1. Cur-EGCG-Fe coordination nanoparticle-functionalized gelatin hydrogel biointerfaces for antibacterial and inflammatory microenvironment-regulatory wound repair.
1. 用于抗菌和炎症微环境调控伤口修复的Cur-EGCG-Fe配位纳米颗粒功能化明胶水凝胶生物界面PMID:日期:2026-10-02Bacterial colonization, oxidative stress, and persistent inflammation jointly impair infected wound healing. Here, curcumin (Cur)-containing epigallocatechin-3-gallate (EGCG)-Fe coordination nanoparticles (CEF NPs) were incorporated into gelatin through Fe-mediated interactions. The resulting hydrogels (Gel/CEF) exhibited injectability, self-healing behavior, a porous structure, and favorable hemocompatibility and cytocompatibility. Under 808 nm near-infrared (NIR) irradiation, Gel/CEF3 generated heat and reduced viable Staphylococcus aureus and Escherichia coli in vitro. Gel/CEF3 extracts decreased intracellular reactive oxygen species and lipid peroxidation in HO-challenged fibroblasts. They also preserved superoxide dismutase activity and improved cell migration. In lipopolysaccharide-stimulated macrophages, Gel/CEF3 extracts decreased the proportion of CD86-positive cells and increased CD206-positive cells, supporting a shift toward an M2-like phenotype. Transcriptomic analysis revealed downregulation of TNF and NOD-like receptor signaling-associated gene sets. Protein analyses further showed reduced levels of NLRP3, ASC, IL-1β, and GSDMD, supporting attenuation of an NLRP3-associated inflammatory profile. In S. aureus-infected full-thickness wounds, Gel/CEF3 reduced culturable bacterial burden during the early treatment phase and accelerated wound closure, re-epithelialization, collagen deposition, and angiogenesis. These effects were more pronounced with NIR treatment and were accompanied by improvements in local oxidative stress and inflammatory markers. Major-organ histology and serum biochemical assessments revealed no apparent systemic toxicity at day 14. These findings support Gel/CEF as a metal-phenolic hydrogel platform that combines NIR-responsive antibacterial activity with redox regulation and macrophage-associated inflammatory modulation for S. aureus-infected wound repair.

2. A biotin-functionalized calix[4]arene nanocarrier with intrinsic antitumor activity for combined chemo-photodynamic therapy.
To address these issues, herein we rationally design and construct a novel calix[4]arene derivate CMP, whose upper rim is functionalized with pH-responsive biotin-PEG hydrophilic chains, and lower rim is dually modified with hydrophobic alkyl chains and antitumor pharmacophores. This "all-in-one" supramolecule can not only serve as an excellent supramolecular chemotherapeutic agent itself, but also act as a versatile carrier to co-deliver doxorubicin (DOX) and chlorin e6 (Ce6). The nanoplatform therefore integrates two types of chemotherapeutic modalities and biotin-targeted photodynamic therapy for multi-modal tumor treatment. Notably, in vivo experiments demonstrate that the resultant DOX@CMP@Ce6 nanosystem achieves a 90.5% tumor growth inhibition rate, while displaying biotin-mediated tumor tropism, excellent biocompatibility, and robust triple-modality combinatorial therapeutic effects. Collectively, this work provides key insights into the rational construction of multifunctional supramolecular platforms, reinforcing the broad prospects of calixarene derivatives for nanomedical applications.

3. Antineoplastic ultrasmall Zn-doped Prussian blue mediating catalytic therapy and endogenous molecular pool modulation for tumor microenvironment regulation.
3. 抗肿瘤超小锌掺杂普鲁士蓝介导催化治疗与内源性分子池调节用于肿瘤微环境调控PMID:日期:2026-10-01Developing novel nanomedicines based on clinically approved agents has emerged as a promising anticancer therapeutic strategy. Nevertheless, the tailored optimization of clinically approved Prussian blue (PB) via Zn doping has yet to attract sufficient attention, particularly its capacity to intervene in the tumor microenvironment. Herein, we employed doping engineering to modulate the structural and functional properties of PB nanoparticles (NPs), with results revealing that zinc doping possessed intrinsic advantages over Co and Mn in narrowing size distribution and generating ultrasmall nanostructures. In addition, these ultrasmall Zn-doped PB NPs displayed outstanding tumor theranostic performance, featuring boosted enzyme-mimetic catalytic activity, favorable MRI imaging capacity, and diverse bioactivities. By catalyzing HO substrates and regulating endogenous molecular pools, they could remodel the breast tumor microenvironment and trigger apoptosis and ferroptosis. This study offers new inspirations for the rational design of metal-doped nanomaterials and delivers valuable strategies to optimize clinically approved therapeutic agents.

