1. Thai native chickens show improved growth performance and gut health following supplementation with Lactiplantibacillus plantarum-fermented noni juice.
PMID:日期:2026-11-01This study evaluated the dose-dependent effects of Lactiplantibacillus plantarum-fermented noni juice (OLP-FNJ) on growth performance and intestinal physiology in Thai native chickens (Surat Thani strain). A total of 240 birds were randomly allocated to four treatments (0, 2.5, 5.0, and 10.0 mL L OLP-FNJ administered via drinking water) in a completely randomized design and reared for 16 weeks. Growth performance, serum biochemical indices, digestive enzyme activities, intestinal morphology, tight junction-related and immune-related gene expression, and cecal microbiota composition were evaluated. Dose-response relationships were assessed using polynomial regression analysis. OLP-FNJ supplementation exerted a significant linear dose-response reduction in feed conversion ratio over the entire experimental period (3.11 vs. 2.70; linear p < 0.001) and significantly increased body weight gain during the starter phase (280.06 vs. 307.08 g; linear p < 0.05). The greatest numerical responses were observed at 10.0 mL L without adverse effects. Performance enhancement was associated with increased digestive enzyme activities, greater villus height-crypt depth ratios, upregulation of tight junction-related genes, and reduced expression of pro-inflammatory markers. Cecal microbial diversity increased in a dose-dependent manner, accompanied by shifts in dominant bacterial phyla and an elevated Firmicutes-to-Bacteroidetes ratio. Serum biochemical parameters remained within physiological ranges, indicating systemic safety. In conclusion, OLP-FNJ enhanced nutrient utilization efficiency in a dose-dependent manner and was associated with coordinated changes in digestive, structural, barrier, immune, and microbial functions. Fermented noni juice shows potential as a functional drinking-water additive to support sustainable production of Thai native chickens.

2. Acute effects of direct exposure to moxifloxacin on the porcine corneal endothelium: An ex vivo study.
PMID:日期:2026-11-01Intracameral administration of an ophthalmic solution of moxifloxacin has been proposed as a prophylactic strategy for the prevention of postoperative endophthalmitis following intraocular surgery, especially after phacoemulsification. The present study evaluated the acute effects of direct exposure to moxifloxacin on the porcine corneal endothelium using a worst-case ex vivo exposure model. A total of 48 porcine corneas, obtained from six-month-old male pigs, were allocated into three experimental groups: group A (control; n = 24) received balanced salt solution applied directly to the endothelial surface; while groups B (n = 12) and C (n = 12) were treated with a 0.25% or 0.5% ophthalmic solution of moxifloxacin, respectively. Scanning electron microscopy (SEM) was used to identify areas of endothelial cell loss, which were quantified using ImageJ software. Although exposure to 0.5% moxifloxacin resulted in significant endothelial morphological changes consistent with cellular damage, no acute endothelial injury was observed following exposure to 0.25% moxifloxacin. Compared to the other groups, the mean endothelial cell loss for Group C was 27.83%. Direct exposure of the corneal endothelium to moxifloxacin ophthalmic solution at concentrations ≥0.5% may induce endothelial toxicity. Therefore, during intracameral administration, care should be taken to avoid direct contact between moxifloxacin and the corneal endothelium in order to minimize potential endothelial damage.

