Frequency-specific high-definition transcranial alternating current stimulation (HD-tACS) may facilitate recovery in patients with prolonged disorders of consciousness (pDoC), but predictive biomarkers and mechanistic insights are limited. To investigate the therapeutic efficacy of 40 Hz or 4 Hz HD-tACS in pDoC. Sixty pDoC patients were randomized into four groups receiving 40 Hz or 4 Hz HD-tACS, each with a corresponding sham condition. Behavioral recovery was assessed via Coma Recovery Scale-Revised (CRS-R), network connectivity was analyzed with electroencephalography (EEG), and baseline 40 Hz auditory steady-state response (ASSR) was explored as a predictive biomarker. Five days of 40 Hz HD-tACS yielded greater improvements of total CRS-R scores than 4 Hz or sham, particularly in visual and arousal subscales. EEG revealed strengthened short- and long-range connectivity quantified by phase-locking value (PLV), correlating with behavioral gains. At six months, 37.5% of 40 Hz patients got a good outcome with Glasgow Outcome Scale-Extended (GOS-E) score ≥ 4, which was significantly higher than that observed in the sham group (P = 0.019). Higher baseline ASSR predicted stronger network enhancement and clinical improvement. 40 Hz HD-tACS promotes recovery and network reorganization in pDoC. Baseline ASSR may provide complementary information for understanding individual variability in responses to frequency-specific neuromodulation.

The choice of preprocessing pipeline for T1-weighted structural MRI substantially influences grey-matter measurements, brain-trait associations, and prediction accuracy. Such analytic variability challenges reproducibility and generalizability in large-scale neuroimaging. Same-Data Meta-Analysis (SDMA), a multiverse framework that combines statistical maps derived from multiple analytic workflows, has been proposed to increase robustness of inference. However, SDMA has not been applied to structural MRI nor evaluated with respect to predictive performance and spatial harmonization constraints. We analyzed structural MRI data from 39,955 UK Biobank participants (discovery n = 23,288; replication n = 16,538). Grey-matter maps were derived using three widely used volume-based pipelines (FSLVBM, FSLANAT, CAT12-ENIGMA), which differ in spatial templates, voxel dimensions, and grey-matter masks. For four phenotypes highly correlated with brain measurements (diabetes, high blood pressure, maternal smoking around birth, and alcohol consumption frequency), we conducted Brain-Wide Association Studies (BWAS). Pipeline-specific association maps were combined using two SDMA approaches (Stouffer and generalized least squares). Because pipelines operate in distinct anatomical spaces, association maps were projected across templates prior to combination, requiring the selection of a reference space. We systematically evaluated how projection strategy and choice of reference space influenced regional discovery, replication, and prediction. SDMA increased the number of significant clusters (+18% on average - standard deviation of 7.8- across all reference space) and improved cluster-level replication rates (e.g., from 5% to 21% in CAT12) compared to single-pipeline analyses, identifying additional cerebellar and subcortical regions not consistently detected by individual workflows. However, results varied depending on the chosen reference space, highlighting that projection between templates introduces non-trivial spatial distortions and affects regional detection. Despite improved regional discovery, SDMA did not enhance prediction accuracy when using significant voxels. When applied to Best Linear Unbiased Predictors (BLUP), SDMA partially mitigated cross-pipeline performance loss but remained inferior to single-pipeline training. In contrast, stacking consistently improved prediction accuracy by ∼23%. These findings demonstrate that while SDMA enhances robustness of regional inference across preprocessing pipelines, projection choices critically shape multiverse outcomes and predictive gains remain limited. Our results provide practical guidance for combining structural MRI workflows and clarify methodological trade-offs in large-scale brain mapping.

Adaptive behavior requires balancing generalization from past experience with the flexible selection of goal-relevant information. Cognitive maps provide a structured basis for this balance, yet how a stable relational map supports task-appropriate decision coordinates remains unclear. Combining fMRI with CNN-LSTM modeling, we show that a shared two-dimensional social map coexists with localized, goal-dependent geometric reformatting. A hippocampal-orbitofrontal/medial prefrontal network preserves a reusable 2D relational structure, whereas a frontoparietal control network expresses goal-aligned 1D decision projections. Crucially, these readouts show systematic geometric shaping along distinct functional axes. Representational stretching along a "congruent axis" was associated with redundant structure across dimensions, whereas goal-directed warping along a "task-relevant axis" selectively emphasized currently relevant information during decision conflict. This localized geometric reformatting was accompanied by dynamic hippocampus-insula-precuneus interactions. As similar patterns emerged spontaneously in recurrent networks, our findings support geometric reformatting as a complementary readout strategy through which a stable multidimensional relational map can be flexibly adapted to current decision demands.

