1. Cyclosporin A triggers a RIG-I-mediated interferon response through multiple cellular processes.
PMID:日期:2026-09-16The development of broad-spectrum antiviral agents is essential for preparedness against emerging viruses with pandemic potential. Cyclosporin A (CsA), a clinically approved immunosuppressant, has been shown to exert antiviral activity against a wide range of viruses. Recent evidence suggests that this effect is partially mediated through the induction of interferon lambda (IFN-λ), which in turn stimulates a set of interferon-stimulated genes (ISGs) that establish an antiviral cellular state. However, the molecular mechanisms underlying IFN-λ induction by CsA have remained poorly understood. Here, we demonstrate that CsA triggers a retinoic acid-inducible gene I (RIG-I)-dependent activation of the IFN-λ/ISG signaling axis. This is accompanied by the upregulation of mitochondrial double-stranded RNA (dsRNA). Importantly, we show that CsA causes pronounced alterations in the mitochondrial cristae network. CsA also impairs autophagy, as evidenced by reduced autophagosomal membrane formation and increased levels of the autophagy marker p62. Supporting a functional link, treatment with the autophagy inducer Torin-1 reverses CsA-induced dsRNA accumulation, IFN-λ expression and ISG transcription. Together, our data indicate that the antiviral activity of CsA is partially driven by converging cellular processes involving mitochondrial remodeling, dsRNA accumulation, and impaired autophagy, which collectively activate the RIG-I/IFN-λ/ISG axis, offering mechanistic insights with therapeutic implications.

2. Deterministic and ANN-based modelling of measles transmission using real epidemiological data.
PMID:日期:2026-09-16This study aims to develop a nonlinear SVLIQR epidemic model to investigate measles transmission dynamics by incorporating vaccination, latency, quarantine, and treatment effects. The qualitative properties of the model, including positivity and boundedness of solutions, are established. The basic reproduction number is derived using the next-generation matrix method, and the existence of disease-free and endemic equilibrium points is examined. The local stability of the disease-free equilibrium is analysed using the Routh-Hurwitz criterion, while global stability is studied through the Castillo-Chávez approach. In addition, model parameters are estimated using the least squares method based on reported measles cases in China from 2005 to 2017. To improve the numerical approximation and capture the nonlinear dynamics of the system, an ANN-based solver coupled with the ode45 scheme is implemented. The results show that the model fits the reported data well, and the ANN framework provides highly accurate and convergent approximations under different epidemiological parameter settings. Numerical simulations further indicate that higher vaccination and quarantine rates significantly reduce measles transmission. Overall, the proposed framework provides a useful and reliable tool for understanding measles dynamics and assessing effective intervention strategies.

3. Searching for separation between frequency-modulated calls of blue and fin whales.
PMID:日期:2026-09-15Reliable differentiation of similar acoustic signals remains a major challenge for passive acoustic monitoring (PAM) of sympatric marine mammal species. Blue (Balaenoptera musculus) and fin whales (Balaenoptera physalus) produce frequency-modulated (FM) downsweeping calls with overlapping frequency ranges and durations, making species attribution difficult. This study combines long-term PAM datasets with animal-borne tag recordings to investigate how these FM calls can be reliably distinguished by species. Tag recordings with confirmed species identity showed substantial overlap in most acoustic features, but the parameter Duration 90% offered partial separation: with most fin whale FM calls < 0.6 s, and blue whale FM calls > 0.7 s. Applied to high-quality calls from moored data with signal-to-noise ratios above 12 dB, this approach may provide a conservative, rule-based means of classification. A more robust approach was achieved by combining deep learning feature extraction with non-linear dimensionality reduction. Integrating confirmed ground-truth tag recordings as a reference space enabled previously unseen FM calls from long-term PAM recordings to be attributed by acoustic similarity to known calls while maintaining an explicit measure of uncertainty by leaving acoustically ambiguous calls unassigned. Together, these approaches provide a practical framework for more reliable call attribution, enhancing long-term PAM-based monitoring of blue and fin whales.

PMID:日期:2026-09-15Plant H⁺-ATPases are activated through tightly regulated hexameric assembly, relieving autoinhibition via displacement of a regulatory domain during the oligomerization process. Sub-hexameric assembly states also exist but their role in activity regulation remains elusive. To characterize sub-hexameric plant H⁺-ATPases, we isolated homo-oligomers of the H⁺-ATPases AHA2 and determined their sizes with size exclusion chromatography multi-angle light scattering. AHA2 predominantly presented as a monomer but also assembled into stable dimers and larger complexes. Activity assays on solubilized and liposome-reconstituted AHA2 monomers and dimers revealed that dimerization did not affect enzymatic activity, suggesting that AHA2 dimerization does not represent a separate regulatory pathway. Instead, AHA2 dimers may serve as reservoirs for rapid hexamer assembly, facilitating the dynamic regulation of protein activity.

