1. Seasonality, mass vaccination and critical policy evaluation on global exit strategy of COVID-19 crisis.
PMID:日期:2023-03-05There is a strong coordinated effort by vaccination groups all over the world to put an end to the current crisis of COVID-19. Now sufficient data are available to analyse and compare some results to explore the aftereffects of vaccination. Some influence variables on transmissions of the disease were discussed e.g., mass vaccination, lockdown and seasonality. Most studies covered here are up to the beginning of July 2022, while some analyses focused on the earlier period of mass vaccination. Well established, simple statistical techniques to evaluate results were presented those used open data sources of authoritative bodies. Some comparisons between vaccinated vs. unvaccinated were also discussed based on data from UK Government Health Security Agency (UHSA). In terms of mass vaccination, adverse reactions after vaccination received attention, as health and safety issues of the general public are of prime importance. Apart from direct side effects, the secondary effect of mass vaccination needs attention too. After the initiation of the vaccination programme, almost all countries experienced a sudden surge in transmission and most countries had to impose strict lockdown measures. Many countries, with a low prevalence of disease, suddenly showed a steep jump and some countries even followed a synchronized pattern between the rate of transmissions and the variation of vaccine doses. Time series analyses and bar diagram presentations were able to capture those features. In that context, fast mutation of the virus and new variants after mass vaccination and possible mechanisms/consequences were also attended. To understand the effect of seasonality, similarities between COVID-19 and the seasonal Flu are discussed for Europe and US to gain useful insight. Using time series analyses and spatial plots of regional temperature composites we showed, like Flu, seasonality played a dominant role in transmissions of COVID-19 in the Europe. Regulations of vaccine dose and policy implication were explored too. From 22 December 2021, global vaccine doses were reduced substantially, which followed a dramatic reduction in cases and thereafter deaths with around one month's lag between each. As strong dependency on seasonality is noticed in certain countries and observing that regulation of vaccine doses has roles in modulating the transmission with certain lags, globally as well as regionally, our results have policy implications for the management of COVID. Debating, questioning and criticism are always the foundation of great science and the major pillars of its progress. Following that objective, it is an effort to explore pragmatically, supported by scientific analyses, areas relating to the effectiveness of the COVID-19 vaccine and the exit strategy via the pathway of vaccination.

2. In silico evaluation of potential intervention against SARS-CoV-2 RNA-dependent RNA polymerase.
PMID:日期:2022-12-15With few available effective interventions, emergence of novel mutants responding poorly to existing vaccines and ever swelling newer waves of infection, SARS-CoV-2 is posing difficult challenges to mankind. This mandates development of newer and effective therapeutics to prevent loss of life and contain the spread of this deadly virus. Nsp12 or RNA-dependent RNA polymerase (RdRp) is a suitable druggable target as it plays a central role in viral replication. Catalytically important conserved amino acid residues of RdRp were delineated through a comprehensive literature search and multiple sequence alignments. PDB ID 7BV2 was used to create binding pockets using SeeSAR and to generate docked poses of the FDA approved drugs on the receptor and estimating their binding affinity and other properties. approach used in this study assisted in prediction of several potential RdRp inhibitors; and re-validation of the already reported ones. Five molecules namely Inosine, Ribavirin, 2-Deoxy-2-Fluoro-D-glucose, Guaifenesin, and Lamivudine were shortlisted which exhibited reasonable binding affinities, with neither torsional nor intermolecular or intramolecular clashes. This study aimed to widen the prospect of interventions against the SARS-CoV-2 RdRp. Our results also re-validate already reported molecules like 2-Deoxy-D-glucose as a similar molecule 2-deoxy-2-fluoro-D-glucose is picked up in this study. Additionally, ribavirin and lamivudine, already known antivirals with polymerase inhibition activity are also picked up as the top leads. Selected potent inhibitors of RdRp hold promise to cater for any future coronavirus-outbreak subject to and validations.

3. Combating COVID-19 crisis and exploring heat-based simple solutions.
PMID:日期:2022-11-30Covid-19 pandemic affected whole of the world taking many lives and impacting the economy and mental health severely. Exit pathways via vaccination though ignited optimism initially but attenuated by the emergence of several new variants which are less sensitive to vaccines. Considering emergency situations, some urgent, simple heat-based solutions for the initial stages of the disease were also proposed at the beginning of pandemic and further elaborated here. Solutions were proposed based on science as follows: exploring results of statistical analyses on the global transmission of COVID-19; observed temperature-dependent behaviours of similar category viruses; temperature-based clinical trial experiments with similar category viruses; successful clinical trial experiments with heat-based intervention for COVID-19 patients; and finally, biological mechanism/response in human bodies to heat-based solution for COVID-19 from medical doctor's perspective. Solutions proposed are practically without side effects, can be even practised in own home and there is no vested interest involved.