Imaging genomics provides a powerful paradigm for decoding the complex interplay between molecular variation and brain functional phenotypes in neurodegenerative disorders. In this study, the molecular modality is specifically gene expression data rather than time-invariant genetic variants. However, statistically integrating these high-dimensional modalities remains challenging, as existing methods often rely on incomplete biological priors or overlook the latent modular structure of the data. To address this, we propose BRIDGE (Biclustering and ensemble penalized Regression for Integrative Data in Genomics and nEuroimaging), a unified statistical framework that synergistically integrates biclustering with ensemble penalized regression. Our approach proceeds in three interdependent stages: (1) accurate estimation of individual brain functional networks via sparse precision matrices; (2) data-driven discovery of regulatory modules-defined as subsets of gene expression features co-varying with subsets of brain connections-using a sparse biclustering algorithm; and (3) stable disease classification and biomarker identification via dimensionality reduction integrated with an ensemble penalized logistic regression model. Simulation studies demonstrate that BRIDGE outperforms ablation variants of the proposed framework and external competing benchmark methods in both feature selection performance and classification accuracy. In an application to the Alzheimer's Disease Neuroimaging Initiative (ADNI) cohort, the framework achieved robust out-of-sample classification performance and uncovered biologically interpretable disease-related modules. These results highlight BRIDGE as a rigorous tool for integrative imaging genomics, capable of identifying pathogenic factors without reliance on a priori knowledge.

Transcutaneous auricular vagus nerve stimulation (taVNS) is a non-invasive neuromodulation approach that may influence memory-related brain activity. Theta oscillations are closely related to episodic memory processing, but it remains unclear whether taVNS modulates memory-related theta activity in a frequency-specific manner and how these effects change after stimulation. This study investigated the sustained effects of 20 Hz and 40 Hz taVNS on free recall and resting-state EEG theta oscillations in healthy young adults. Forty-four participants were enrolled in this randomized, single-blind, sham-controlled, three-arm parallel-group study. They were randomly assigned to the sham stimulation group, the 20 Hz taVNS group, or the 40 Hz taVNS group. Free recall performance was assessed before and after stimulation, and resting-state EEG was recorded at baseline and multiple time points after stimulation. The results showed that participants receiving 40 Hz taVNS exhibited better free recall performance than those receiving sham stimulation or 20 Hz stimulation. 40 Hz taVNS increased the relative power of theta waves, particularly in the frontal region. This enhancement was an immediate increase followed by a gradual decrease over time. Frontal theta activity and stimulation-induced theta changes were positively correlated with free recall performance. Furthermore, exploratory Bayesian mediation analysis supported a theta-dependent model. This study suggests that 40 Hz taVNS may promote verbal free recall performance by inducing transient enhancement of frontal theta wave activity, providing preliminary evidence for the modulation of memory-related neural oscillations by frequency-specific taVNS.

The behavioural activity of freely moving animals is a confounding factor that affects the recording, analysis, and final results of animal EEG experiments. Along with the lack of standardisation in animal in vivo electrophysiology experiments, this could lead to huge inconsistencies, especially in the analysis of centrally acting drugs. Therefore, the main aim of this paper is to investigate the effects of behavioural activity versus inactivity on the multichannel EEG in freely moving rats. In a large sample (n = 116) of waking recordings from 12 cortical electrodes (ECoG) in Wistar rats, we evaluated behavioural activity-related changes in the power spectrum, current source density, and power-based global functional connectivity (GFC) in a 3D rat brain model, according to the TOHOKU Rat Brain Atlas. The main findings were that behavioural activity was associated with 1) a robust power increase in 6-8 Hz, peaking at 7 Hz with maximum changes over the parietal and temporal cortex, 2) an increase in gamma (30-40 Hz) and high-gamma (55-100 Hz) power across the whole brain, 3) a decrease in delta (1-4 Hz) and beta (12-25 Hz) and high-beta (25-30 Hz) power across the whole cortex. Changes were also localised in subcortical regions, particularly in the diencephalon/thalamus. The GFC analysis showed a similar pattern of changes across the 6-8 Hz, delta, and beta bands; however, GFC in the gamma band decreased, while high-gamma GFC increased. Again, the GFC analysis revealed changes in connectivity within subcortical structures, primarily in the thalamus. None of the measures was affected in the alpha band (8-12 Hz). These findings emphasise behavioural state as a critical factor influencing EEG outcomes, with important implications for the standardisation and translational validity of preclinical neurophysiological studies.

