1. PD-L1 immunoreactivity and its association with tumor grade in canine mammary carcinomas.
PMID:期刊:日期:2026-09-24Programmed death ligand 1 (PD-L1) is a transmembrane protein involved in tumor immune evasion. Although widely studied in human oncology, its role in canine mammary carcinoma remains poorly characterized. We quantified gene mRNA levels by reverse-transcription quantitative real-time PCR (RT-qPCR), evaluated PD-L1 immunoreactivity by immunohistochemistry (IHC) in canine mammary carcinomas, and tested their association with tumor grade and epidemiologic factors using 96 formalin-fixed, paraffin-embedded tissue samples. The mean age of affected dogs was 9.6 y, with Miniature Poodles and mixed-breed dogs most frequently represented. We found no statistically significant associations between mRNA levels with histologic subtype or epidemiologic factors. However, PD-L1 immunoreactivity was significantly associated with tumor grade in the bivariate analysis ( 0.017; Cramer V = 0.3), supporting the diagnostic value of PD-L1 IHC in canine mammary carcinomas and suggesting its utility as a marker of tumor aggressiveness.

2. Diagnostic and prognostic value of hematology measurands and lactate dehydrogenase isoenzyme activities in canine immune thrombocytopenia at a primary care hospital.
PMID:期刊:日期:2026-09-24Immune thrombocytopenia (ITP) is a common hematologic disorder in dogs for which reliable diagnostic and prognostic markers are limited. We evaluated retrospectively the diagnostic and prognostic value of hematology measurands and lactate dehydrogenase (LDH) isoenzyme activities in a primary care setting. Dogs with thrombocytopenia were classified into ITP ( = 14) and non-ITP ( = 27) groups, and platelet count, plateletcrit, mean platelet volume (MPV), and LDH isoenzyme activity profiles were compared between groups. Platelet count and plateletcrit were significantly lower in the ITP group, with sensitivities and specificities of 92.3% and 100% and 85.7% and 100% at cutoffs of 8 × 10/L and 0.0001 L/L, respectively. LDH1-3 activities were significantly higher, whereas LDH5 activities were significantly lower in the ITP group, although specificity was limited. Within the ITP group, dogs with bleeding ( = 8) had significantly lower MPVs. Dogs responding to prednisolone monotherapy ( = 7) had higher platelet counts and MPVs. The outcome predictive performance of a platelet count ≥3 × 10/L or MPV ≥15.8 fL was excellent (100% sensitivity and specificity in this cohort). Platelet count and plateletcrit were reliable diagnostic indicators for canine ITP in primary care. LDH isoenzyme activities offer limited standalone diagnostic value but may provide supplementary information. MPV may serve as an indicator of bleeding risk and a predictor of treatment response.

PMID:期刊:日期:2026-09-22Here, we describe a case of rhinitis in a cow and review the literature on diseases caused by spp. and dematiaceous fungi historically classified morphologically as , , or in domestic ruminants. In March 2024, an adult Holstein cow from a dairy herd in Uruguay developed bilateral nodules in the nasal mucosa, resulting in dyspnea, snoring, nasal discharge, and weight loss. Histologically, severe chronic eosinophilic granulomatous rhinitis was found, with giant cells and intra- and extracellular fungi. was identified by PCR and sequencing of the extended 28S rDNA region. Our literature search retrieved ~40 bovine cases in the United States, Australia, South Africa, Namibia, and Uruguay, 1 caprine case in Brazil, and no ovine cases. Clinical signs included chronic dyspnea, stertor, mucopurulent nasal discharge, epistaxis, and weight loss, generally affecting adult beef cattle. The lesions usually affected the nasal mucosa and submucosa, rarely involved the skin or regional lymph nodes, and occasionally were restricted to the skin. Bilateral, raised, 0.1-5-cm granulomatous nasal nodules resulted in restriction of airflow. The hallmark microscopic lesion is chronic eosinophilic granulomatous inflammation with giant cells and intralesional fungal structures, including ~5-30-µm, round-to-ovoid chlamydospores and short ~2-6-µm-wide septate hyphae, which may have brown-pigmented walls. Definitive fungal identification requires molecular testing, which is rarely pursued.

5. Biomarkers for monitoring racehorse musculoskeletal injuries and injury risk.
PMID:期刊:日期:2026-09-11In most catastrophic racehorse fractures, postmortem examinations have found evidence of chronic accumulation of microdamage attributed to high-stress loading during high-speed exercise. Biological responses to this microdamage accumulation may be an avenue to identify biomarkers indicative of this damage, offering insight into musculoskeletal adaptation and dysregulation. Since the first description of an equine bone degradation biomarker in 1975, our understanding of various biological molecules associated with bone modeling, remodeling, and damage has greatly improved, although none has become a mainstream, clinical screening tool. Here, we describe the history and current state of research on biomarkers in racehorse musculoskeletal health. Although not yet in clinical use, biomarkers present a promising area of research for screening and monitoring racehorses at risk for musculoskeletal injury.