4. Modeling COVID-19 infection in high-risk settings and low-risk settings.
PMID:日期:2022-11-02In this research paper we present a mathematical model for COVID-19 in high-risk settings and low-risk settings which might be infection dynamics between hotspots and less risky communities. The main idea was to couple the SIR model with alternating risk levels from the two different settings high and low-risk settings. Therefore, building from this model we partition the infected class into two categories, the symptomatic and the asymptomatic. Using this approach we simulated COVID-19 dynamics in low and high-risk settings with auto-switching risk settings. Again, the model was analyzed using both analytic methods and numerical methods. The results of this study suggest that switching risk levels in different settings plays a pivotal role in COVID-19 progression dynamics. Hence, population reaction time to adhere to preventative measures and interventions ought to be implemented with flash speed targeting first the high-risk setting while containing the dynamics in low-risk settings.

5. Advances on concrete strength properties after adding polypropylene fibers from health personal protective equipment (PPE) of COVID-19: Implication on waste management and sustainable environment.
PMID:日期:2022-10-11Using Health personal protective equipment (PPE) such as face masks, safety foot shoes and protective suits has expanded dramatically due to COVID-19 pandemic leading to a widespread distribution of the PPE, particularly the face masks, in the environments including streets, dump sites, seashores and other risky locations. The environmental degradation of polypropylene, the essential plastic component in single-use face masks (SUM), takes between 20 and 30 years and thus it is essential to develop experimental approaches to recycle the polypropylene or to reuse it in different ways. This paper explores the integration of SUM into concrete structures to improve its mechanical properties. We first to cut the inner nose wire and ear loops, then distribute the PPE material among five different mixed styles. The PPE were applied by volume at 0%, 1%, 1.5%, 2.0%, and 2.5%, with tests focusing on UCS, STS, FS, and PV to determine the concrete's overall consistency and assess the improvement in its mechanical properties. The results showed that adding PPE improves the strength properties and general performance of the concrete specimens. The pattern of rising intensity started to fade after 2%. The findings demonstrated that adding PPE fibers enhanced the UCS by 9.4% at the optimum 2% PPE. The PPE fibers, on the other side, are crucial in calculating the STS and FS of the reinforcement concrete.

6. Medicinal plants used for the management of respiratory diseases in Zimbabwe: Review and perspectives potential management of COVID-19.
PMID:日期:2022-09-21Respiratory diseases have in the recent past become a health concern globally. More than 523 million cases of coronavirus disease (COVID19), a recent respiratory diseases have been reported, leaving more than 6 million deaths worldwide since the start of the pandemic. In Zimbabwe, respiratory infections have largely been managed using traditional (herbal) medicines, due to their low cost and ease of accessibility. This review highlights the plants' toxicological and pharmacological evaluation studies explored. It seeks to document plants that have been traditionally used in Zimbabwe to treat respiratory ailments within and beyond the past four decades. Extensive literature review based on published papers and abstracts retrieved from the online bibliographic databases, books, book chapters, scientific reports and theses available at Universities in Zimbabwe, were used in this study. From the study, there were at least 58 plant families comprising 160 medicinal plants widely distributed throughout the country. The Fabaceae family had the highest number of medicinal plant species, with a total of 21 species. A total of 12 respiratory ailments were reportedly treatable using the identified plants. From a total of 160 plants, colds were reportedly treatable with 56, pneumonia 53, coughs 34, chest pain and related conditions 29, asthma 25, tuberculosis and spots in lungs 22, unspecified respiratory conditions 20, influenza 13, bronchial problems 12, dyspnoea 7, sore throat and infections 5 and sinus clearing 1 plant. The study identified potential medicinal plants that can be utilised in future to manage respiratory infections.

7. The City of Harare's response to COVID-19: A case for international co-operation and solidarity.
PMID:日期:2022-08-28This article uses the case of the City of Harare to offer insights into how the coronavirus pandemic shaped paradiplomacy in Zimbabwe. It argues that the City of Harare's international partnerships played a nominal role in helping its response to COVID-19. There is strong evidence that the coronavirus pandemic undermined the significance of international co-operation and solidarity by African subnational governments in pursuit of their development and service delivery mandates. Better leveraging of the City of Harare's international partnerships had the potential to transform many of the challenges it faced in fighting the pandemic. The absence of a robust international dimension in the city's handling of the COVID-19 pandemic was one of the main factors that resulted in most of its responses being ineffective. The article concludes that the City of Harare's experience offers important lessons to, among others, African governments on the need to integrate decentralised responses and city-to-city co-operation into their future national disaster and economic response, recovery and resilience strategies.

