1. GnRH and PGF2α administered at the time of AI differently affect conception rates of non-estrous Nelore cows according to Body Condition Score.
PMID:日期:2026-10-03This study evaluated the effects of GnRH, PGF2α, and their combination at the time of fixed-time artificial insemination (timed-AI) on pregnancy per AI (P/AI) and pregnancy loss (PL) in non-estrous suckled multiparous Nelore cows. Of 8061 cows submitted to a timed-AI protocol, those expressing estrus (n = 5400) served as a positive control, while non-estrous cows (n = 2661) were randomly assigned to saline solution (Control; n = 791), GnRH (n = 656), PGF2α (n = 667), or PGF2α+GnRH (n = 547) at AI. Pregnancy per AI was diagnosed on Day 41 (D41) and PL assessed on D71. Pregnancy per AI was greater (P < 0.0001) in estrous (59%) than non-estrous cows (34%), and among non-estrous cows a treatment × BCS interaction was detected (P < 0.0001). In cows with BCS < 2.75, PGF2α (34%) and PGF2α+GnRH (31%) achieved greater P/AI (P = 0.006) than GnRH (18%). Conversely, in cows with BCS ≥ 2.75, GnRH achieved the highest P/AI (49%), exceeding PGF2α (26%) and PGF2α+GnRH (28%; P < 0.0001), though not significantly different from Control (41%; P = 0.053). Overall PL was lower (P = 0.042) in the PGF2α+GnRH group (2.5%) than Control (9%). Integrating P/AI and PL outcomes, the final conception rates at D71 demonstrate that BCS influenced the success of the treatment administered at the time of AI. GnRH improved fertility of cows with BCS ≥ 2.75 (Control: 38%, GnRH: 47%, PGF2α: 25%; PGF2α+GnRH: 28%; P < 0.0001) while PGF2α increased pregnancy of cows with BCS <2.75 (Control: 16%, GnRH: 17%, PGF2α: 32%, PGF2α+GnRH: 30%; P < 0.0001). In conclusion, the effectiveness of hormonal treatment at AI in non-estrous Nelore cows depends on BCS: GnRH improved fertility in cows with adequate body condition (BCS ≥ 2.75), while PGF2α-based treatments were more beneficial in cows with poor nutritional status (BCS < 2.75).

2. The corpus luteum transcriptome is dysregulated in dairy cows with mastitis.
PMID:日期:2026-10-03Mastitis-induced inflammation in dairy cows can trigger premature luteolysis and subsequent pregnancy loss; however, the molecular mechanisms underlying these effects remain poorly understood. This study aimed to characterize changes in gene expression in the corpus luteum of cows with clinical or subclinical mastitis compared with healthy cows. Multiparous pregnant cows from a commercial pasture-based dairy herd were classified into three groups: subclinical mastitis (SCM; n = 6), clinical mastitis (CLM; n = 6), and healthy controls (CON; n = 5), based on California Mastitis Test scores and somatic cell counts. Corpus luteum biopsies were collected at pregnancy diagnosis, between 28 and 42 days post-insemination, using a transvaginal ultrasound-guided technique and subjected to RNA sequencing. Differential expression analysis identified 428 differentially expressed genes (DEG) between CLM and CON cows (263 upregulated and 165 downregulated), 186 between SCM and CON cows (76 upregulated and 110 downregulated), and 391 between CLM and SCM cows (236 upregulated and 155 downregulated; FDR < 0.05). Functional enrichment of DEG between CLM and CON showed dysregulation of pathways related to immune response, cortisol, GnRH, and oxytocin signaling. These transcriptional changes may reflect an adaptive luteoprotective response aimed at maintaining luteal function under systemic inflammatory stress. Overall, the results demonstrate that mastitis, particularly in its clinical form, markedly alters the luteal transcriptome during early pregnancy, potentially affecting corpus luteum function and contributing to an increased risk of pregnancy loss.

3. Factors associated with pregnancy following transfer of Japanese Black embryos into Japanese Black, Holstein, and crossbred heifers.
PMID:日期:2026-10-01We investigated factors associated with pregnancy approximately 60 days after transfer of Japanese Black (JB) embryos into JB, Holstein (HO), and their crossbred (JB×HO) heifers. A total of 26,196 embryo transfer (ET) records collected between 2008 and 2023 were analyzed. Recipient-, embryo-, and donor-related factors were initially evaluated using Akaike information criterion (AIC)-based variable selection, and the retained fixed-effect structure was subsequently evaluated in a mixed-effects logistic regression model with donor as a random intercept. The overall pregnancy rate was 70.7%. Pregnancy rates for JB, HO, and JB×HO recipients were 62.0%, 71.1%, and 71.6%, respectively. Embryo quality, embryo type, ET technician, farm, and ET attempt number showed prominent associations with pregnancy. Donor-related associations were statistically detectable but small. Embryo stage was retained in the AIC-selected model, but its overall effect was not statistically significant in the likelihood-ratio test (P = 0.1222). These findings indicate that embryo- and recipient-related factors were more strongly associated with pregnancy rate than donor superovulatory response traits.