3. Evaluation of machine learning models for predicting mastitis in dairy cows using dielectric properties and other milk quality parameters.
PMID:日期:2026-11-01Early and accurate detection of mastitis is essential for improving udder health, milk quality and dairy farm productivity. This study evaluated machine learning models for estimating somatic cell count (SCC) from physico-chemical milk parameters integrated with dielectric properties and for assigning SCC derived udder health categories. A total of 982 longitudinal milk samples obtained from 100 dairy cows at LUVAS cattle farm (HISAR, HARYANA) were analyzed. Model inputs comprised pH, electrical resistance, fat, protein, lactose, milk temperature, udder surface temperature, and dielectric properties measured at six microwave frequencies (54, 104, 254, 500, 915, and 2450 MHz). Experimentally measured SCC served as target variable and was not included among the input predictors. After prediction, measured and predicted SCC were grouped using thresholds of <2 × 10, 2 × 10-5 × 10 and > 5 × 10 cells/ml. Feed Forward Neural Network (FFNN), Support Vector Machine (SVM) and Multiple Linear Regression (MLR) models were developed and evaluated using mean square error (MSE), correlation coefficient (R), sensitivity, specificity, and area under the receiver operating characteristic curve (AUC). SVM demonstrated highest predictive performance (MSE = 2152.196, R = 0.98), followed by FFNN (MSE = 2447.60, R = 0.97), while MLR model exhibited lower accuracy (MSE = 13,231.630, R = 0.89). For SCC-based classification, FFNN achieved 100% sensitivity, 97.2% specificity and 0.986 AUC. SVM achieved 100% sensitivity, 98.7% specificity and 0.994 AUC, whereas MLR achieved 85.7% sensitivity, 98.6% specificity and 0.922 AUC. As repeated samples came from same cows, all samples were collected from one farm without external validation. These findings should be considered preliminary.

4. Anatomical study of the thorax and abdomen of the domestic cat: Correlation of cross-sectional anatomy with contrast-enhanced computed tomography and 3D reconstruction.
PMID:日期:2026-11-01Computed tomography (CT) has become an essential diagnostic and educational tool in veterinary medicine; however, accurate interpretation relies on precise knowledge of normal cross-sectional anatomy. This study aimed to develop a detailed anatomical atlas of the feline thorax and abdomen by correlating cross-sectional anatomical slices with contrast-enhanced CT images and three-dimensional (3D) reconstructions. Ten adult domestic cats of both sexes were used. Five cats were serially sectioned at defined thoracic and abdominal levels, and the sections were examined in both fresh and fixed states. The remaining five cats underwent plain and contrast-enhanced CT examinations following intravenous administration of iodinated contrast medium. CT datasets were converted to DICOM format and used to generate 3D reconstructions of the thoracic skeleton, heart, major vessels, and abdominal viscera. All major bones, muscles, visceral organs, and vascular structures of the thorax and abdomen were successfully identified and correlated between anatomical sections and CT images. Contrast-enhanced CT markedly improved visualization of vascular anatomy and visceral relationships. The reconstructed anatomical, radiological, and 3D reconstruction atlas provides a reliable reference for veterinary anatomy education, diagnostic imaging interpretation, and surgical planning in cats.

5. Characterization of eukaryotic communities in small ruminant bile shows substantial extra-intestinal location of Cryptosporidium parvum.
PMID:日期:2026-11-01Next-generation sequencing has expanded the study of microbial communities, yet the eukaryotic fraction of host-associated microbiomes remains poorly characterized, particularly in non-intestinal matrices. This study aimed to assess eukaryotic community diversity in bile from small ruminants. A total of 400 bile samples from sheep and goats collected at a Portuguese slaughterhouse were analyzed using 18S rRNA PCR and long-read Oxford Nanopore sequencing on 80 pooled samples. Overall, 12.5% (10/80) of pools were found to have eukaryotic elements, and Cryptosporidium-assigned reads were detected in 30% (3/10) of these, all originating from adult sheep. These findings provide molecular evidence of C. parvum in bile, extending its known tissue distribution beyond the gastrointestinal tract, suggesting potential chronic or subclinical biliary carriage and expanding its recognized tissue tropism. This work highlights the utility of long-read 18S rRNA metabarcoding for detecting protozoan parasites in unconventional biological matrices and characterizing eukaryotic diversity in livestock systems.

