1. Spatial relationships between mosquito breeding sites and malaria cases in rural Uganda: Implications for larval source management.
PMID:日期:2026-09-24Larval source management (LSM) has been a core strategy of malaria control but is difficult to implement in holoendemic regions of rural sub-Saharan Africa. More targeted approaches that reduce the burden of effort, but maintain impact are needed. The prospective cohort study examined the spatial relationship between mosquito breeding sites and malaria cases to guide targeted LSM. After enrolling households across three villages, we recorded malaria cases from neighboring health facilities and conducted larval and adult mosquito surveillance. Breeding site density was assessed within different search radii from malaria cases using GIS. Households were grouped by environmental similarity using Factor Analysis of Mixed Data (FAMD) and k-means clustering, and clusters were compared in terms of malaria incidence and mosquito abundance. We identified and surveyed 69 breeding sites (March-May 2023, 90% larva-positive) in the study area and documented 56 clinical malaria cases among 366 households (April-June 2023). Malaria incidence was higher within 200 m of breeding sites than in households without (0.20 vs 0.09 cases per household; p = 0.005). When searching outward from cases, the number of covered breeding sites increased rapidly, reaching 60% within 200 m and 75% within 300 m, before plateauing. Malaria risk and mosquito abundance varied across clusters, particularly near the hydroelectric station spillway and in areas with different house construction. The findings support the potential of reactive LSM by targeting breeding sites near malaria cases. Further research is needed to assess its effectiveness and its added value in malaria control programs.

2. Brief exposure to hyperoxia is associated with reduced Plasmodium falciparum in vitro tolerance to Dihydroartemisinin.
PMID:日期:2026-09-24The ring-stage survival assay (RSA) is a valuable tool for assessing in vitro Plasmodium falciparum tolerance to Dihydroartemisinin. However, laboratories with unreliable or no access to premixed commercial gas may not perform the RSA under the recommended physiological normoxia. We measured survival rates of ART-sensitive and ART-resistant strains in the normoxic commercial gas and hyperoxic candle jar gas. RSA read-outs for ART-resistant strains were significantly lower (p<0.05) under hyperoxia compared to physiological normoxia, whereas those of ART-sensitive strains were unaltered. These findings have important implications for interlaboratory comparison of RSA data and understanding the mechanisms of resistance to endoperoxide drugs.

3. The Effect of Pertussis Vaccination During Pregnancy on Pertussis and Diphtheria Immunity in Children at 3 and 4 Years of Age.
PMID:日期:2026-09-23Pertussis vaccination during pregnancy protects young infants but blunts their antibody responses to primary immunization; the duration of this effect beyond infancy is unknown. In a clinical trial, we followed children born to tetanus-diphtheria-acellular pertussis-vaccinated (n=47) and unvaccinated (n=22) mothers to 4 years of age. At both 3 and 4 years, children born to Tdap-vaccinated mothers had lower anti-pertussis toxin and anti-diphtheria toxin IgG concentrations than control children, and pertussis toxin seropositivity and diphtheria seroprotection rates were low in both groups. After a preschool acellular pertussis booster at 4 years, antibody concentrations increased to comparable levels in both groups.

4. Rationale for a live-attenuated vaccine for cryptosporidiosis.
PMID:日期:2026-09-23Natural Cryptosporidium infection causes development of protective immunity. Live-attenuated vaccines stimulate both adaptive and "trained" innate immune responses. With advances in parasite genetics, target identification, and in vitro cultivation, we now propose development of a live-attenuated vaccine for cryptosporidiosis.

5. The Phosphate-Specific Transport System Gene pstA1 Contributes to Rifampin Tolerance in Mycobacterium tuberculosis.
PMID:日期:2026-09-23Tuberculosis (TB) caused an estimated 10.7 million new cases and 1.23 million deaths in 2024. Antibiotic tolerance, the ability of bacteria to survive bactericidal antibiotics without genetic resistance mutations, contributes to prolonged treatment. Targeting tolerance mechanisms could promote accelerated clearance of Mycobacterium tuberculosis (Mtb). Our previous genetic screen identified pstA1, involved in phosphate-specific import. PstA1 has previously been implicated in Mtb virulence, as mutants lacking this gene exhibit defective survival during phosphate limitation in vitro, in macrophages, and in immunocompetent mice. We evaluated rifampin susceptibility of a pstA1 deletion mutant (ΔpstA1) using minimum inhibitory concentration (MIC) and time-kill assays, assessed survival during phosphate limitation and in macrophages, and performed differential expression analysis. Rifampin MIC was not altered in ΔpstA1, suggesting that the difference in susceptibility was not due to antibiotic resistance. Time-kill assays revealed a shift in the mean duration of killing (2-log reduction) from 1.6 days in wild-type to 1.0 day in ΔpstA1, and complementation partially restored this phenotype. pstA1 was specifically required for Mtb survival in the absence of exogenous inorganic phosphate and was important for growth adaptation in culture without detergent and within macrophages in an interferon-γ-dependent manner. ΔpstA1 exhibited distinct transcriptional reprogramming with 58 differentially expressed genes, including altered metabolic, DNA damage repair, and secretory pathways. These findings identify PstA1 as a contributor to rifampin tolerance and suggest that phosphate transport pathways influence antibiotic susceptibility in Mtb.

