1. Evaluation of Two Induction Protocols for Oclacitinib in Dogs.
PMID:日期:2026-09-20Oclacitinib is approved for the treatment of canine atopic dermatitis (AD) and pruritus. The conventional administration protocol includes twice-daily oral dosing for 14 days with a subsequent reduction to once-daily administration. This reduction sometimes leads to an increase in pruritus. The aim of this study was to compare the changes in skin lesions and pruritus following a shortened induction versus the conventional protocol for oclacitinib. Forty-five client-owned dogs with AD were randomised into two groups. The conventional protocol group (CP) received oclacitinib tablets at 0.4-0.6 mg/kg twice-daily for 14 days, then once-daily for 7 days. In the shortened induction protocol group (SIP), oclacitinib was administered twice-daily for 6 days, then once-daily for 15 days. Pruritus was assessed daily by the owners using a pruritus Visual Analog Scale (PVAS). Lesions were scored with the Canine Atopic Dermatitis Lesion Index (CADLI) at Day (D) 0 and D21. Data from 40 dogs were available. Mean CADLI scores decreased from 13.4 to 8 in the CP group (p = 0.12) and from 12.9 to 6.9 in the SIP group (p = 0.01), respectively. Mean pruritus scores decreased from 6.4 to 2.1 and from 5.7 to 1.9, respectively (p < 0.0001 each). No between-group differences were observed. An increase in pruritus following dose reduction was seen in two dogs (CP) and one dog (SIP). The SIP was not significantly different to the CP with respect to improvement in lesion severity or pruritus.

4. Efficacy and Field Safety of the Second-Generation, Highly Selective JAK1 Inhibitor Atinvicitinib for the Treatment of Canine Atopic Dermatitis: A Multicentre, Blinded, Randomised, Placebo-Controlled Clinical Trial in Client-Owned Dogs.
PMID:日期:2026-09-17Janus kinase (JAK) inhibitors alleviate pruritus and clinical signs of canine atopic dermatitis (cAD). To confirm the optimal dosing regimen of atinvicitinib, a second-generation highly selective inhibitor of JAK1 in dogs. Sixty-one client-owned dogs, ≥ 12 months and ≥ 2 kg, with cAD. In a randomised, masked, placebo-controlled clinical trial, dogs received atinvicitinib, with food, at 0.8-1.2 mg/kg, once daily for 28 days (q.d. group, n = 21) or 0.4-0.6 mg/kg twice daily for 14 days followed by once daily dosing for 14 days (b.i.d.-q.d. group, n = 21); or placebo b.i.d.-q.d. (control group, n = 19). Owners assessed pruritus (Day [D]0-7, D14, D28) using a Visual Analog Scale (PVAS); veterinary surgeons assessed skin lesions (D0, D28) using the cAD Extent and Severity Index, 4th iteration (CADESI-04). On D0, D14 and D28, blood and urine samples were collected. Treatment success was defined as a ≥ 50% reduction from baseline PVAS or CADESI-04 on D28. Proportions of dogs with ≥ 2 PVAS reduction, and reduction in PVAS and/or CADESI-04 from baseline, also were assessed. Treatment success was 87.5% for the q.d. group (p = 0.011) and 73.3% for the b.i.d.-q.d. group (p = 0.033) (23.1% control group). There was ≥ 2 decrease in PVAS in 80.0% of q.d. dogs. (16.7% controls, p = 0.042). Compared to controls, atinvicitinib-treated dogs had a significantly greater reduction in PVAS (q.d., p = 0.003, b.i.d.-q.d., p = 0.001) and CADESI-04 (q.d. group, p = 0.046). Mean results of clinical pathological values remained within reference ranges. Treatment was well-tolerated. Once-daily atinvicitinib was safe, well-tolerated and effective at rapidly reducing pruritus and resolving cAD-associated skin lesions.

6. Radiation-Induced Alopecia in Dogs With Nasal Tumours: Coat Type Influences Development and Onset.
PMID:日期:2026-09-10Radiation-induced alopecia (RIA) is a common adverse effect of radiation therapy, yet factors influencing its development in dogs remain unclear. To characterise acute RIA in dogs with nasal tumours and to evaluate associations with coat type and radiation dose. Thirty-eight client-owned dogs were treated with radiation therapy for nasal tumours. Medical records were reviewed retrospectively. Dogs were classified by coat type (straight versus curly or wire-haired). Time to onset of alopecia was assessed as the interval from the first irradiation to the first detection of alopecia, and cumulative skin dose was calculated up to alopecia onset. Dogs developing acute RIA during radiation therapy were compared with those without RIA. Dogs developing alopecia only after completion of radiation therapy were excluded. Seventeen of 34 dogs developed acute RIA during radiation therapy. Coat type was significantly associated with RIA development (p = 0.003), with all dogs with curly or wire-haired coats affected. These dogs developed RIA earlier than those with straight-haired coats (median 14 versus 20 days; p = 0.047). Within affected dogs, RIA developed first in regions receiving higher skin doses and subsequently progressed to regions receiving lower skin doses. Coat type influences the development of acute RIA in dogs, with curly or wire-haired breeds showing earlier onset and greater extent of alopecia. Alopecia progression reflects regional skin dose distribution. These findings may assist clinicians in predicting the risk of RIA and informing owners about the potential development and timing of radiation-induced alopecia.