6. Quantitative multiplex detection of 9 feline respiratory pathogens with enhanced sensitivity: a cost-effective integrated platform combining RT-PCR/PCR and capillary electrophoresis.
PMID:期刊:日期:2026-09-11Pathogen infections and coinfections are critical determinants of feline upper respiratory tract disease; although the etiologic agents are diverse, their clinical manifestations often overlap. Established detection panels for feline upper respiratory tract infections (FURTIs) primarily focus on core pathogens-feline calicivirus (FCV), felid alphaherpesvirus 1 (FeAHV1), (MF), (CF), (BB), and influenza A virus (IAV)-and frequently do not incorporate emerging zoonotic agents (e.g., SARS-CoV-2) and overlooked streptococcal species ( subsp. [SEZ] and [SC]) in a multiplex assay. Furthermore, simultaneous quantification remains technically challenging. We developed an integrated multiplex reverse-transcription PCR (RT-PCR)/PCR-capillary electrophoresis (CE) platform for simultaneous quantitative detection of 9 feline respiratory pathogens (FCV, FeAHV1, MF, CF, BB, IAV, SARS-CoV-2, SEZ, SC). Requiring only a standard thermal cycler and a basic CE instrument, our platform is highly accessible to resource-limited laboratories, offering high specificity and a 1.6-h turnaround time. With a limit of detection of 1 copy/µL across all targets, our multiplex assay had exceptional sensitivity, surpassing conventional singleplex RT-PCR/PCR on both agarose gel and real-time PCR platforms. Notably, we achieved a 2.2-fold higher detection rate than the conventional gold standard (singleplex RT-PCR/PCR with agarose gel). This enhanced sensitivity enabled us to identify even low-pathogen-load samples, often missed by conventional assays. When used with clinical specimens, the platform offered comprehensive coverage of major and emerging zoonotic FURTI pathogens, confirming its potential as a cost-effective, high-efficiency tool for differential diagnosis, targeted therapy, and epidemiologic surveillance.

7. {"_":"Acute hepatic necrosis associated with poisoning in cattle in Brazil.","i":["Trema micranthum"]}

Blume (Jamaican nettletree) is a hepatotoxic tree that is widely distributed throughout the Americas and has been associated with poisoning in several domestic animal species. To our knowledge, spontaneous intoxication in cattle has not been reported previously. Here, we describe 2 spontaneous outbreaks of poisoning in Nelore cows in the state of Mato Grosso, Brazil. Affected cattle developed apathy, ataxia, and recumbency, followed by death within hours. Autopsy revealed jaundice and enlarged livers with a diffuse mottled red-and-white appearance. Histologically, severe centrilobular-to-massive hepatocellular coagulative necrosis was observed. Epidemiologic investigation identified recent access to paddocks containing vigorously sprouting following deforestation or tree fall, with clear evidence of grazing. The combination of pathology findings and field investigation established the diagnosis of acute toxic hepatopathy caused by .

8. Methomyl poisoning in dogs: report of 5 cases and literature review.
PMID:期刊:日期:2026-09-11Methomyl is a widely available carbamate pesticide that acts as an acetylcholinesterase inhibitor. Here, we describe 5 cases of methomyl poisoning in dogs. The main gross and microscopic findings included pulmonary edema and multisystemic congestion and hemorrhages, which were most severe in the respiratory tract. The diagnosis of poisoning was confirmed in all cases by detecting methomyl in gastric contents using gas chromatography-mass spectrometry. Differential diagnoses include poisoning by organophosphates, coumarin rodenticides, and sodium fluoroacetate, which share clinical and pathology features but differ in pathogenesis. Additionally, we conducted a review of the literature on methomyl poisoning in animals focused on pathogenesis, clinical signs, lesions, and diagnosis.

9. Analytical validation of the Mindray BC-75R Vet hematology analyzer and comparison with the Siemens Advia 2120i for application in dogs and cats.
PMID:期刊:日期:2026-09-04The BC-75R Vet (Mindray Animal Medical) is a multi-species hematology analyzer that uses electrical impedance and SF-Cube technology (combining laser scatter, fluorescence, and 3-dimensional analysis) for CBC and reticulocyte analysis. We evaluated its analytical performance and compared it with the Advia 2120i (Siemens), a widely used analyzer in veterinary clinical pathology. CBCs were performed on EDTA-anticoagulated blood samples from 411 dogs and 225 cats submitted for routine testing. Analytical precision, linearity, carryover, and total observed error (TE) were assessed according to ASVCP guidelines. We used a 2120i as the reference method and manual differential counts for WBC evaluation. RIs were calculated from 131 healthy dogs and 53 cats. The BC-75R Vet fulfilled the ASVCP requirements for repeatability (CVs < 5% for most variables) and linearity ( > 0.97), with negligible carryover. TE values were within ASVCP allowable error limits, except for WBC types in QC material, which remained acceptable for in-clinic use. Agreement with the 2120i was acceptable for most RBC-related variables; however, systematic and proportional errors were found for mean corpuscular hemoglobin concentration (MCHC) in both species, and a systematic error for mean corpuscular volume (MCV) was found in dogs. Agreement for WBC and platelet counts was generally good, although discrepancies were noted for monocyte and mean platelet volume in both species. These differences likely reflect methodologic differences and known challenges in feline hematology. Despite these limitations, the BC-75R Vet met ASVCP standards and appears suitable for routine testing in dogs and cats. Manual smear review remains essential in selected cases.

PMID:期刊:日期:2026-09-03An 8-mo-old Japanese Black beef heifer calf was presented with anorexia, dysuria, and abdominal pain. Ultrasonographically, the left renal pelvis and calyces were markedly dilated and contained numerous calculi; the right renal pelvis was filled with hyperechoic material without obvious calculi. Despite supportive care and antimicrobial treatment with ceftiofur, the calf died 4 d after presentation. Autopsy revealed severe hydronephrosis with numerous struvite calculi in the left kidney and advanced pyonephrosis in the right kidney. was isolated antemortem from urine and postmortem from purulent material in both kidneys, ureters, urinary bladder, and an associated urachal remnant abscess. As found in our case, infection in cattle can be associated with both obstructive and suppurative renal lesions within a single animal. Our case emphasizes the importance of comprehensive diagnostic investigation, including bilateral renal ultrasonography and thorough bacteriologic evaluation, and offers diagnostic insight into the variable pathologic manifestations of -associated urinary tract disease in cattle.