Efficient auditory attention requires prioritizing target over nontarget information. Although attentional deficits are a core feature of attention-deficit/hyperactivity disorder (ADHD), the neural mechanisms underlying auditory prioritization during cross-modal attention remain unclear. We examined neural differentiation between target and nontarget auditory information in children with ADHD. Ninety-five school-aged children (mean age = 10.81 years), including 46 children with ADHD and 49 typically developing (TD) children, performed a visually cued auditory spatial attention task during electroencephalography (EEG) recording. Auditory selective attention was assessed using the N2-anterior-contralateral (N2ac) component and multivariate pattern analysis (MVPA). TD children showed enhanced MVPA decoding of target locations and reduced decoding of nontarget locations. Greater target-nontarget neural differentiation was associated with larger N2ac amplitudes. In contrast, children with ADHD showed reduced target lateralization, smaller N2ac amplitudes, weaker target decoding, and enhanced nontarget decoding, resulting in diminished neural differentiation between relevant and irrelevant auditory information. Reduced target-related lateralization was associated with greater response-time variability, whereas reduced target decoding accuracy was linked to more severe hyperactivity-impulsivity symptoms. Together, these findings indicate that children with ADHD show altered neural prioritization of auditory information within a visually cued attention context. Altered neural prioritization was associated with behavioral variability and symptom severity, providing new insight into the neurocognitive mechanisms underlying attentional dysfunction in ADHD.

Recent advances in fMRI-based semantic decoding and visual reconstruction have achieved remarkable progress. However, severe inter-subject variability and the scarcity of paired fMRI-image data remain major obstacles to cross-subject generalization. To overcome these challenges, we propose DeID-Aligner, a brain functional alignment framework that enables multi-task visual decoding across subjects under limited fMRI data. The method first constructs a region-of-interest (ROI)-wise fMRI encoder that captures regional functional representations. A dynamic Patch-to-ROI fusion mechanism with Progressive Modal Attention Transition (PMAT) explicitly associates image patches with cortical regions, enabling fine-grained cross-modal semantic alignment while gradually shifting from cross-modal to fMRI-only for robust inference. To remove subject-specific information, we introduce a de-identification task implemented with a graph attention network (DIT-GAT), which models inter-ROI interaction patterns to reduce subject-specific information and suppress identity cues. Extensive experiments on the Natural Scenes Dataset (NSD) demonstrate that DeID-Aligner significantly outperforms state-of-the-art methods in cross-subject category classification, multi-label classification, image captioning, and image reconstruction. Voxel-wise unique variance analysis further reveals that the Patch-to-ROI fusion mechanism substantially enhances semantic representations in high-level visual areas while maintaining structural fidelity in early visual cortices. These results suggest that DeID-Aligner effectively bridges inter-subject functional disparities and provides a unified, interpretable framework for multi-task visual neural decoding.

Language relies on a left-lateralized fronto-temporal network whose functional organization likely reflects structural factors that emerge early in neurodevelopment. One such factor is sulcal depth, which has been proposed as a key anatomical marker of functional specialization. Here, we investigated potential structure-function relationships with respect to phonological encoding, a core linguistic process linking lexical representations and articulatory planning. Thirty-two healthy adults performed a rhyming judgment task, known to be sensitive to phonological coding, during fMRI. Activation during task performance was related to cortical morphology derived from structural MRI in the same subjects, focusing on sulcal depth and other standard measures (i.e. cortical volume, surface area and thickness). Rhyming task performance elicited predominantely left-lateralized activation in the inferior frontal cortex and other regions including the precentral, postcentral, supramarginal, and superior temporal cortex, as well as the supplementary motor area, the middle cingulate and calcarine cortex, the left cerebellum and the basal ganglia. A cluster in the left inferior frontal gyrus (pars opercularis, a subregion of Broca's area) showed a significant association between sulcal depth and activation during the rhyming task. No associations were observed for other regions or for other structural measures. These findings raise the possibility that phonological encoding may have an origin in early-emerging cortical folding patterns and support the view that core language functions are scaffolded by genetically driven neurodevelopmental processes.

    10. Functional organization of the perirhinal cortex in natural scene processing and memory.

The perirhinal cortex (PRC) has long been implicated in object perception and recognition memory; however, its role in processing complex natural scenes and its internal functional organization are not well understood. In the current study, we investigated the relationship between object- and scene-related visual feature (faces, bodies, and place) representations in PRC and the visual cortex, and the spatial pattern within PRC for visual processing and memory. Utilizing fMRI data from a continuous recognition task involving natural scenes (from the Natural Scenes Dataset), we first quantified the trial-by-trial decoding accuracy for visual features across distributed visual cortical regions and within the PRC. Our results demonstrate that multivariate pattern feature decodability in the PRC increase significantly when visual features are broadly represented across distributed regions in visual cortex. The PRC exhibited a robust subsequent memory effect, though the memory outcome was not directly predicted by the level of distributed visual feature representation. Critically, left PRC showed a posterior bias for the multivariate decoding of the place feature. Vertex-level univariate analyses uncovered a posterior bias for face-feature-driven vertices and an anterior bias for memory-driven vertices, which is more consistent in the right PRC. Together, these findings suggest that the PRC may play a flexible role in scene processing that extends beyond object representation and highlights an anterior-posterior functional gradient within the human PRC that supports perceptual or mnemonic processes.
