1. Oral and topical phytopharmaceuticals for bovine mastitis: Effects of Terminalia bellerica, Boswellia serrata, Bergenia ciliata, and Piper betle formulations on udder health and immune responses.
PMID:日期:2026-09-20This study evaluated the efficacy of oral and topical phytopharmaceutical treatments containing extracts of Terminalia bellerica, Boswellia serrata, Bergenia ciliata, and Piper betle in modulating immune response and reducing mammary gland inflammation in Karan Fries cows. Sixty-six early lactating cows were screened for subclinical (SCM) and clinical mastitis (CM) and divided into treatment groups. In the first experiment, SCM cows (n = 6 per group) were allocated to six groups. One group received no treatment (negative control), one group received Mastilep gel (positive control), and three groups received topical mastitis-formulated gels containing extracts of Terminalia bellerica, Boswellia serrata, Bergenia ciliata, and Piper betle. MST-F1 contained the four plant extracts alone, MST-F2 contained the four plant extracts with 1% citronellol oil, and MST-F3 contained the four plant extracts with 1% citronellol oil and 1% eucalyptol. The sixth group received an oral phytopharmaceutical product (PPP) containing the same four plant extracts. Results indicated that topical MST-F2 and oral PPP reduced mammary inflammatory indicators and maintained neutrophil viability and phagocytic activity. The second experiment combined the most effective topical formula (MST-F2) with oral PPP to treat SCM and CM cows (n = 6 per group). Controls included untreated SCM and CM cows (negative) and antibiotic-treated CM cows (positive). The combined treatment significantly (P < 0.05) reduced mammary infections, demonstrating efficacy comparable to antibiotic therapy. This treatment decreased somatic cell counts and was associated with maintained neutrophil viability and phagocytic activity. It helped in shifting from neutrophil dominance to increased lymphocyte percentages. The treatment downregulated neutrophil chemokine receptors (CXCR1, CXCR2) and the pattern recognition receptor CD14 while reducing pro-inflammatory cytokines (TNF-α, IL-6), indicating an attenuation of the inflammatory cascade. These findings suggest that phytopharmaceutical products offer a promising alternative to antibiotics for managing mastitis, improving both udder health and immune function in dairy cows.

2. Mapping immune targets in peste des petits ruminants virus hemagglutinin: An integrated computational framework for vaccine candidate prioritization.
PMID:日期:2026-09-17Peste des petits ruminants virus (PPRV) is a major transboundary viral pathogen of small ruminants and causes substantial economic losses in endemic regions. The hemagglutinin (H) protein mediates receptor recognition and host-cell attachment and is an important target for vaccine development. This study applied an integrated computational framework to identify conserved immunogenic regions within the PPRV H protein. A total of 64 unique PPRV H protein sequences were analyzed using multiple sequence alignment, entropy-based conservation profiling, conservation-aware epitope prediction, comparison with experimentally characterized immune determinants, structural mapping, candidate-region ranking, and exploratory neural-network attribution analysis. Predicted epitopes were compared with reported immune determinants and contextualized using conservation and structural data. The workflow identified 9 predicted B-cell epitope candidates and 151 predicted T-cell peptide candidates distributed throughout the H protein sequence. The highest-scoring predicted B-cell epitope candidate was localized within residues 399-407 (SGPWSEGRI, Epitope_Score: 1.0000, length: 9 aa), whereas the highest-scoring T-cell peptide candidate corresponded to residues 36-44 (YILLGVLLV; score: 0.889). Conservation analysis identified 412 residues with conservation scores greater than 0.9. Comparison with experimentally characterized immune determinants showed literature-based correspondence with selected predicted regions. Structural mapping provided three-dimensional context for conserved and predicted regions. Exploratory neural-network attribution scores were generated descriptively, without biological interpretation as validated antigenicity measures. Integrated computational analysis combining conservation profiling, immune epitope prediction, comparison with experimentally characterized immune determinants, structural interpretation, candidate-region ranking, and exploratory neural-network attribution analysis identified conserved and computationally predicted immune candidate regions within the PPRV H protein. Residues 399-407 (SGPWSEGRI) represented the highest-scoring computationally predicted B-cell epitope region and warrant further investigation and experimental confirmation.