6. Comparative cytotoxic and transcriptional effects of methanol, ethanol, and DMSO in the CMT-U27 canine mammary carcinoma cell line.
PMID:日期:2026-11-01Organic solvents such as dimethyl sulfoxide (DMSO), ethanol, and methanol are widely used as vehicles in in vitro oncology research, yet their inherent toxicities can confound experimental outcomes. This study systematically evaluates the comparative cytotoxicity and transcriptional signatures of cell death pathways induced by these solvents in the canine mammary carcinoma cell line (CMTU27). Cells were treated with concentrations from 0.25% to 5% for 24, 48, and 72 h. Viability was assessed using the XTT assay, and cell death modes were characterized using Hoechst 33342/Annexin V/PI triple-fluorescence staining. Transcriptional alterations of apoptotic (CASP3, BAX, PUMA, ATM, BID), anti-apoptotic (BCL2, MCL1), and DNA damage/cell cycle (P53, P21, NOXA) genes were quantified via RT-qPCR. DMSO exhibited significant time- and dose-dependent cytotoxicity. No 50% viability concentration was reached after 24 h, whereas linear interpolation yielded approximate concentrations of 4.34% and 3.67% corresponding to 50% viability after 48 and 72 h, respectively. Fluorescent imaging revealed staining patterns consistent with apoptosis and secondary necrosis. Transcriptionally, DMSO markedly upregulated CASP3, BAX, PUMA, and ATM and downregulated MCL1, while BID and BCL2 showed concentration-dependent but not consistently significant changes. Conversely, ethanol and methanol maintained viability above 80% up to 5% (IC not calculated). PCA modeling revealed distinct spatial dissociation: ethanol predominantly induced genotoxic stress, strongly correlating with the P53, P21, and NOXA vectors, whereas methanol clustered closest to the control, despite showing significant gene-specific transcriptional alterations. In conclusion, our findings suggest that DMSO concentrations should be kept below 1% in CMT-U27 assays to minimize off-target apoptotic effects. Although ethanol showed limited cytotoxicity, it was associated with DNA damage-related transcriptional responses. Methanol exhibited the most favorable cytotoxicity profile among the tested solvents, although significant gene-specific transcriptional responses were observed, suggesting that its suitability as a vehicle solvent for CMT-U27-based in vitro studies should be evaluated according to the experimental endpoint.

7. Characterization of primary three-dimensional cell culture from spontaneous canine tumors.
PMID:日期:2026-11-01This study aimed to generate primary three-dimensional (3D) cell cultures derived from spontaneous canine tumors and to compare two culture systems: a ready-to-use bioreactor (VITVO®) and a miniaturized plate-based system (MPBS). A total of 27 tumors, including mainly hemangiosarcomas, oral melanomas, urothelial carcinomas, and osteosarcomas, were collected from three Italian veterinary hospitals. Fresh samples were used to establish 3D cell cultures and to perform histological and immunohistochemical analyses for phenotypic characterization and comparison with primary tumors. Tissue dissociation proved to be a critical step. A manual protocol combining mechanical and enzymatic methods significantly improved cell viability and cellularity compared to automated dissociation, which yielded fewer viable single cells and limited cell survival. Both culture systems successfully supported 3D cell cultures using the same scaffold material. The plate-based system was particularly suitable for small biopsy samples without compromising cellularity, whereas VITVO® was more appropriate for larger tissue specimens. An inverse correlation was observed between the extent of necrosis in primary tumors and the cellularity of the corresponding 3D cell cultures. Immunohistochemical analyses confirmed that most 3D cell cultures retained the phenotypic characteristics of the original tumors, while osteosarcoma models exhibited limited cell maintenance, likely due to stromal predominance. Overall, these findings highlight the importance of optimizing dissociation protocols, scaffold size, and culture conditions to preserve tumor heterogeneity and viability. This workflow provides a platform for generating 3D models with preserved morphological and immunophenotypic concordance with the primary tumors.

