1. Identification of MRI-Derived Structural Biomarkers in Female Alzheimer's Disease Subjects Using CAT12 and Mimics: Effects of Voxel Geometry on Biomarker Estimation.
PMID:日期:2026-09-06Alzheimer's disease (AD) is the leading cause of dementia and disproportionately affects women, who experience a higher lifetime risk and more rapid structural brain changes than men. Reliable imaging biomarkers are essential for detecting these changes, although their estimation may be influenced by voxel geometry, image resolution, and segmentation methodology. In this study, magnetic resonance imaging (MRI) scans from 40 female participants with AD obtained from the Alzheimer's Disease Neuroimaging Initiative (ADNI) database were analyzed using the Computational Anatomy Toolbox 12 (CAT12), implemented within Statistical Parametric Mapping 12 (SPM12), and Materialise Mimics to identify structural biomarkers associated with neurodegeneration. Cortical thickness, gray matter (GM), white matter (WM), cerebrospinal fluid (CSF), and total intracranial volume (TIV) were quantified, while Brain Parenchymal Volume (BPV) and Brain Parenchymal Fraction (BPF) were calculated to assess global brain tissue preservation. The analyses demonstrated characteristic AD-related changes, including cortical thinning, GM loss, ventricular enlargement, and increased CSF volume. Significant correlations among cortical thickness, tissue volumes, BPV, and BPF further supported their complementary role in characterizing disease-related structural changes. Overall, these findings suggest that MRI-derived measures of cortical thickness, tissue volumes, BPV, and BPF provide useful structural biomarkers of AD and emphasize the importance of considering voxel geometry and segmentation methodology when evaluating neuroimaging biomarkers.

(yellow sweet clover) is a medicinally valuable Fabaceae species with recognized antioxidant and antimicrobial properties, yet the impact of cultivation system on its secondary metabolism remains poorly characterized for Central Asian populations. This study compared wild and in vitro-grown from northern Kazakhstan (Astana city), examining phenolic composition, organic acids, antioxidant capacity, and antimicrobial activity as functions of cultivation system. Hydroethanolic extracts were analyzed by HPLC-DAD-ESI-Orbitrap MS/MS and UFLC-PDA, while antioxidant activity was assessed by TBARS and ABTS assays and antimicrobial activity by broth microdilution against eight bacterial and two fungal strains. Thirteen phenolic compounds were tentatively identified, revealing higher total flavonoid content in in vitro plants, dominated by apigenin di-C-pentoside isomers and vicenin-3, whereas wild plants showed greater diversity of phenolic acids and flavonols, including kaempferol and quercetin glycosides. Despite lower total phenolics, wild extracts more effectively inhibited lipid peroxidation, while ABTS radical scavenging was comparable between sources, and antimicrobial effects were strain-specific. Oxalic acid was approximately three-fold higher in in vitro material, a biochemical feature relevant for downstream use. These findings indicate that cultivation system substantially shapes the secondary metabolite profile and bioactivity of this regionally important medicinal species.

PMID:日期:2026-08-21This study aimed to illustrate the effects of plant-based yogurts made from bio-fermentative green rice milk yogurt (GRMY), enhanced with powder (WGP) and infused with Thai green tea. Different formulations (F1 to F4) were analyzed to assess the impact of varying weight ratios of WGP, ranging from 0% to 15%. The goal was to improve nutritional values and bioactive components. The F4 formulation of plant-based yogurt demonstrated enhanced nutritional values, including higher levels of dietary fiber, starch, protein, and fat contents, which were 42.45, 26.55, 22.48, and 2.29% DW, respectively. It was increased by bioactive compounds such as total phenolic content (52.99 mg GAE/g), total flavonoid content (60.45 mg QUE/g), and β-carotene (1.81 mg/g). Additionally, the F4 formulation exhibited the highest antioxidants in terms of DPPH activity (5.48 mg TROE/g), ABTS activity (424.15 mg TROE/g), and FRAP reducing capacity (106.35 mg TROE/g) and antidiabetic activity of α-glucosidase inhibition in IC at 1.41 mg/mL. The yogurts were evaluated through metabolomic network analysis as a complex bioactive food system, wherein fermentation-derived metabolites synergistically interact with tea polyphenols, amino acids, phytosterols, and carotenoids. The prevailing metabolic profiles indicate an increase in antioxidant capacity, a reduction in inflammatory burden, enhanced glucose metabolism, alterations in gut microbiota, cardiovascular protection, and neuroprotective potential. These findings suggest that the product represents a promising plant-based functional food, offering numerous health benefits. This assertion is supported by a diverse metabolite network identified through UPLC-MS/MS metabolomic research.