3. Variation in immune responsiveness is associated with pregnancy status in beef heifers.
PMID:日期:2026-09-15Variation in immune responsiveness among heifers may contribute to differences in reproductive success. Herein, we evaluated whether cell-mediated immune (CMI) responses at early pregnancy (day 62) differ between pregnant and non-pregnant heifers by characterizing responses to viral and mitogen stimulation under standardized in vitro conditions. To this end, a subset of pregnant and non-pregnant heifers (n = 12 per group) was selected and assessed for immune responses using in vitro stimulation with BVDV-1, BVDV-2, and mitogen. No differences were observed in immune cell population frequencies or responses to non-specific mitogen stimulation and in non-stimulated cells (P > 0.05). Non-pregnant heifers exhibited significantly greater antigen-specific IFN-γ response following BVDV-2 stimulation for the CD4 population P ≤ 0.05), and a similar trend was also observed for the CD335 population While similar results were observed for the BVDV-1 stimulation, these observations were not significant (P > 0.05). These findings indicate that pregnant heifers demonstrate reduced antigen-specific CMI responses while maintaining overall immune competence. Collectively, our results suggest that immune responsiveness was associated with pregnancy status, and further studies are needed to validate these relationships and elucidate underlying mechanisms.

4. Host C3 complement restricts bovine coronavirus infection in vitro via virus-induced IL-1β signaling.
PMID:日期:2026-09-12Bovine coronavirus (BCoV) is a major enteric and respiratory pathogen of cattle; however, its interactions with the host complement system remain poorly characterized. We previously reported that BCoV modulates host complement C3 expression. Here, we demonstrate that C3 functions as a bona fide cell-intrinsic restriction factor against BCoV infection. Using bovine C3 overexpression, recombinant human C3 supplementation, siRNA-mediated knockdown, and pharmacological inhibition with Compstatin, gain- and loss-of-function experiments consistently demonstrated that enhanced C3 activity suppresses viral replication, whereas C3 depletion or inhibition promotes it. Mechanistically, we identify a novel IL-1β-p38 MAPK-C/EBP-β signaling axis: BCoV infection induces IL-1β expression, which in turn activates p38 MAPK and drives C/EBP-β phosphorylation, leading to transcriptional upregulation of C3. This virus-triggered, cytokine-dependent induction of C3 establishes an intracellular antiviral state that restricts BCoV replication, revealing a previously unappreciated, non-canonical role for complement C3 in intrinsic immunity.

5. A serum-reduced culture system for canine PBMC-derived macrophages enables characterization of M0, M1, and M2 phenotypes and tumor-associated interactions.
PMID:日期:2026-09-10Canine macrophages are increasingly recognized as important regulators of tumor immunity; however, in vitro culture systems that minimize serum-derived variability remain limited. In this study, we established a serum-reduced culture system for canine peripheral blood mononuclear cell (PBMC)-derived macrophages and characterized their phenotypic, transcriptional, and functional properties. PBMCs from healthy Beagle dogs (n = 3) were differentiated into M0, M1, and M2 macrophages using Macrophage Base Medium supplemented with canine serum or recombinant cytokines. Morphology was assessed by phase-contrast microscopy, macrophage identity was evaluated by Iba1 immunostaining and AIF1 expression, and exploratory transcriptomic profiles were examined by RNA sequencing (n = 1). Phagocytic activity was assessed using FITC-labeled latex beads and flow cytometry, and macrophage-tumor cell interactions were evaluated by co-culture with canine urothelial carcinoma cells. M0, M1, and M2 macrophages exhibited distinct morphologies and transcriptional patterns. M1 macrophages showed increased expression of inflammatory genes, including IL1B and IFNG, whereas M2 macrophages showed increased expression of genes associated with alternative activation and immune regulation, including CD209, CD5L, and IL33. Phagocytic activity was observed in all subsets. In co-culture experiments, macrophages exhibited polarization-dependent interaction patterns with tumor cells, with M2 macrophages exhibiting features reminiscent of tumor-associated macrophages. These findings provide a pilot evaluation of this serum-reduced culture system, which generates phenotypically distinct canine macrophage subsets and may serve as a useful in vitro platform for future studies of canine macrophage polarization and tumor-associated immune responses.