8. Histological differences in the placenta of three mares treated for ascending bacterial placentitis compared to physiological pregnancies.
PMID:日期:2026-11-01Ascending bacterial placentitis is a frequent cause of late pregnancy loss in mares. The histology of placentitis has been described but mainly with a focus on inflammatory changes. In this study, we assessed several aspects beyond inflammatory processes between mares foaling close to term during treatment for placentitis and physiological pregnancies. We included placental tissue from physiological term pregnancies (group PP; n = 8) and clinical cases with confirmed ascending bacterial placentitis (group PL; n = 3). Samples were collected from several locations of the allantochorion (cervical star, base, and tip of both uterine horns). Immunostaining was performed for caspase 3 (apoptosis), Ki67 (proliferation), and MAC387 (macrophages); histochemical staining was made with PAS (Periodic acid Schiff, blood vessels) and Kossa stain (microcalcification). Allantochorionic cell proliferation was increased (p < 0.05) in PL mares compared to PP mares at several locations; no group differences in caspase 3 indicating apoptotic activity were detected. Whereas the blood vessel area in the allantochorion was increased in PP mares in some locations, a larger number of blood vessels (p < 0.05) was determined at the base of the pregnant horn in PL mares. In conclusion, bacterial ascending placentitis was characterized by tissue proliferation and vascularization. This might suggest compensatory mechanisms that were induced because of tissue function loss due to inflammation.

10. Outer membrane vesicles in Pseudomonas aeruginosa pathogenesis: a systematic review of virulence dissemination, immune modulation, and antimicrobial resistance, with perspectives for veterinary and one health practice.
PMID:日期:2026-11-01Pseudomonas aeruginosa is an opportunistic pathogen recognised for its intrinsic and acquired antibiotic resistance, its capacity to establish chronic infections in cystic fibrosis (CF) airways, and its clinical importance in immunocompromised human and animal hosts. While its extensive secretome has been intensively studied, a mechanistically distinct dissemination strategy has emerged: the constitutive production of outer membrane vesicles (OMVs) that concentrate, protect, and deliver virulence cargo to host cells beyond direct bacterial contact. This systematic review evaluates the molecular mechanisms, biological consequences, antimicrobial resistance implications, and therapeutic prospects associated with OMV-mediated virulence in P. aeruginosa infection, and additionally considers the veterinary and One Health relevance of this evidence base. A systematic search of PubMed/MEDLINE, Scopus, Web of Science, and EMBASE was conducted from database inception to January 2025 following PRISMA 2020 guidelines. Studies investigating OMV production, cargo characterisation, or biological activity in the context of P. aeruginosa infection were eligible. Risk of bias was assessed using the ToxRTool (in vitro) and SYRCLE tool (in vivo). Inter-rater agreement was quantified by Cohen's kappa (kappa = 0.82). Fifty-eight studies (predominantly in vitro and murine models) met the inclusion criteria. P. aeruginosa OMVs (50-250 nm) carry biologically active type III secretion effectors (ExoS, ExoU), elastase (LasB), alkaline protease (AprA), pyocyanin, quorum-sensing autoinducers (3-oxo-C12-HSL), Hcp1, siderophores, and beta-lactamase enzymes including VIM and OXA-type carbapenemases. OMV-packaged beta-lactamases were shown, principally in co-culture experiments, to confer transferable resistance by degrading beta-lactam antibiotics in the extracellular milieu, shielding susceptible bystander bacteria. OMVs drive biofilm maturation, suppress CF airway innate immunity, and induce TLR4-mediated hyperinflammation. OMV output was consistently increased under antibiotic selection pressure, in the CF mucus environment, and during quorum-sensing activation, although the magnitude of increase varied considerably across experimental conditions and should be interpreted qualitatively rather than as directly comparable pooled estimates. Therapeutic strategies targeting OMV biogenesis, cargo neutralisation, and OMV-based vaccine platforms are evaluated, and remain at a predominantly preclinical stage of development. P. aeruginosa OMVs constitute a versatile virulence amplification platform that contributes to tissue destruction, immune evasion, biofilm consolidation, and antibiotic resistance dissemination. Disrupting this multi-functional delivery system represents a promising, though still largely preclinical, antibiotic-independent therapeutic direction for managing refractory P. aeruginosa infections, particularly in CF and critical care settings. Extension of this evidence base to animal-associated P. aeruginosa infections and One Health surveillance is identified as an important priority for future research.