4. New Strategies in Tuberculosis Prediction: TB-CAST, an Explainable Baseline Score for Two-Month Sputum Culture Positivity.
PMID:日期:2026-08-21Early identification of patients who will remain sputum-culture-positive at two months could support risk-adapted tuberculosis (TB) monitoring. We developed a compact baseline score from routine admission data measured before anti-TB therapy. The cohort comprised 155 adults (62 culture-positive and 93 culture-negative at two months). Random Forest ranking and correlation clustering were used as exploratory selection steps. The complete development pipeline was evaluated by repeated fully nested cross-validation; a secondary conditional analysis held the full-cohort-selected candidate set fixed. The fully nested pipeline yielded a mean test-fold AUC of 0.70 ± 0.09 (fold-level percentile interval 0.53-0.85). Conditional on full-cohort predictor preselection, mean test-fold AUC was 0.74 ± 0.08, and the patient-level mean repeated out-of-fold score AUC was 0.75 (bootstrap 95% CI 0.67-0.83). At provisional thresholds of ≥2, ≥3 and ≥4 points, conditional procedure sensitivity/specificity were 0.90/0.39, 0.80/0.60 and 0.57/0.74, respectively. The conditional repeated out-of-fold Brier score was 0.200, with calibration intercept approximately 0.00 and slope 0.74. TB-CAST is an exploratory score with moderate internal discrimination. The fully nested result is the primary internal estimate for the complete data-driven pipeline; the conditional results and provisional thresholds require prospective external validation before clinical use.

5. Safety and Efficacy Assessment of Cannabis Plant Compared to Atorvastatin for Lipid Lowering in Diabetic and Obese Wistar Male Rats.
PMID:日期:2026-08-21Statins' established cardiovascular benefits are often undermined by hepatic adverse effects in obese diabetic patients with suspected non-alcoholic fatty liver disease. We evaluated whether cannabis plant treatment is a safe and effective alternative to atorvastatin in a metabolically challenged rodent model. Fourteen obese and diabetic Wistar rats received either atorvastatin for 30 days or cannabis plant extract for 30 days. Main outcome: lipid profile; secondary outcomes: renal function, glycemia and endothelial health assessed through atherogenic indices. Safety was assessed using plasma liver enzyme concentrations. Baseline biochemical profiles were similar between groups. Atorvastatin increased HDL-Col more than cannabis plant (MD: 44.3; IQR [33, 57.5] mg/dL, Hedges' 2.713 vs. MD: 14.3; IQR [14, 17] mg/dL, Hedges' 5.049), and cannabis plant did not change LDL-Col. Atherogenic index of plasma and Castelli Risk Index 1 improved with both interventions, more so with atorvastatin. Liver enzymes increased with atorvastatin (ALAT, MD: 15.9; IQR [12.5, 19.5] U/L; ASAT, MD: 33; IQR [26.5, 38] U/L) but remained virtually unchanged with cannabis. Serum creatinine increased with cannabis plant, with moderate effect size (Hedges' 0.798), but negligibly with statins (Hedges' 0.122). Cannabis plant extract modestly improved the lipid profile and atherogenic indices, with differences often not exceeding the change in control groups. Despite apparent liver safety, the undesired renal and neurological consequences limit applicability in humans. Further studies should prioritize addiction development and renal function.