6. Small nucleolar RNA landscapes uncover ribosome-modification potential and immune-metabolic signatures in bovine subclinical mastitis.
PMID:日期:2026-09-09Subclinical mastitis (SCM) is a clinically silent inflammatory disorder that impairs udder health and milk productivity, yet how small nucleolar RNAs (snoRNAs) participate in epigenome-linked mammary remodeling remains poorly defined. We profiled snoRNAs in milk somatic cells from healthy and SCM-affected Vrindavani cows and integrated differentially expressed (DE) snoRNAs with matched methylome-transcriptome-derived differentially methylated expression genes (DMEGs). A catalogue of 2186 snoRNA loci was generated, and RNA-seq analysis identified 25 DE snoRNAs (FDR < 0.05, |log2FC= > 1), all showing increased abundance in SCM, including 19 C/D-box and 6 H/ACA-box snoRNAs. Computational target prediction identified 18 candidate H/ACA-box snoRNA-rRNA interactions consistent with pseudouridylation guidance and 47 candidate C/D-box interactions consistent with 2'-O-methylation guidance, predominantly involving 28S rRNA. Correlation-based integration of DE snoRNAs with DMEGs identified co-expression patterns associated with immune and metabolic processes. Positively correlated genes were enriched for leukocyte migration, granulocyte chemotaxis, cytokine/chemokine activity, immune receptor activity and MHC-associated host-defense pathways, whereas negatively correlated genes were enriched for oxoacid metabolism, glycolysis/gluconeogenesis, AMPK signaling, glycerolipid metabolism and peroxisome pathways. Together, these findings characterize SCM-associated snoRNA expression changes, predicted rRNA guide-target interactions and cross-layer associations with methylation-associated transcriptional programs, providing a hypothesis-generating framework for prioritizing snoRNAs for further experimental investigation.

7. Short communication: Th1 immunity is associated with Staphylococcus chromogenes intramammary infection.
PMID:日期:2026-09-08Staphylococcus chromogenes is one of the most prevalent non-aureus staphylococci associated with persistent intramammary infections (IMIs) in dairy cattle. However, the local cellular immune response to naturally occurring S. chromogenes IMI remains poorly characterized. Thus, this study aimed to assess milk T cell subsets, macrophages, neutrophils, and key intracellular cytokine production by milk T cells in mammary gland quarters naturally infected with S. chromogenes. Milk samples were used from non-infected and S. chromogenes-infected quarters from clinically healthy Holstein-Friesian dairy cows. The intensity of intracellular IFN-γ production by CD4 (P = 0.048) and CD8 (P = 0.035) T lymphocytes was higher in S. chromogenes-infected quarters than in non-infected controls, with a similar trend observed for γδ T cells (P = 0.06). Furthermore, the proportion of milk γδ T cells was higher in S. chromogenes-infected quarters (P = 0.05). Differences were observed neither in intracellular IL-17A production by T cell subsets nor in the proportions of milk CD4 and CD8 T lymphocytes, macrophages and neutrophils. Our findings identify T cell responses as a key component of the mammary immune response to S. chromogenes IMI and open new avenues for future studies investigating its potential in vivo protective response against major mastitis pathogens.

8. Preparation and evaluation of a monovalent foot and mouth disease vaccine as a rapid response to the emerging serotype SAT1 in Egypt.
PMID:日期:2026-09-04The recent emergence of foot and mouth disease virus (FMDV) serotype SAT1 in Egypt necessitated the development of a specific vaccine directed against the newly introduced serotype. The present study describes the preparation and evaluation of a monovalent inactivated SAT1 vaccine with potential for inclusion in national FMD control programs. Three SAT1 field isolates were adapted in BHK-21 cells through serial passage, after which an optimal master seed virus was selected based on reproducible and stable infectivity characteristics. The vaccine was formulated as a double oil emulsion using Montanide ISA 206. The vaccine potency was assessed in guinea pigs through determination of the protective dose (GPPD) following challenge with virulent guinea pig adapted virus that revealed satisfactory protective potency. The humoral immune response induced by such vaccine was subsequently evaluated in vaccinated cattle (2 mL dose), buffaloes (2 mL dose), and sheep (1 mL dose) through monitoring of their serum neutralizing antibody titers. Protective antibody levels were detected at 14 days in cattle and buffaloes and at 21 days in sheep post vaccination. The exhibited immunity had a duration extended to 32 weeks in cattle and buffaloes and to 28 weeks in sheep. These findings indicate that the prepared SAT1 vaccine was capable of inducing protective and sustained humoral immunity in the principal livestock species targeted within Egyptian vaccination campaigns and support its potential application in national FMD control programs.

