1. A Simple and Fast Method for Derivation of blastocyst-like cell aggregates (BLCAs) from Murine Embryonic Stem Cells.
PMID:日期:2018-07The study of blastocyst differentiation from stem cells is still at its stage of infancy. In this study, a simple and efficient method was developed to derive blastocyst-like cell aggregates (BLCAs) from murine embryonic stem cells (mESCs). This was achieved by culturing the mESCs in non-adherent dishes.0020BLCAs (13±2 per million mESCs) were obtained in only 5 days and they were found to express the protein and gene markers of trophoblasts. Furthermore, the BLCAs showed the normal trophoblasts' capability of penetrating the uterine wall after being transferred into pseudo-pregnant mice, indicating the trophoblast-like functionality of the derived BLCAs. Collectively, we successfully developed a simple and fast 3D suspension culture approach to differentiate mESCs into BLCAs, which may provide important clues into the development of early embryos and stem cells. This may also offer a simple and efficient approach to isolate trophoblast cells for studying the native microenvironment of pluripotent stem cells without the need to sacrifice animals.

2. Engineering Microvascularized 3D Tissue Using Alginate-Chitosan Microcapsules.
PMID:日期:2017-02-01Construction of vascularized tissues is one of the major challenges of tissue engineering. The goal of this study was to engineer 3D microvascular tissues by incorporating the HUVEC-CS cells with a collagen/alginate-chitosan (AC) microcapsule scaffold. In the presence of AC microcapsules, a 3D vascular-like network was clearly observable. The results indicated the importance of AC microcapsules in engineering microvascular tissues -- providing support and guiding alignment of HUVEC-CS cells. This approach provides an alternative and promising method for constructing vascularized tissues.

3. Patterning Pluripotent Stem Cells at a Single Cell Level.
PMID:日期:2013-08-01Studies of cell-extracellular matrix (ECM) interactions at a single cell level have drawn interest from scientists around the world. Subcellular ECM micropatterning techniques allow researchers to control cell shape, migration, and spindle orientation during mitosis potentially influencing the stem cell fate. Generally these studies have been limited to somatic cells rather than human pluripotent stem cells (hPSCs) which are capable of enormous differentiation potential. hPSCs require a defined ECM for attachment and express characteristic integrins mediating cell-substrate interactions. hPSCs also rely on cell-cell contacts for survival and to maintain self-renewal properties, but these circumstances also significantly limit hPSC observation at a single cell level. In addition, currently available methods for ECM micropatterning generally require a facility with trained personnel and intricate equipment to produce protein micropatterns. To overcome this problem, we have developed a protocol for vitronectin micropatterning using simple UV/ozone modification of polystyrene. Single hPSCs were able to attach and form characteristic stress fibers and focal adhesions similar to somatic cell types which demonstrate hPSC responsiveness to extracellular adhesive cues. Micropatterned hPSCs were able to be cultured for up to 48 hours while maintaining expression of pluripotency-associated transcription factor OCT4. Although further studies are necessary, the results of our investigation will potentially have a large impact on cell regenerative medicine and tissue engineering.

4. A sulfated nanofibrous mesh supporting the osteogenic differentiation of periosteum-derived cells.
PMID:日期:2013-08-01The periosteum is a thin fibrous membrane covering the surface of long bone and is known to play a critical role in bone development and adult bone fracture healing. Loss or damage of the periosteum tissue during traumatic long bone injuries can lead to retarded healing of bone graft-mediated repair. The regenerative potential of periosteum-derived progenitor cells (PDCs) has inspired their use as an alternative to bone marrow-derived mesenchymal stromal cells (MSCs) to augment scaffold-assisted bone repair. In this study, we first demonstrated that PDCs isolated from adult rat long bone exhibited innate advantages over bone marrow-derived MSCs in terms of faster proliferation and more potent osteogenic differentiation upon induction in plastic-adherent culture. Further, we examined the potential of two electrospun nanofibrous meshes, an uncharged regenerated cellulose mesh and a sulfated mesh, to support the attachment and osteogenic differentiation of PDCs. We showed that both nanofibrous meshes were able to support the attachment and proliferation of PDCs and MSCs alike, with the sulfated mesh enabling significantly higher seeding efficiency than the cellulose mesh. Both meshes were also able to support the osteogenic differentiation of adherent PDCs upon induction by osteogenic media, with the sulfated mesh facilitating more potent mineral deposition by adherent PDCs. Our study supports the sulfated nanofibrous mesh as a promising synthetic periosteal membrane for the delivery of exogenous PDCs to augment bone healing.