6. Impact of Red Blood Cell Deformability and Local Hematocrit on Microvascular Flow Resistance: Insights from In Vitro Microfluidic Measurements and Viscosity Modeling.
PMID:日期:2026-08-21Altered red blood cell (RBC) deformability can impact microvascular flow resistance through changes in cell partitioning at bifurcations, and local hematocrit distribution, a key determinant of the effective viscosity of blood. In this work we report on resistance-relevant viscosity distributions extracted from in vitro microfluidic observations of a Y-junction type blood flow. Spatially resolved hematocrit distributions in the parent channel and daughter branches, determined by experimental measurements in earlier work, for healthy and stiffened RBC suspensions over a range of daughter to parent flow-split ratios, Q*, are combined with a recently proposed viscosity model enabling the estimation of the local relative viscosity profiles ηr. The maximum of the RBC elongation index EI, was utilized to define of the deformability parameter λ, which with the local RBC volume fraction, Φ(y), were used as inputs in the relative viscosity, ηr. The latter is a modified Krieger-Dougherty viscosity expression, accounting explicitly for deformability and RBC concentration. The resulting viscosity profiles are averaged in the channel regions of interest to provide an apparent viscosity in the branches, used here as an indicator of microvascular flow resistance. The results illustrate that deformability and hematocrit-dependent RBC enrichment or depletion in the branches of a vascular junction may amplify or attenuate the resistance asymmetry in the daughters of the Y-junction.

7. Integrated Mendelian Randomization and Single-Cell Transcriptomics Reveal T Cell Immune Mechanisms in Systemic Lupus Erythematosus-Bladder Cancer Comorbidity.
PMID:日期:2026-08-21Patients with systemic lupus erythematosus (SLE) exhibit elevated malignancy risk, with increased bladder cancer incidence. This study integrated Mendelian randomization (MR) with single-cell sequencing (scRNA-seq) to nominate exploratory prioritized candidate genes in SLE-bladder cancer comorbidity and their T cell regulatory roles. Single-cell datasets for SLE (GSE266852) and bladder cancer (GSE222315) were retrieved from GEO. Quality control, clustering, and annotation were performed using Seurat. T cell differentially expressed genes were intersected for bidirectional two-sample MR using IEU Open GWAS statistics. Heterogeneity, pleiotropy, and sensitivity analyses assessed robustness. GeneMANIA, miRNA databases, and CTD were used for network and functional analyses. Wilcoxon tests and Monocle 2 were used to characterize expression and T cell differentiation trajectories. Cross-disease intersection nominated 1010 candidate genes. In an exploratory MR screen (uncorrected < 0.05), four candidate genes were nominated (GBP3, LMAN1, SLC40A1, MIS18BP1); none survived FDR correction in both directions. At the uncorrected threshold, LMAN1 showed a shared risk direction (OR > 1) and MIS18BP1 a protective direction (OR < 1). Both showed significant T cell differential expression ( < 0.001) and elevated late differentiation expression. LMAN1 was involved in COPII vesicle transport; MIS18BP1 in CENP-A chromatin assembly. Twenty high-confidence miRNAs targeted each gene. CTD indicated liver injury associations and cisplatin/cyclosporine interactions. LMAN1 and MIS18BP1 are proposed as hypothesis-generating exploratory candidate genes in SLE-bladder cancer comorbidity, potentially involved in immune dysregulation through T cell terminal differentiation modulation. This study provides preliminary evidence suggestive of autoimmune-malignancy comorbidity mechanisms.

