Many over-the-counter drug products lack official compendial analytical methods. As a result, the United States Pharmacopeia and the United States Food and Drug Administration are seeking to develop and validate new methods to establish analysis standards for the assessment of the pharmaceutical quality of over-the-counter drug products. Diphenhydramine and phenylephrine hydrochloride oral solution, a combination drug product, was identified as needing a compendial standard. Therefore, an ultra-high-performance liquid chromatography method was developed to separate and quantify the two drug compounds and eleven related organic impurities. As part of a robustness study, the separation was demonstrated using different high-performance liquid chromatography systems and columns from different manufacturers, and showed little dependence with changes in flow rate, column temperature, detection wavelength, injection volume and mobile phase gradient. The method was then validated conformant with the International Council for Harmonisation guidelines. For impurities, adequate specificity, linearity, accuracy and precision were demonstrated. For assay, a slight modification to the injection volume was necessary to achieve adequate analytical performance. With successful development and validation, these methods were shown to be suitable for their intended purpose and may be considered for adoption as compendial procedures.

4. Rapid Formation of Polymer Frits in Fused Silica Capillaries Using Spatially defined Thermal Free-Radical Initiated Polymerization.
PMID:日期:2018-11-19Column preparation in capillary chromatography commonly relies upon the generation of on-column porous frits. Here, we report a simple, robust and low-cost approach for preparing polymer frits on-column, in a rapid and spatially controlled manner using thermal free-radical initiated polymerization. In this approach, a simple, temperature-controlled heating apparatus is positioned adjacent to a 100 m i.d. fused-silica capillary for a defined duration. Frits were synthesized in 3-(trimethoxysilyl)propyl methacrylate modified capillaries using a monomer solution of 2,2-azobisisobutyronitrile, glycidyl methacrylate, ethylene glycol dimethacrylate, and decanol. Frit length and stability were investigated as a function of polymerization time and temperature. Frit length was easily controlled via a combination of polymerization time and temperature and position was readily controlled using a simple mechanical placement jig. Thermal initiated frits were stable throughout column packing and did not require removal of the capillary polyimide coating. The thermal initiation approach offers higher throughput, with polymerization times of < 2 min compared to ≥ 30 min for UV-initiated polymerization and significantly reduces the cost, enabling broader access to on-column frit technology for a variety of capillary separation applications.