4. Bioactive fucoidan-coated polylactic acid-based nanoparticles for co-delivery of vincristine and thymoquinone: Enhanced antitumor efficacy with reduced vincristine-associated toxicity.
4. 生物活性岩藻多糖包覆的聚乳酸基纳米颗粒用于长春新碱和百里醌的共递送:增强抗肿瘤疗效并降低长春新碱相关毒性PMID:日期:2026-09-30The clinical efficacy of vincristine (VIN) is often restricted by dose-limiting toxicity, poor tumor selectivity, and rapid systemic distribution. Another biomolecule, thymoquinone (TQ), has negligible toxicity, but poor aqueous solubility and limited bioavailability hamper its use in cancer therapy. In this study, a fucoidan (Fu)-based methoxy-poly (ethylene glycol)-block-poly (lactic acid) (mPEG-PLA) nanoformulation has been optimized for the co-delivery of VIN & TQ to synergistically enhance cancer therapy. Besides acting as a surface stabilizer, Fu provides anticancer therapeutic responses owing to its intrinsic anti-cancer, anti-inflammatory, and tumor-targeting properties, thereby enhancing the overall therapeutic efficacy of the nanoformulation. Molecular docking was first performed to investigate the drug and the polymer interactions, which revealed several favourable bindings of both the biomolecules with the hydrophobic domain of the mPEG-PLA polymer. Formed Fu-based co-loaded nanoparticles (VIN-TQ-PLA-CN-Fu-NPs) exhibited narrow size distribution, high drug entrapment efficiency, slow and sustained pH-responsive drug release. In-vitro studies using the triple negative breast cancer (TNBC) cell line showed excellent cellular uptake, cytotoxicity, apoptosis, and inhibition of tumor proliferation using the Calcein-AM/PI live/dead assay, JC-1 assay, and DCFDH-DA reactive oxygen species (ROS) generation assay compared to free drugs. In-vivo experiments further confirmed remarkable tumor growth inhibition, achieving 90.2% tumor regression, prolonged survival periods, while exhibiting excellent biocompatibility without obvious changes to vital organs, probably due to toxicity-alleviating properties of TQ. Collectively, the combination of VIN, TQ, and Fu in a single nanoformulation produced a multitargeted synergistic therapeutic system for combination cancer chemotherapy and offers significant potential for use in clinical settings.

5. Bioinspired Fe-polyphenol nanozymes: harnessing natural coordination assembly for biomedical applications.
5. 仿生铁-多酚纳米酶:利用天然配位组装实现生物医学应用PMID:日期:2026-09-30Fe-polyphenol nanozymes are assembled from natural polyphenols and Fe ions via coordination. They combine polyphenol bioactivity with iron based enzyme like catalysis. This review covers their composition, classification, performance, and applications. Based on polyphenol types, we summarize the morphology, coordination mechanism, iron valence, and loading capacity of systems including tannic acid, curcumin, quercetin, gallic acid, and polydopamine. These nanozymes exhibit multiple enzyme like activities: peroxidase like, superoxide dismutase like, and catalase like. They achieve targeted lesion delivery via passive or active targeting and respond to pH, enzymes, or substrates. In applications, Fe-polyphenol nanozymes improve tumor therapy through cascade catalysis, multimodal synergy, and hypoxia relief. They reduce inflammation by scavenging reactive oxygen species and modulating immune balance. They enable simultaneous bacterial killing and tissue regeneration in antimicrobial and wound repair settings. They also show high sensitivity in sensing and detection. Finally, we discuss future directions in standardized evaluation, AI-assisted design, in vivo mechanism, and clinical translation.