8. Flaxseed Influences Basic Health Parameters, Fatty Acid Content Cell Membrane Integrity, Cell Proliferation and Angiogenesis in Fattening Pigs.
8. 亚麻籽对育肥猪基础健康参数、脂肪酸含量、细胞膜完整性、细胞增殖和血管生成的影响PMID:日期:2026-08-21This study focused on dietary supplementation with flaxseed in fattening pigs to observe health benefits and important biological findings, including cell membrane integrity, proliferation, and angiogenesis. Animals received flaxseed for 6 or 3 weeks prior to slaughter. Pigs were divided into a control group (standard diet) and experimental groups F6 (6 weeks) and F3 (3 weeks), which received flaxseed in the feed. Spectroscopy followed by an electrophoretic method was used to study the levels of basic biochemical parameters, including LDH. Subsequently, gas chromatography revealed fatty acid concentrations in the pigs' livers, and finally, immunohistochemical analysis was used to detect CD31 and PCNA antigens in the liver and jejunum. The highest weight gain was measured in the F6 group. After slaughter, blood samples revealed a decreased glucose level in the F6 group, maintained levels of basic biochemical parameters, and decreased concentrations of LDH in blood, heart, liver, and skeletal muscle extracts, thus improving cell membrane integrity. A long-term supplementation period led to better utilization of essential fatty acids, an improved n-6:n-3 ratio, and other atherogenic indices related to cardiovascular health. Flaxseed also affected the expression of CD31 and PCNA proteins in the liver and jejunum, mainly in the F6 group.

9. Environmental Exposure to Endocrine-Disrupting Chemicals and SARS-CoV-2 Infection History Among Blood Donors.
PMID:日期:2026-08-21The present study evaluated the association between plasma concentrations of selected endocrine-disrupting chemicals (EDCs)-bisphenol A (BPA), methylparaben (MeP), propylparaben (PrP), 4-nonylphenol (4-NP), 4-octylphenol (4-OP), bis(2-ethylhexyl) phthalate (DEHP), and imazalil (IMZ)-and prior coronavirus disease 2019 (COVID-19) status. Plasma EDC levels were quantified via gas chromatography-mass spectrometry in 262 voluntary blood donors (121 convalescents and 141 controls) at the Regional Centre for Transfusion Medicine in Bialystok, Poland. The cohort exhibited widespread environmental exposure to all selected EDCs (BPA: 1.14 ng/mL, MeP: 1.20 ng/mL, PrP: 1.38 ng/mL, 4-NP: 1.90 ng/mL, 4-OP: 0.57 ng/mL, DEHP: 49.68 ng/mL, IMZ: 20.06 ng/mL). Convalescents had significantly higher concentrations of MeP, PrP, 4-OP, and 4-NP. Logistic regression revealed that MeP, PrP, and 4-NP were significantly associated with prior COVID-19 status, increasing infection odds by 80%, 70%, and 63%, respectively. While 4-OP was strongly associated with prior COVID-19 status (odds ratio > 300 per unit change and 1.79 per 0.1 unit change), the wide confidence interval for the unit change estimate requires cautious interpretation. These findings suggest a strong link between selected paraben and alkylphenol exposure and prior COVID-19 status in blood donors. Additionally, the high IMZ levels highlight the need for urgent evaluation of its safety and environmental prevalence.

10. Atrial Fibrillation Burden: Assessment, Clinical Significance, and Therapeutic Implications.
PMID:日期:2026-08-21Atrial fibrillation (AF) is traditionally classified using categorical clinical patterns and binary recurrence endpoints. However, these measures do not fully reflect arrhythmia frequency, duration, temporal distribution, or progression. AF burden, defined as the proportion of monitored time spent in AF, provides a more granular measure of disease activity and therapeutic response. Recent advances in cardiac implantable electronic devices, insertable cardiac monitors, wearable technologies, and digital platforms have significantly improved burden assessment, although considerable heterogeneity remains in monitoring methods, definitions, and clinically relevant thresholds. This review examines contemporary approaches to AF burden measurement and its associations with thromboembolic risk, heart failure, hospitalization, symptoms, quality of life, disease progression, and mortality. We particularly discuss the reduction of AF burden as a potential therapeutic target following antiarrhythmic drug therapy and catheter ablation, and the limitations of conventional recurrence definitions based on episodes lasting more than 30 s. This review also focuses on the implications for rhythm-control selection, post-ablation monitoring, anticoagulation, and clinical-trial design, while proposing priorities for standardized burden-based assessment.