4. Singleton freemartinism in cattle: A structured narrative review and historical case reappraisal.
PMID:日期:2026-10-01Freemartinism classically affects a female calf gestated with a male co-twin. A female may be born as an apparent singleton when a male co-conceptus is lost after placental vascular communication has permitted cellular exchange and exposure to testicular products. This structured narrative review and historical case reappraisal evaluates evidence for singleton freemartinism, clarifies diagnostic boundaries and reassesses a 1995 case following multiple-embryo transfer. PubMed, Scopus and FAO AGRIS were searched through 14 September 2026, supplemented by citation searching. Evidence supports the existence of singleton freemartinism but does not establish its prevalence. An apparent singleton birth does not demonstrate that gestation was singleton throughout. In an appropriate developmental context, persistent XX/XY blood chimerism, the coexistence of XX and XY blood-cell populations, is evidence that fetal cell exchange occurred. Detection of a Y-linked sequence establishes only that target DNA is present in the specimen. Neither finding alone demonstrates male-type reproductive-tract development or pathological freemartinism. Diagnosis therefore requires integration of gestational history, characterized chromosome-based or molecular findings, reproductive anatomy, functional findings and histopathology. Archival sections show ovarian follicles and testis-like tubular structures in the same gonad. Interpretation is limited by the unknown sex of the lost embryo, incomplete gonadal examination, lack of cell-level localization and cell-type identification, and unknown fertility. Twinning, multiple-embryo transfer and conceptus loss may increase opportunities for the condition to arise, but evidence demonstrates neither increased prevalence nor a quantitative contribution to reproductive failure. Prospective multicenter studies linking gestational records with molecular, anatomical and functional outcomes are required.

5. Melatonin synergizes with constant FSH stimulation to improve oocyte yield and in vitro embryo production in dairy goats.
PMID:日期:2026-09-29Ovum Pick-Up (OPU) enables efficient, repeated oocyte retrieval from elite dairy goats, making it a critical technology for accelerating genetic progress through in vitro embryo production (IVEP) in dairy goat breeding. However, the limited quality and quantity of retrieved oocytes often restrict its overall efficiency. Melatonin (MT) has been known to regulate mammalian reproductive physiology and the ovarian microenvironment, but its specific effects across different follicle-stimulating hormone (FSH) superovulation protocols to enhance oocyte yield and quality remain unclear. This study investigated the effects of MT supplementation (20 mg/dose, total 120 mg/goat across 6 injections) on oocyte recovery in dairy goats subjected to three different FSH injection protocols (incremental, constant, and decremental). Following in vitro fertilization, embryo development and pregnancy rates after embryo transfer (ET) were subsequently evaluated. MT supplementation significantly increased the total number of recovered oocytes (P < 0.05), with the constant FSH protocol yielding optimal outcomes. MT increased blastocyst rates (P < 0.05), although the improvement of cleavage and fresh ET pregnancy rates were not significant. MT elevated both estrogen and progesterone levels in the serum of dairy goats (P < 0.05). Further researches to determine the mechanism of melatonin effects was performed by evaluation of serum hormones and oxidative stress related molecule, follicular fluid metabolomics, and ovarian transcriptomics. Multi-omics analysis revealed that MT significantly altered the follicular fluid metabolic profile, particularly affecting antioxidant and steroid hormone metabolism (Epigallocatechin, Quercetin 3-O-Xylosyl-Rutinoside, Hexahydrocurcumin, Protocatechuic Aldehyde, Isodityrosine, Pregnanediol, 16-Ketoestradiol), and regulated antioxidant and steroid hormone-related genes (GSTM1, MGST2, RXFP1, 3βHSD, HSD17B7, and PAQR7). Furthermore, ovarian redox homeostasis was enhanced by increasing glutathione peroxidase (GSH-Px) activity and reducing malondialdehyde (MDA) levels (P < 0.05). Overall, MT exerts multidimensional beneficial effects on ovarian microenvironment to enhance oocyte developmental competence. Combining a constant FSH protocol with MT achieves optimal superovulation efficiency, providing a robust strategy for advancing overall IVEP efficiency in dairy goat.