6. Nicotinium-based surface-active ionic liquids as antimicrobial surfactants: Micellization properties, antibiofilm activity, and biocompatibility.
6. 尼古丁基表面活性离子液体作为抗菌表面活性剂:胶束化性质、抗生物膜活性及生物相容性PMID:日期:2026-09-30A new family of nicotinium-based surface-active ionic liquids (CₙNicBr, n = 10-16) was synthesized by N-alkylation of nicotine and characterized by NMR, MS and thermogravimetry (stable to 215 °C). Aqueous association, assessed by conductimetry, fluorescence, tensiometry and dynamic light scattering, gave critical micelle concentrations of 0.58-36 mM and a chain-length-independent apparent monomeric pKa of 5.6 ± 0.1 for the pyrrolidinium nitrogen. CₙNicBr showed generally superior antimicrobial activity against a broad panel of Gram-positive and Gram-negative bacteria and fungi, including MRSA and Candida albicans, versus equivalent-chain cholinium homologues (CₙCholBr), most pronounced at shorter chains, and 2-4-fold greater eradication of preformed MRSA and C. albicans biofilms (MBEC₈₀ 16-128 μg/mL), matching or exceeding benzalkonium chloride and didecyldimethylammonium bromide. Because the pyrrolidinium pKa places the dicationic form below physiological pH, planktonic potency is governed mainly by hydrophobicity, while pH-driven charge enhancement in the acidic biofilm microenvironment (pH 5.5-6.0) may explain the superior eradication. Haemolytic activity (HC₅₀ 17-576 μM) and cytotoxicity against 3T3 and HaCaT cells defined the biocompatibility window: C₁₄NicBr gave the best selectivity against S. aureus/S. epidermidis (HC₅₀/MIC = 25.5), and C₁₂NicBr the widest safety margin (HC₅₀ = 230 μM), the only homologue reaching a rabbit-erythrocyte-based therapeutic index ≥ 10 against MRSA; nucleated-cell-based selectivity indices were substantially narrowed, indicating that the erythrocyte-based indices overestimate the practical safety margin. CₙNicBr thus emerges as a structurally novel antimicrobial surfactant series that, in most comparisons performed here, showed higher antimicrobial and antibiofilm activity than the cholinium benchmark.

7. Engineered polymeric nanosponge in-situ gel enables sustained and enhanced nose-to-brain delivery of quetiapine fumarate.
7. 工程化聚合物纳米海绵原位凝胶实现富马酸喹硫平的持续和增强鼻-脑递送PMID:日期:2026-09-30Quetiapine fumarate is a second-generation antipsychotic with limited oral bioavailability owing to extensive hepatic first-pass metabolism, making it a suitable candidate for intranasal brain delivery. This study aimed to develop a quetiapine fumarate-loaded nanosponge in-situ gel for sustained and enhanced brain delivery. Nanosponges were prepared by the quasi-emulsion solvent diffusion method and screened using a Plackett-Burman design to identify critical formulation and process variables. Stirring speed, drug amount, and Eudragit L100 amount were further optimized using a Box-Behnken design with particle size, PDI, encapsulation efficiency, and 24 h drug release as responses. The optimized nanosponges showed a particle size of 248.7 nm, PDI of 0.250, and encapsulation efficiency of 97.48%. FTIR, DSC, and XRD analyses confirmed successful drug incorporation and reduced crystallinity of quetiapine fumarate within the nanosponge matrix. The optimized in-situ gel containing quetiapine fumarate-loaded nanosponges (QF-NS) showed physiological pH, appropriate viscosity and gelation, and defined textural characteristics. In vitro release studies showed an apparent sustained-release profile under dialysis conditions, reaching 87.37 ± 3.7% over 24 h, while ex vivo permeation across goat nasal mucosa demonstrated controlled permeation compared with QF solution. Histopathological evaluation showed no overt mucosal damage following short-term ex vivo exposure. In vivo pharmacokinetic studies in rats showed greater brain exposure following intranasal administration than with the intravenous comparator, with a brain Cmax of 9.27 ± 2.34 μg/g compared with 0.57 ± 0.12 μg/g. The observed brain AUC0-6 h was 7.73 μg·h/g after intranasal administration and 2.01 μg·h/g after intravenous administration. The intranasal formulation produced a brain-to-plasma exposure ratio substantially greater than that of the intravenous solution, with DTE and DTP values of 536.51% and 81.36%, respectively. Overall, these findings support the optimized QF-NS-loaded in-situ gel as a promising intranasal platform for sustained drug delivery and enhanced relative brain exposure to quetiapine fumarate.