7. Systemic Inflammation in Dogs With Atopic Dermatitis Compared to Healthy Dogs and Dogs With Pemphigus Foliaceus: A Study Using a Limited Cytokine Panel.
PMID:日期:2026-09-03Atopic dermatitis (AD) is a common chronic, pruritic and inflammatory skin disease in both humans and dogs. In people, AD is increasingly recognised as a systemic inflammatory disease in which immune dysregulation drives chronic inflammation extending beyond the skin, particularly in moderate-to-severe disease. Whether a comparable systemic inflammation exists in canine AD, however, remains poorly characterised. We hypothesised that serum cytokine concentrations would differ between dogs with varying AD severity and healthy controls. This study aimed to identify systemic inflammatory markers correlating with AD severity, by comparing cytokine profiles in dogs with mild AD and moderate-to-severe AD against healthy dogs (negative control) and dogs with active pemphigus foliaceus (PF) lesions (positive control). Forty client-owned dogs were enrolled, with 10 dogs in each of four groups: healthy controls, mild AD, moderate-to-severe AD and PF. Serum concentrations of tumour necrosis factor (TNF)-α, TNF-β/lymphotoxin-α, interleukin (IL)-1β, IL-6, IL-12 and IL-1 receptor antagonist (IL-1RA) were measured using standardised immunoassays to evaluate systemic inflammation across groups. Marked interindividual variability was observed across all groups. No statistically significant differences in serum concentrations of any of the six cytokines were detected between the healthy, mild AD, moderate-to-severe AD and PF groups. In a limited cytokine panel, no evidence of differential systemic inflammation was detected across groups, including dogs with moderate-to-severe AD compared to healthy controls. These findings may reflect true lack of systemic inflammation in dogs or may be attributable to the small sample size, high interindividual variability or the limited scope of the cytokine panel evaluated.

8. Diagnostic Performance of Immunoglobulin E Serology, Prick Test and Patch Test With Food Allergens in Dogs With Atopic Dermatitis and Adverse Food Reactions Using the Double-Blind Placebo-Controlled Food Challenge as Gold Standard.
PMID:日期:2026-08-28In dogs with adverse food reactions (AFRs), immunoglobulin (Ig)E serology, prick test and patch test with food allergens have shown variable diagnostic performance for the identification of the offending foods, when the latter were identified using open food challenges. To evaluate the diagnostic performance of these tests in dogs with atopic dermatitis (AD) and AFRs, using the double-blind placebo-controlled food challenge (DBPCFC) as the gold standard. Twelve dogs with AD and AFRs underwent DBPCFCs with eight food items. IgE serology, prick test and patch test with the same eight food allergens were performed after the diagnosis of AD and before the diagnostic investigation for AFRs and the DBPCFCs. The sensitivity (3%-60.6%), specificity (54.8%-98.4%), positive predictive value (25%-66.7%) and negative predictive value (64.1%-72.3%) of the three tests and of their combination were unsatisfactory. Furthermore, their positive and negative likelihood ratio values were not in the clinically informative range, apart from the prick test having a weakly informative positive likelihood ratio value of 3.81. Under the conditions of this study, the diagnostic performance of IgE serology, prick test and/or patch test with food allergens was poor using the DBPCFC as gold standard.

9. Time Required for Topical Difluprednate to Suppress the Intradermal Histamine-Induced Vascular Reaction in Dogs.
PMID:日期:2026-08-27Topical glucocorticoid (GC) is used to manage inflammatory skin diseases in dogs. However, contact times associated with detectable topical GC effects have not been clarified in dogs. Uncertainty regarding contact time may lead to premature removal or overuse, compromising efficacy and safety. To determine contact times at which detectable GC effects are observed after topical difluprednate lotion application in dogs. Six male Beagle dogs. Suppression of the histamine-induced vascular reaction (HVR), based on the principles of the vasoconstrictor assay, was used as an indicator of detectable GC effects. Difluprednate lotion was applied to a circular area (2 cm in diameter) on the lateral trunk 5, 10 and 30 min before intradermal histamine injection. Fluorescein was administered intravenously 5 min before histamine injection to visualise HVR. HVR images were obtained 15 min after histamine injection under UV illumination, and the fluorescent area was measured using ImageJ. Statistical analysis was performed using Dunnett's multiple comparison test. When difluprednate application was performed 5, 10 and 30 min before histamine injection, HVR areas were smaller than untreated control sites (p < 0.01). No significant differences were detected among sites treated 5, 10 and 30 min before histamine injection. Difluprednate lotion suppressed the HVR when applied 5, 10 and 30 min before histamine injection, indicating detectable dermal GC activity at the evaluated time points. Information regarding contact times at which topical GC effects can be detected may help clinicians to provide contact-time instructions and improve treatment management.