6. Effects of cordycepin on antioxidant capacity and proliferative potential of buffalo spermatogonial cells in vitro.
PMID:日期:2026-09-29Buffalo spermatogonial cells (bSCs) are essential for maintaining spermatogenesis and male fertility. Oxidative stress impairs bSCs proliferation through ROS-induced damage and apoptosis. Cordycepin, a bioactive compound from Cordyceps militaris, exhibits potent antioxidant effects but has not been systematically evaluated in bSCs. This study aimed to investigate the effects of cordycepin(0-5 μM) on the in vitro growth and survival of buffalo spermatogonial cells (bSCs) and to characterize associated changes in redox status and mitochondrial function.Cell proliferation was assessed by cell counting, and cellular metabolic activity was evaluated by CCK-8 assay. Proliferation markers (PCNA, ETV5, CDC25B) and reproductive markers (PGP9.5, DDX4, NANOS2, OCT4) were analyzed by qPCR and Western blotting. Antioxidant enzyme activities (SOD, CAT, GSH-Px) and oxidative stress markers (MDA) were measured by ELISA. Intracellular ROS, apoptosis, mitochondrial membrane potential, and mitochondrial dynamics proteins (MFN-1, OPA1, DRP-1, FIS-1) were assessed by DCFH-DA staining, flow cytometry, JC-1 probe, and immunofluorescence/Western blotting, respectively. At 1.25 μM, cordycepin significantly increased cell proliferation and upregulated PCNA and ETV5 expression. Cordycepin reduced intracellular ROS, enhanced antioxidant enzyme activities, and decreased MDA levels. Treatment increased JC-1 fluorescence intensity and was associated with selective mRNA- and protein-level changes in mitochondrial dynamics-associated factors; however, these data did not directly assess mitochondrial morphology or establish altered fusion/fission activity. Cordycepin decreased apoptosis and was associated with increased expression of selected germ cell- and spermatogonial cell-associated markers. At 1.25 μM, cordycepin treatment was associated with increased bSCs number, reduced intracellular ROS accumulation, enhanced antioxidant enzyme activities, altered mitochondrial membrane potential, reduced apoptosis, and increased expression of spermatogonial cell-associated markers. These findings support the potential utility of cordycepin as a culture supplement for improving the in vitro maintenance and expansion of bSCs.

7. Effect of domperidone and/or oxytocin on milk bromatological composition in bitches with hypogalactia after elective caesarean section.
PMID:日期:2026-09-28Lactational disorders significantly increase puppy mortality, due to delayed colostrum intake and suboptimal nutrition to fulfill growth and development. Hypogalactia is a common consequence of c-section, even at term, and requires treatment to ensure adequate puppies' nutritional requirement. For such purpose, galactagogues can be used to stimulate synthesis of mammary secretion and/or induce its ejection. This study aimed to perform a bromatological analysis of colostrum and milk from bitches with post-surgical hypogalactia treated with domperidone to stimulate lactogenesis and/or oxytocin to increase milk withdraw. Twenty bitches diagnosed with hypogalactia immediately after c-section were enrolled and assigned to four experimental groups of treatment: Oxytocin (n = 5; 0.1 IU/kg within 100 mL of saline solution, IV, BID for 3 days), Domperidone (n = 5; 1 mg/kg, PO, BID for 3 days), Oxytocin + Domperidone (n = 5), and Control (n = 5; normogalactia bitches). Milk was collected at 0h (before treatment), 24h, 3d, 7d, and 21d postpartum and subjected to bromatological analysis. In Domperidone Group, percentage and concentration of total solids and milk protein decreased from pre-treatment to day 3 postpartum, while carbohydrate concentration increased. Oxytocin-treated group showed decrease in total solids, but higher lipid content from day 3 onward. Milk carbohydrate and lipid concentrations increased on day 3 and day 7 postpartum, respectively, in bitches treated with both domperidone and oxytocin. In conclusion, treatment with domperidone and/or oxytocin in bitches with hypogalactia does not compromise colostrum composition, albeit milk protein, fat and carbohydrate content varied further in lactation according to the treatment protocol, comparable to that of bitches with normal lactation.