5. Cold-induced vasomotion changes in healthy individuals and patients after median or ulnar nerve repair.
PMID:日期:2026-09-15This exploratory study investigated digital vasomotion during and after a standardized cold stress test in healthy subjects and patients previously treated for median or ulnar nerve injury. Twenty-five healthy individuals and 20 patients with a median or ulnar nerve injury were included. Finger perfusion was measured before and after a Cold Stress Test, using Laser Speckle Contrast Imaging. Vasomotion activity was quantified as the area under the curve of synchronized perfusion dips, and comparisons were made within and between the groups. Synchronized perfusion dips were observed in healthy participants both before and after cold provocation but were attenuated in patients with nerve injury. This suggests an impaired neural modulation of digital microvascular tone among nerve-injured patients. Among male patients with ulnar nerve injury, baseline perfusion was lower in the injured digit V than in the corresponding uninjured digit V (132 vs. 157 PU, p = 0.001), accompanied by reduced vasomotion activity (AUC 1.07 vs. 1.92, p = 0.010). In conclusion, neural mechanisms may be important in regulating vasomotion in the hand. Patients with a nerve trunk injury exhibit a different vasomotion pattern compared with healthy participants, supporting an important role of neural regulation in the alteration of digital vasomotion.

6. Integrated expression profiling reveals molecular crosstalk between CFTR, FASN, FRAP (mTOR), and BANK1 in hepatocellular carcinoma.
PMID:日期:2026-09-15Hepatocellular carcinoma (HCC) is a common malignancy characterised by molecular heterogeneity and poor prognosis. The purpose of this study was to determine differences in the expression of Cystic Fibrosis Transmembrane Conductance Regulator (CFTR), Fatty Acid Synthase (FASN), FKBP12-rapamycin-associated protein (FRAP; mTOR), and B-cell scaffold protein with ankyrin repeats 1 (BANK1) between HCC patients and controls. A total of 200 blood samples (100 from HCC patients and 100 from controls) were analysed using Reverse Transcription Polymerase Chain Reaction (RT-PCR), with Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) used as an internal control. The results demonstrated significant upregulation of FASN and FRAP (mTOR) (p < 0.0001) and downregulation of CFTR and BANK1 (p < 0.0001), suggesting potential oncogenic and tumour-suppressive roles, respectively. Gene expression levels also differed significantly across Stages I, II, III, and IV. The Ct values for BANK1 were higher than those for CFTR, FASN, and FRAP (mTOR). Because the expression measurements for the four genes were obtained from blood rather than tumour parenchyma, these findings were not interpreted as direct evidence of gene expression within tumour tissue. Survival analysis using a public dataset revealed that dysregulation of these genes was associated with lower survival rates. In silico pathway analysis demonstrated complex interactions among the four genes, linking them to metabolic regulation, immune signalling, and cellular proliferation pathways. Overall, the integrated network involving CFTR, FASN, FRAP (mTOR), and BANK1 may have potential as a novel prognostic marker by reflecting combined metabolic and immune pathway activity. A combined four-gene logistic regression model demonstrated promising discriminative performance in the present cohort (AUC = 0.89). However, this finding requires validation in independent cohorts before any diagnostic application can be considered.

7. Low-cost IoT real-time environmental monitoring and one-hour LSTM forecasting for small-scale agricultural applications.
PMID:日期:2026-09-15Standard equipment is too expensive and complex, frequently limiting continuous environmental monitoring in small-scale farming. To overcome this, a low-cost seven-channel IoT monitoring and forecasting system was developed and deployed, utilizing an Arduino Uno R3 and ESP8266 NodeMCU Wi-Fi module. The system acquired temperature, humidity, precipitation, soil moisture, methane, carbon monoxide, and air quality, transmitting data to the ThingSpeak cloud platform at an average interval of approximately one minute. The DHT22 temperature and humidity sensor carries a manufacturer-rated accuracy of ± 0.5 °C and ± 2-5% RH; the remaining sensors report uncalibrated raw output, as no reference-instrument validation was performed. The device was deployed for 21 days at a residential garden site in the Nile Delta region of Egypt. A total of 30,981 of 31,460 exported observations, 98.48% of the exported record were retained after the application of the preprocessing pipeline, which consisted of time-aware interpolation and Augmented Dickey-Fuller (ADF)-guided stationarity conditioning. A 70/15/15 chronological train/validation/test split was used to prevent temporal leakage. The analysis revealed distinct environmental patterns, including a rising trend of 0.296 °C/day, an anti-phase coupling between temperature and humidity, and an increase in raw gas sensor output overnight. To make use of this data for predictive insights, seven different univariate LSTM models were trained to forecast one hour ahead. When evaluated against persistence and seasonal-naive baselines on the same test set, the LSTM held a modest edge over persistence for methane (Mean Absolute Percentage Error, MAPE: 0.81%) and air quality (MAPE: 0.31%), while a simple persistence forecast matched or outperformed the LSTM for the remaining five channels. This field deployment demonstrates the technical feasibility of pairing affordable hardware with deep learning forecasting for continuous environmental monitoring, providing a foundation for future irrigation-support applications in resource-constrained settings.