9. Pharmacokinetics and safety of an engineered anti-feline PD-1 chimeric antibody following repeated intravenous and subcutaneous administration in healthy cats.
PMID:日期:2026-09-03Immune checkpoint inhibitors targeting programmed cell death-1 (PD-1) represent a promising strategy in feline oncology. We previously developed an anti-feline PD-1 monoclonal antibody and engineered an effector-silent variant, 1A1-2-fIgG-M2, to abolish cytotoxic functions. However, the safety profile and plasma pharmacokinetics (PK) of anti-feline PD-1 antibodies remain uncharacterized. This study aimed to evaluate the safety and PK profiles of 1A1-2-fIgG-M2 in healthy cats to support its clinical translation. Cats received 1A1-2-fIgG-M2 intravenously (IV; 3 or 10 mg/kg) or subcutaneously (SC; 3, 7, or 10 mg/kg) every 2 weeks for five doses, or saline as control (n = 3/group). Clinical signs, hematology, serum biochemistry, and necropsy findings were evaluated over 70 days. Anti-idiotype monoclonal antibodies were generated to establish an anti-idiotype antibody-based capture ELISA for plasma drug quantification. While IV groups displayed predictable elimination, SC groups exhibited marked PK variability at 3 and 10 mg/kg. Conversely, the 7 mg/kg SC group (SC-7) consistently maintained stable plasma concentrations (10-80 µg/mL). No mortality, severe adverse events, or acute infusion reactions were observed during the study period. Anti-idiotype ELISA provided robust PK quantification. Repeated SC administration at 7 mg/kg every 2 weeks maintained measurable plasma concentrations and was generally well tolerated in this small exploratory study. This regimen provides a foundation for clinical trials in cancer-bearing cats.

10. Immunostimulatory effects of dietary fucoidan from Padina boergesenii on growth, digestive performance, immune response and transcription of immune-related genes in Channa striata and disease resistance to Aeromonas hydrophila.
PMID:日期:2026-09-01The objective of this research was to evaluate the antibacterial properties against Aeromonas hydrophila and histological responses, immunological mechanisms, gene expression and disease resistance in Channa striata. The fish were separated into five groups and given meals with various amounts of fucoidan diet (0%, 0.5%, 1.5%, 2.5%, and 3.5%) for 45 days. The study's findings show that adding 2.5% fucoidan to the diet resulted in a significant improvement in the final weight, weight gain, and specific growth rate (SGR) of C. striata. Furthermore, the feed conversion ratio (FCR) decreased significantly fucoidan fed groups. The dietary intake of 2.5% fucoidan resulted in an elevation in the levels of amylase, protease, and lipase enzymes. The antibacterial activity results showed significant dose-dependent antibacterial activities. The metabolic enzymes aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) were significantly increased. The antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT) were also highly elevated during A. hydrophila infection and were modulated by fucoidan 2.5% supplementation in the diet. The supplementation of 2.5% fucoidan to fish diets enhanced non-specific immune responses such as Phagocytic activity, Nitroblue tetrazolium test (NBT), lysozyme activity, Complement C3, immunoglobulin (IgM) levels, and histological improvements after exposure to A. hydrophila. The fish-fed diets supplemented with fucoidan resulted in a significant increase (P < 0.05) in the mRNA expression of TNF-α and NF-κB in liver tissues. This study demonstrates that incorporating fucoidan to fish diets may promote growth, immunological and TNF-α and NF-κB increased, and resistance to A. hydrophila.