8. Use of plant-derived compounds for chilled gamete storage in felids and canids.
PMID:日期:2026-09-28Wild felids and canids play important roles in maintaining ecosystem stability and biodiversity, yet many species are threatened by habitat loss, climate change, urbanization, and poaching. Assisted reproductive technologies (ARTs) have therefore become valuable tools for wildlife conservation and genetic resource banking. However, prolonged storage and post-mortem deterioration remain major limitations affecting gamete quality and fertilizing ability. Oxidative stress is a major cause of gamete deterioration during chilled storage. During chilled storage, excessive reactive oxygen species (ROS) production can induce membrane damage, mitochondrial dysfunction, DNA fragmentation, apoptosis, and impaired fertilization capacity. Because endogenous antioxidant defenses progressively decline during chilled storage, supplementation with exogenous antioxidants has gained increasing attention for improving gamete survival during storage. This review summarizes the potential applications of plant-derived antioxidants in chilled storage of reproductive tissues and cells, focusing on chamuangone from Garcinia cowa Roxb. (Cha Muang), green tea polyphenols, and curcumin from Curcuma longa. These compounds have demonstrated beneficial effects on sperm motility, viability, membrane integrity, oocyte maturation, and fertilization-related biomarkers during chilled storage of feline and canine reproductive tissues. Antioxidant supplementation has also been associated with storage of IZUMO1 and integrin β5 expression together with reduced apoptosis-related markers such as cleaved caspase-3. Collectively, plant-derived antioxidants may improve short-term chilled storage of gamete through antioxidative, anti-apoptotic, and membrane-stabilizing mechanisms, highlighting their potential applications in reproductive biotechnology and conservation programs.

9. CATH-1 supplementation improves sperm quality and fertility of rooster semen during cold storage by reducing bacterial contamination.
PMID:日期:2026-09-28The present study investigated the effects of antibiotic and antimicrobial peptide supplementation on semen quality, viable bacterial load, and fertility potential in Thai native chickens during cold storage. Semen samples were diluted in an extender supplemented with kanamycin (31.20 μg/mL) or CATH-1 (3.75 or 7.50 μg/mL) and stored at 5 °C for up to 72 h. Sperm quality parameters, viable bacterial load, and reproductive outcomes following artificial insemination were assessed at 0, 24, 48, and 72 h. Storage duration significantly affected all sperm quality parameters (P < 0.001), while fertility and hatchability were progressively lower across the sequential storage-time phases. Kanamycin and CATH-1 at 3.75 μg/mL consistently maintained higher sperm quality and reduced susceptibility to lipid peroxidation compared with the control (P < 0.05), with no significant difference between the two treatments. Both treatments effectively suppressed bacterial proliferation; however, viable bacterial load increased progressively beyond 24 h in all groups. Scanning electron microscopy revealed marked bacterial surface alterations following CATH-1 treatment, consistent with membrane-disruptive antibacterial activity. In contrast, CATH-1 at 7.50 μg/mL compromised sperm quality and reproductive outcomes from the initial time point onward, despite achieving the greatest bacterial suppression among all treatments. Across the sequential storage-time phases, fertility potential was progressively lower, although the kanamycin and CATH-1 (3.75 μg/mL) groups maintained higher fertility than the control during prolonged storage. These findings demonstrate a dose-dependent dual effect of CATH-1, with 3.75 μg/mL performing comparably to kanamycin and representing a promising antibiotic alternative in the context of One Health concerns in poultry artificial insemination programs.

10. Zygote origin shapes sensitivity to electrical voltage field and editing susceptibility after CRISPR/Cas9-RNP electroporation in bovine embryos.
PMID:日期:2026-09-26Despite major progress in generation of edited embryos and offspring via CRISPR/Cas9-RNP electroporation, editing outcomes remain highly variable. This variability may be due to intrinsic differences between in vivo-derived (IVV) and in vitro-derived (IVT) zygotes, which may play a more decisive role than previously anticipated. Therefore, this study aimed to determine how zygote origin shapes the sensitivity to electrical voltage field and editing susceptibility after CRISPR/Cas9-RNP electroporation in the bovine model. The present study systematically compared subsequent developmental competence and editing events in IVV- and IVT-derived bovine zygotes across defined electroporation voltages and pulse durations. Notably, CRISPR/Cas9-RNP electroporation of IVV-derived bovine zygotes of comparable age and collected by tubal embryo flushing (10-18 h post ovulation, hpo) resulted in markedly lower survival rates at 25 and 30 V compared to 20 V. In contrast, higher (p < 0.05) editing frequencies were observed at 25 V and 30 V. Clear differences, however, were observed between the two zygote groups. Following CRISPR/Cas9 RNP electroporation, IVV-derived zygotes yielded a higher proportion of unedited blastocysts (p < 0.05), absence of full edited embryos, a significantly increased frequency of monoallelic edits and reduced rates of mosaicism compared to IVT-derived zygotes. Collectively, the results of the present study clearly underscore a significant lower editing efficiency to pulse sensitivity ratio in IVV-derived zygotes compared to their IVT counterparts. To our knowledge, this is the first study to use bovine IVV-derived zygotes for genome editing using RNP-electroporation to elucidate how zygote origin shapes sensitivity to electrical voltage field and editing efficiency.