8. Forgotten frontline workers in higher education: Aiding Ghana in the COVID-19 recovery process.
PMID:日期:2022-07-31The sudden emergence of the COVID-19 triggered a chain of events in the global education system; suspended onsite instruction; migration to online learning; adoption of mobile technologies for mobile learning, and diverse technological innovations. All this was done with the objective of adhering to COVID-19 lockdown protocols to speed global recovery from the disruptive effect of the COVID-19 crisis. In many countries, online learning became the only legal means to achieve lifelong and progressive education for over 91% of the world's student population whose education was at risk. Stakeholders in higher education (parents, students, teachers, and administrators) across the globe also became "frontline workers" in a collective effort to combat the spread of the virus. The study explores the integral role of these frontline workers in curbing the virus. The researchers draw from qualitative interviews involving twenty (20) tertiary students in Ghana who experienced a physical resumption of school in 2021. Findings suggest that a policy shift by school leaders led to the adoption of a multi-track year-round education (MT-YRE) system to promote social distancing. Students were provided with personal protective equipment (PPEs), teachers educated students on COVID-19 prevention and fostered good relationships with their students. Students adhered to institutional protocols to study. Parents also provided psychological and financial support. Policymakers in education should provide clear guidelines, resources, funds, and recognition to school stakeholders as they collectively tackle the virus to ensure economic, health, and education recovery post-COVID-19. Future studies should focus on how to establish a crisis-management framework for higher education.

9. COVID-19, the environment and animal life in Malawi compared to other countries: A brief scooping review for a research agenda in the developing countries.
PMID:日期:2022-07-07The impact of COVID-19 on the human population in Malawi has been documented. However, its impact on the animal population and the environment has not been thoroughly researched. Because of the well-known inter-relationship between human and animal populations and the environment, a study based on a brief scooping review of previous related studies, media and survey reports, was conducted. The findings reveal that except for a few selected studies, the research gap on COVID-19's impact on the environment and animals in Malawi is wide compared to other countries. Nonetheless, from the few identified related studies, this study has revealed that as the restriction of movement and closure of borders disrupted the supply chain of forest resources in the country, the COVID-19 pandemic has led to increased pressure on forests as a coping strategy due to significant loss of jobs in the informal sector. Although the quality of water and air improved in most parts of the globe due to reduced human activity, there is no substantial literature on the same in Malawi partly due to ineffective monitoring systems. However, COVID-19 has exposed the deficiencies in water security in Malawi, thereby creating opportunities to address them. Conversely, increased demand for water at household levels due to restricted movements contributed to environmental pollution at suburb levels. In particular, the less developed and overpopulated countries suffered from land pollution due to poor disposal of plastic generated from hospitals and personal protection equipment. Elsewhere, studies show that minimal human interference with animals outside homes resulted in an increase of fish and bird biomasses. But, unemployment rates caused by the pandemic have seriously contributed to illegal poaching in developing countries. Therefore, a rapid assessment of the impact of the pandemic on environment in Malawi, to generate the evidence needed for policy makers to use in support of the affected and also plan for the recovery and sustainability of wildlife, is recommended.

10. In-silico screening to delineate novel antagonists to SARS-CoV-2 nucleocapsid protein.
PMID:日期:2022-06-21Since its inception, SARS-CoV-2 has crossed all borders and continues rampaging around the globe, causing profound economic damage and heavy burden on the scientific community and the healthcare fraternity and facilities. With the emergence of new variants, the global pandemic has prolonged and raised concerns regarding the existing therapies. Most of the identified mutants have the potential to exacerbate the already existing crisis. In line with the urgent need for promising antivirals against the novel coronavirus, we conducted an in-silico drug docking study using SeeSAR and other bioinformatics tools and identified prospective molecules that target the nucleocapsid protein of SARS-CoV-2. The highly conserved N protein plays a crucial role in viral assembly and pathogenicity by interacting with the host ribosomal subunits and suppressing nonsense mediated decay (NMD) of viral mRNA by the host cell. In the current study, FDA approved drugs were docked into pockets created within the N protein including the crucial conserved residues and analyzed for their affinity. The docked compounds give us novel plausible models that can be inspected further and paves way for the development of potent therapeutics against SARS-CoV-2.