5. Laminin Functionalized Biomimetic Nanofibers For Nerve Tissue Engineering.
PMID:日期:2013-08-01Large-gap peripheral nerve injuries present a significant challenge for nerve regeneration due to lack of suitable grafts, insufficient cell penetration, and repair. Biomimetic nanofibrous scaffolds, functionalized on the surface with extracellular matrix proteins, can lead to novel therapies for repair and regeneration of damaged peripheral nerves. Here, nanofibrous scaffolds electrospun from blends of poly(caprolactone) (PCL) and chitosan were fabricated. Taking advantage of the amine groups on the chitosan, the surface of the scaffolds were functionalized with laminin by carbodiimide based crosslinking. Crosslinking allowed laminin to be attached to the surfaces of the PCL-chitosan nanofibers at relatively high concentrations that were not possible using conventional adsorption methods. The nanofibrous meshes were tested for wettability, mechanical properties and cell attachment and proliferation. Blending of chitosan with PCL provided more favorable surfaces for attachment of Schwann cells due to the reduction of the contact angle in comparison to neat PCL. Proliferation rates of Schwann cells grown on PCL-chitosan scaffolds with crosslinked laminin were significantly higher than the rates for PCL-chitosan nanofibrous matrices with adsorbed laminin. PCL-chitosan scaffolds with modified surfaces via crosslinking of laminin could potentially serves as versatile substrates with excellent mechanical and surface properties for in vivo cell delivery for nerve tissue engineering applications.

6. {"_":"Rat Cortical Oligodendrocyte-Embryonic Motoneuron Co-Culture: An Axon-Oligodendrocyte Interaction Model.","i":["In Vitro"]}
PMID:日期:2012-09Mechanisms that control the differentiation and function of oligodendrocytes in the central nervous system are complex and involve multiple inputs from the surrounding environment, including localized concentrations of growth factors and the extracellular matrix. Dissection and analysis of these inputs are key to understanding the pathology of central nervous system demyelinating diseases such as multiple sclerosis, where the differentiation of myelinating oligodendrocytes from their precursors underlies the remission phase of the disease. co-culture models provide a mechanism for the study of factors that regulate differentiation of oligodendrocyte precursors but have been difficult to develop due to the complex nature of central nervous system myelination. This study describes development of an model that merges a defined medium with a chemically modified substrate to study aspects of myelination in the central nervous system. We demonstrate that oligodendrocyte precursors co-cultured with rat embryonic motoneurons on non-biological substrate (diethylenetriamine trimethoxy-silylpropyldiethylenetriamine), can be induced to differentiate into mature oligodendrocytes that express myelin basic protein, using a serum-free medium. This defined and reproducible model of myelination could be a valuable tool for the development of treatments for demyelinating diseases such as multiple sclerosis.

7. Fabrication of Mineralized Collagen from Bovine Waste Materials by Hydrothermal Method as Promised Biomaterials.
PMID:日期:2011-12In the present study, we aimed to produce mineralized-collagen by hydrothermal process. A simple method not depending on additional foreign chemicals has been employed to isolate the mineralized-collagen fibers from bovine waste. The process of extraction involves the use of hydrothermal method from available bovine bones. The structural and morphological properties of the collagen fibers were characterized by using scanning electron microscopy and transmission electron microscopy. These results indicated well received collagen fibers, having a diameter less than 1 m and with established mineral content in the individual fibers. The X-ray diffraction showed the crystalline feature of the obtained nano-compounds. The thermo gravimetric analysis was used to differentiate between the collagen and mineral parts of obtained product. Overall, the results generously indicated production of well received collagen fibers from bovine bones.

8. Thermoresponsive Nanocomposite Hydrogels: Transparency, Rapid Deswelling and Cell Release.
PMID:日期:2011-06-01Thermal modulation reversibly switches poly(-isopropylacrylamide) (PNIPAAm) hydrogels between a water-swollen and a deswollen state which is useful for a variety of biomedical applications. The utility and efficiency of PNIPAAm hydrogels requires tailoring their rate of deswelling/reswelling, mechanical properties and/or optical clarity. In the current work, we prepared novel thermoresponsive nanocomposite hydrogels comprised of a PNIPAAm hydrogel matrix and polysiloxane colloidal nanoparticles (~54 nm ave. diameter) via photopolymerization of aqueous solutions of NIPAAm monomer, ,'-methylenebisacrylamide (BIS, crosslinker), photoinitiator and 0.5-4.0 wt% polysiloxane nanoparticles (wt% solids of nanoparticles with respect to NIPAAm weight) at ~7 °C. At these nanoparticle concentrations, the nanocomposite hydrogels were more optically transparent versus those prepared with analogous larger nanoparticles (~219 nm ave. diameter). The volume phase transition temperature (VPTT) of the nanocomposite hydrogels was conveniently unaltered versus that of the pure PNIPAAm hydrogel. Incorporation of nanoparticles caused enhancement in modulus as well as the extent and rate of deswelling. When cooled from 37 °C to 25 °C, mouse smooth muscle precursor cells (10T1/2) were effectively detached from nanocomposite hydrogel surfaces due to hydrogel swelling.