6. The Power of Storytelling in Science: Perspectives from Two Students and a Community Member.

Through our experiences with The Last Gift Program, we reflect on how engaging with participants and their stories transformed our understanding of science, compassion, and responsibility. These experiences demonstrate how storytelling can foster empathy, strengthen scientific purpose, challenge stigma, and inspire the next generation of researchers to pursue science grounded in humanity. Scientific discovery may preserve samples and data, but storytelling helps preserve the people, lives, and acts of generosity behind them.

8. Chlamydia trachomatis Incidence in Relation to Vaginal Microbiota Dynamics, Immunogenetics and Exposures in a Cohort of Young Student Women in France.
PMID:日期:2026-09-23Given the potential role of the vaginal microbiota in the acquisition of Chlamydia trachomatis infections, we aim to investigate its contribution together with immunogenetics and epidemiological exposures to the incidence of C. trachomatis in young women. This study involved 313 female students aged 18-24 years from the i-Predict prevention trial in France. Participants provided 4 self-collected vaginal samples and filled 4 self-administered questionnaires every 6 months for 18 months. C. trachomatis-positive participants and negative controls with complete follow-up were selected for this analysis and submitted to chlamydia testing and to vaginal microbiota characterization using 16S rRNA amplicon sequencing. Thirteen human single nucleotide polymorphisms (SNPs) related to C. trachomatis susceptibility and severity were also assessed. Compared with 260 noninfected participants, Gardnerella spp., Fannyhessea vaginae and Prevotella timonensis were more abundant in C. trachomatis-incident participants (n = 24) before infection. Having a CST IV at the preceding sample compared with a CST I (OR 3.77, 95%CI [1.12-12.69], P = .033) was associated with increased risk of C. trachomatis acquisition, as well as having had multiple concomitant partners in the last 6 months (5.40 [2.10-13.92], P < .001). Lifetime condom use was associated with decreased incidence (0.38 [0.15-0.95], P = .039). None of the tested human SNPs was associated with C. trachomatis infection. In this low-risk for C. trachomatis population, having a CST IV vaginal microbiota and associated bacterial anaerobes was a risk factor for C. trachomatis acquisition after adjustment for other exposures. 9. Low-Level Viremia and Spatial Heterogeneity Among Individuals on Dolutegravir-Based Antiretroviral Therapy in Botswana: A Nationwide Study (2016-2022).
PMID:日期:2026-09-23The study uses spatial analysis to identify factors associated with human immunodeficiency virus (HIV) detectable viremia in dolutegravir (DTG)-based first-line antiretroviral therapy (ART), which is crucial for targeted interventions to sustain epidemic control in Botswana. This was a longitudinal observational cohort utilizing a dataset of 42 546 people with HIV on DTG-based first-line ART from June 2016 to December 2022 enrolled in the Botswana National Treatment Program. Detectable viremia was defined as viral load (VL) >50 copies/mL and further classified into low-level viremia (LLV; VL from 51 to 999 copies/mL) and virologic failure (VF; VL ≥1000 copies/mL). The rates of detectable viremia, LLV, and VF were stratified by year from 2017 to 2022 and mapped by districts in Botswana. Generalized estimating equation (GEE) models were utilized to evaluate odds of detectable viremia. There was an overall trend of increasing detectable viremia, with rates of 6.7% (2017), 5.6% (2018), 4.7% (2019), 7.1% (2020), 13.5% (2021), and 11.2% (2022) (P < .001). The linear annual trend showed that each additional year from 2017 was associated with 23% increased odds of detectable viremia and 50% increase in the odds of LLV, while odds of VF decreased by 6% per year. Repeated-measures GEE models showed that male sex (adjusted odds ratio [aOR], 1.36 [95% confidence interval {CI}, 1.30-1.43]) and rural residence (aOR, 1.10 [95% CI, 1.05-1.16]) were consistently associated with higher odds of detectable viremia, LLV, and VF over follow-up. While VF is declining, LLV is increasing. We observed spatial heterogeneity in detectable viremia, and higher odds in males, those with rural residence, and young individuals (18-35 years). Targeted interventions are necessary for high-risk groups and regions to further suppress viremia and achieve sustained epidemic control beyond the UNAIDS 95-95-95 targets.