8. Emotion regulation strategy use and daily levels of affective arousal, affective valence and dissociation in a clinical sample.
PMID:日期:2026-09-15Clinical models and cross-sectional research suggest that habitual use of disengagement-oriented emotion regulation strategies is associated with high levels of affective arousal, negative valence, and dissociation, whereas engagement-oriented strategies show the opposite effect. However, intensive longitudinal evidence remains scarce. This study examined whether habitual use of 10 emotion regulation strategies, assessed using the Process Model of Emotion Regulation Questionnaire (PMERQ), predicts affective arousal and valence, and dissociation in daily life. We analysed ecological momentary assessment data from 88 individuals with posttraumatic stress disorder, borderline personality disorder, and dissociative disorders. Baseline PMERQ scores did not robustly predict daily levels of affective arousal and valence, and dissociation. The overall null findings of our study could reflect either a true absence of meaningful effects, insufficient sample size, or a mismatch between trait-level emotion regulation measures and momentary affect/dissociation. Future studies should prioritise momentary assessments of emotion regulation to disentangle state and trait effects.

9. Diverse ossification pathways in the lower jaw of Sclerocephalus (Lower Permian) as revealed by the first record of chondroid bone in Amphibia.
PMID:日期:2026-09-15Dermal bones are typically thought to form directly from mesenchyme, however some can develop via a transient tissue called chondroid bone, which are well documented among fishes and amniotes. Here, we report the first occurrence of chondroid bone among non-amniote tetrapods and the stratigraphically oldest record of this tissue among vertebrates, in the temnospondyl Sclerocephalus nobilis from the Lower Permian of Germany. Histological analysis of its mandible reveals a heterogeneity of developmental pathways: the angular forms through direct intramembranous ossification, the articular ossifies endochondrally from Meckelian cartilage, and the dermal surangular uniquely contains chondroid bone, with chondrocyte-like cells embedded in the matrix composed of interwoven loop complexes. This tissue likely represents an early developmental stage, promoting rapid expansion of the bone surface while potentially providing mechanical support in regions experiencing high stress during feeding. The angular exhibits a diploë structure and is composed of lamellar and parallel-fibred bone, with remnants of avascular faint parallel-fibred bone representing the larval stage. The articular represents the only endochondral bone and consists of an early-formed periosteal bone that is parallel-fibred in nature, with prominent Sharpey's fibers and a secondary trabecular endosteal region. The discovery of chondroid bone in Sclerocephalus expands the known diversity of skeletal tissues among early tetrapods and suggests that transitional bone types may have been more widespread than previously recognized, but are likely underreported due to preservational biases and limited sampling.

10. Amorphous pyrano[3,2-c]quinoline-semicarbazone thin films switch from uniform to heterogeneous morphology and optical modulation upon copper coordination.
PMID:日期:2026-09-15Thermally evaporated pyrano[3,2-c]quinoline semicarbazone (PQMHC) and its Cu(II) complex thin films were systematically investigated to elucidate the effect of metal coordination on their structural, morphological, photoluminescence, and optical properties. X-ray diffraction (XRD) revealed broad halos, confirming the amorphous nature of both films. Field-emission scanning electron microscopy (FE-SEM) showed that the PQMHC film possesses a relatively uniform surface composed of closely packed granular features, whereas Cu(II) incorporation induces a heterogeneous morphology with irregular clusters and elongated structures. Photoluminescence measurements revealed broad visible emission spanning 380-650 nm, with a pronounced red shift toward 520-560 nm upon Cu(II) complexation. Optical measurements showed reduced transmittance and enhanced reflectance following Cu(II) incorporation, accompanied by a decrease in the optical band gap from 2.87 ± 0.02 to 2.57 ± 0.01 eV. Furthermore, Miller's empirical rule was employed to estimate the third-order nonlinear susceptibility and nonlinear refractive index, both of which decreased upon Cu(II) coordination, indicating reduced calculated polarizability. Overall, Cu(II) coordination provides an effective means of tuning the structural, morphological, photoluminescence, and optical characteristics of PQMHC thin films, suggesting their potential for tunable optical and optoelectronic applications.