8. Biogenic selenium nanoparticles as dual-action antithrombotic agents: Targeting platelet activation cascades and thrombus formation.
8. 生物源硒纳米颗粒作为双效抗血栓剂:靶向血小板活化级联和血栓形成PMID:日期:2026-09-29Platelet hyperactivation is a central driver of thrombotic disorders, yet current antithrombotic therapies remain constrained by bleeding risk and other limitations. Here, we investigated protein-capped selenium nanoparticles (SeNPs, ~40 nm) as a potential antithrombotic nanoplatform. SeNPs were synthesized and characterized using spectroscopic and microscopic approaches, demonstrating spherical morphology and protein-associated stabilization. Hemocompatibility and metabolic viability studies showed no hemolysis and no significant reduction in viability at concentrations up to 100 μg/ml under the tested conditions. SeNPs inhibited platelet aggregation and attenuated thrombin-induced cytoskeletal remodeling, integrin αIIbβ3 activation, P-selectin externalization and intracellular Ca⁺ elevation. Mechanistically, SeNP treatment reduced global tyrosine phosphorylation and Akt phosphorylation, together with decreased mitochondrial superoxide detected by MitoSOX, consistent with attenuation of platelet activation-associated signaling and altered mitochondrial redox status. Ex vivo thromboelastography demonstrated delayed clot initiation and reduced clot strength, while fibrin polymerization assays showed prolonged lag time and reduced polymerization rate, indicating platelet-independent modulation of fibrin formation. In vivo, intravenous SeNP administration prolonged tail bleeding time but significantly attenuated FeCl₃-induced carotid artery thrombosis and preserved arterial blood flow. These findings demonstrate that biogenic SeNPs exert complementary effects on platelet activation and fibrin-dependent coagulation processes, while also highlighting the need to define an appropriate therapeutic window between antithrombotic efficacy and bleeding liability.

9. Sequence variations govern the hierarchical self-assembly and structural polymorphism of PSM-α functional amyloids.
9. 序列变异调控PSM-α功能性淀粉样蛋白的分级自组装和结构多态性PMID:日期:2026-09-29Biofilm-associated functional amyloids reinforce extracellular matrices and enhance microbial resistance to environmental and antimicrobial stresses. In Staphylococcus aureus, phenol-soluble modulins (PSMs), particularly the PSM-α family, regulate biofilm maturation and pathogenicity through sequence-dependent self-assembly. However, how subtle sequence variations encode distinct conformational landscapes and supramolecular architectures remains unclear. Here, we combined atomistic discrete molecular dynamics (DMD) and coarse-grained (CG) simulations with all-atom MD refinement to elucidate PSM-α self-assembly. PSM-α1, PSM-α2, and PSM-α4 monomers sampled heterogeneous ensembles with pronounced β-sheet propensities and underwent disorder-to-β-sheet transitions upon self-assembly. Notably, their conserved IIKXI region exhibited the highest β-sheet propensity and inter-peptide backbone contact frequencies, identifying it as a major β-sheet-forming hotspot within the experimentally established amyloidogenic region. In contrast, PSM-α3 intrinsically favored an amphipathic α-helical ensemble and retained its helical organization during assembly through lateral association of preformed helices. CG simulations followed by unrestrained all-atom MD refinement further revealed distinct higher-order architectures and confirmed their structural stability. Collectively, these findings link sequence variation to PSM-α structural polymorphism and highlight the conserved IIKXI region as a potential target for disrupting amyloid assembly and developing anti-biofilm strategies.

10. A smart magneto-responsive ternary 3D platform for actively steering stereoscopic infiltration and chronological osteogenic differentiation of BMSCs.
10. 一种智能磁响应三元3D平台,用于主动调控BMSCs的立体浸润和时序成骨分化PMID:日期:2026-09-29The limited effective infiltration of cells into the interior of scaffolds remains a critical bottleneck limiting the therapeutic application of bone tissue engineering. To address this challenge, this study developed a physically self-assembled ternary smart magneto-responsive composite platform (MGO/CS) based on chitosan, graphene oxide, and superparamagnetic iron oxide nanoparticles (SPIONs). Although the incorporation of SPIONs reduced the mean pore size of the scaffold from 306.64 ± 52.25 μm to 185.39 ± 28.32 μm (p < 0.05), this smart platform, driven by a 3 mT weak static magnetic field (SMF), successfully mitigated the dependence of the three-dimensional colonization of stem cells on pore size, thereby partially overcoming the constraints imposed by conventional geometric topography on deep cell infiltration. By establishing a micro-magnetic environment within the porous scaffold, this magneto-responsive platform may promote BMSC migration across geometric barriers, achieving enhanced deep infiltration throughout the scaffold interior. Furthermore, this intelligent magneto-responsive platform can effectively enhance cellular differentiation signaling, promoting spatiotemporally coordinated osteogenic cascades (characterized by the upregulation of RUNX-2, COL-1, and OPN) and extracellular matrix mineralization. This physical microenvironment modulation strategy mitigates the reliance on geometric topology, offering a novel paradigm for addressing the persistent challenge of insufficient inward cellular infiltration and growth in bone tissue engineering.
