4. Carbon dots derived from natural medicines: Therapeutic advances and challenges.
4. 源自天然药物的碳点:治疗进展与挑战PMID:日期:2026-05-26Carbon dots (CDs) are a novel nanomaterial exhibiting exceptional biocompatibility, tunable fluorescence, and versatile functionality, making them a promising candidate in biomedicine. Natural medicines, including plants, animals, minerals, and their extracts, represent ideal precursors for CD synthesis due to their abundance, renewability, and eco-friendliness. This addresses sustainability and environmental concerns associated with conventional chemical precursors. Natural medicine-derived CDs (NM-CDs) feature quasi-spherical core-shell structures, ranging from 1 to 10 nm, with surface hydroxyl, carboxyl, and amino groups enhancing their hydrophilicity and functionality. Their properties can be further modulated by synthesis temperature and heteroatom doping. Critically, NM-CDs combine the pharmacological activities of natural medicines with the unique nanoscale properties of CDs, bridging traditional medicine and modern nanotechnology. This review provides a comprehensive overview of the development of NM-CDs, including green natural precursors, formation mechanisms, and synthesis strategies. It systematically analyzes the structure and properties of NM-CDs, categorizes their therapeutic applications into seven major disease types, and offers a detailed explanation of their fundamental pharmacological mechanisms. Finally, ten key opportunities and challenges for the sustainable development of NM-CDs are highlighted. By integrating efficient resource utilization, green chemistry principles, and clinical demands, NM-CDs exhibit strong potential to advance bio-based nanotherapeutics and enhance the value of natural medicines through modern scientific approaches.

5. A quantitative screening strategy based on activity-content ratio for prioritizing bioactive compounds in medicinal plants.
PMID:日期:2026-05-21Accurately evaluating the intrinsic bioactivity of individual constituents, independent of their abundance, remains a major challenge in natural product screening. To address this, we proposed the activity-content ratio (ACR) as a novel index for direct intrinsic inhibitory potency characterization of constituents. Utilizing an at-line nanofractionation platform, an ACR-based screening method for acetylcholinesterase inhibitors was established. The method was validated using both mixed standards and a complex extract of alkaloids. Results demonstrated a strong linear correlation ( > 0.93) between ACR% and IC for 16 active constituents. High-ACR% compounds, such as berberine, palmatine, and dehydrocorydalin, exhibited significant inhibitory activities at nanomolar concentrations. The model's broad applicability was further confirmed by screening flavonoids for α-glucosidase inhibitors, showing excellent linearity ( = 0.96) for the five identified active constituents. Notably, rutin displayed the highest ACR% and an IC of 15.21 ± 1.23 μM. The consistent performance of the ACR model across two distinct enzymatic targets and chemical families (alkaloids and flavonoids) demonstrates its reliability and broad applicability. This study provides a reliable and efficient quantitative tool for bioactivity evaluation, which is expected to greatly accelerate the discovery and quality control of bioactive medicinal plants.

7. Genipin induces liver injury by disrupting the CPS1-regulated urea cycle and redox homeostasis.
7. 京尼平通过破坏CPS1调控的尿素循环和氧化还原稳态诱导肝损伤PMID:日期:2026-05-07Genipin (GP) is the key metabolite of geniposide (GE), the primary active component of the traditional Chinese medicine Gardeniae Fructus. Its hepatotoxicity poses a potential risk to safe clinical administration, while its core pathophysiological mechanism remains unclear. This study systematically elucidated the molecular mechanisms of GP-induced hepatotoxicity through integrated multi-omics and functional validation. GP treatment induced significant liver damage and oxidative stress in rats, characterized by elevated serum transaminases (alanine aminotransferase (ALT) increased by approximately 63.5-fold; aspartate aminotransferase (AST) by 18.6-fold), an approximately 1.8-fold decrease in superoxide dismutase (SOD) activity, and an approximately 10-fold accumulation of malondialdehyde (MDA) (all < 0.001). Metabolomic and proteomic analyses revealed that GP binds to carbamoyl phosphate synthetase 1 (CPS1), triggering urea cycle dysfunction marked by citrulline depletion and significant downregulation of its downstream product fumarate. experiments confirmed that disrupting CPS1 impairs the urea cycle and induces oxidative stress, while CPS1 overexpression or exogenous fumarate supplementation significantly reversed oxidative stress. Clinical sample analysis revealed similar urea cycle metabolic disturbances, characterized by citrulline depletion and reduced fumarate levels, in plasma from patients with liver injury due to inappropriate use of Gardeniae Fructus-containing formulas, confirming clinical relevance. This study reveals a novel hepatotoxic mechanism whereby GP targets CPS1 to disrupt the urea cycle, reducing fumarate production and impairing nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant defense, thereby inducing oxidative stress. These findings provide a scientific basis for improving the safety evaluation and rational clinical use of Gardeniae Fructus-containing preparations.

10. Diosgenin presents a novel role in promoting diabetic wound healing: A mechanism involving Sirt6/Nrf2-mediated inhibition of ferroptosis.
10. 薯蓣皂苷元在促进糖尿病伤口愈合中展现出新作用:涉及Sirt6/Nrf2介导抑制铁死亡的机制PMID:日期:2026-04-13Delayed wound healing in patients with diabetes is frequently caused by the dysfunction of human umbilical vein endothelial cells (HUVECs). Under high-glucose and high-fat (HGHF) conditions, the microenvironment induces uncontrolled lipid peroxidation and subsequent iron-dependent ferroptosis. Diosgenin (DG), a steroidal saponin extracted from herbs, including wild yam, exhibits diverse pharmacological activities. The present study aimed to investigate the role and underlying mechanism of DG in HGHF-induced ferroptosis and diabetic wound (DW) healing. A DW model was established in mice by intraperitoneal injection of streptozotocin (STZ). HUVECs were cultured under HGHF conditions to induce the diabetic microenvironment. C11-BODIPY, FerroOrange probes, and hematoxylin-eosin (H&E)/Masson staining were used to determine lipid peroxidation, iron ion levels, and histological changes, respectively. Results demonstrated that DG alleviated HGHF-induced ferroptosis, as evidenced by reduced malondialdehyde (MDA), lipid peroxidation, and mitochondrial reactive oxygen species (ROS) levels, and increased glutathione (GSH) levels. Additionally, DG upregulated the expression of the anti-ferroptotic protein glutathione peroxidase 4 (GPX4), promoted angiogenesis, and accelerated wound healing in diabetic mice. Network pharmacology analysis identified the sirtuin 6/nuclear factor erythroid 2-related factor 2 (Sirt6/Nrf2) pathway as a potential target of DG, a finding validated by mechanistic studies. Silencing Sirt6 in HUVECs negated the protective effect of DG against ferroptosis. In conclusion, DG mitigates ferroptosis and promotes DW healing, at least partially through activating the Sirt6/Nrf2 pathway.
